Background Three randomised trials have demonstrated that combining bevacizumab with first-line chemotherapy significantly improves progression-free survival versus chemotherapy alone in HEr2-negative locally recurrent/metastatic breast cancer （Lr/mBC）. However, data from Chinese populations are limited and possible differences between ethnic and geographic populations are unknown. This study was conducted to determine whether there are differences in safety and efficacy in patients with HEr2-negative Lr/mrC between Chinese and Western populations after they receive first-line bevacizumab combined with taxane-based therapy.

Among the four prostaglandin E2 receptors,Ep3 receptor is the one most abundantly expressed in white adipose tissue(WAT).The mouse Ep3 gene gives rise to three isoforms,namely Ep3α,Ep3β,and Ep3γ,which differ only at their C-terminal *** date,functions of Ep3 receptor and its isoforms in WAT remain incompletely *** this study,we found that the expression of all Ep3 isoforms were downregulated in WAT of both db/db and high-fat diet-induced obese *** ablation of three Ep3 receptor isoforms(Ep3^(−/−)mice)or Ep3αand Ep3γisoforms with Ep3βintact(Ep3βmice)led to an obese phenotype with increased food intake,decreased motor activity,reduced insulin sensitivity,and elevated serum *** the differentiation of preadipocytes and mouse embryonic fibroblasts to adipocytes was markedly facilitated by either pharmacological blockade or genetic deletion/inhibition of Ep3 receptor via the cAMp/pKA/pp.r.pathway,increased adipogenesis may contribute to obesity in Ep3^(−/−)and Ep3β***,both Ep3^(−/−)and Ep3βmice had increased lipolysis in WAT mainly due to the activated cAMp/pKA/hormone-sensitive lipase *** together,our findings suggest that Ep3 receptor and itsαandγisoforms are involved in both adipogenesis and lipolysis and influence food intake,serum lipid levels,and insulin sensitivity.

BACKGrOUND:penehyclidine is a newly developed anticholinergic *** aimed to investigate the role of penehyclidine in acute organophosphorus pesticide poisoning(Op)***:We searched the pubmed,Cochrane library,EMBASE,Chinese National Knowledge Infrastructure(CNKI),Chinese Biomedical literature(CBM)and Wanfang *** controlled trials(rCTs)recruiting acute Op patients were identifi ed for *** outcomes included cure rate,mortality rate,time to atropinization,time to 60%normal acetylcholinesterase(AchE)level,rate of intermediate syndrome(IMS)and rate of adverse drug reactions(ADr).rESULTS:Sixteen rCTs involving 1,334 patients were identifi *** with the atropineor penehyclidine-alone groups,atropine combined with penehyclidine significantly increased the cure rate(penehyclidine+atropine ***,0.97 vs.0.86,rr 1.13,95%CI[1.07–1.19];penehyclidine+atropine ***,0.93 vs.0.80,rr 1.08,95%CI[1.01–1.15])and reduced the mortality rate(penehyclidine+atropine ***,0.015 vs.0.11,rr 0.17,95%CI[0.06–0.49];penehyclidine+atropine ***,0.13 vs.0.08,rr 0.23,95%CI[0.04–1.28]).Atropine combined with penehyclidine in Op patients also helped reduce the time to atropinization and AchE recovery,the rate of IMS and the rate of *** with a single dose of atropine,a single dose of penehyclidine also signifi cantly elevated the cure rate,reduced times to atropinization,AchE recovery,and rate of ***:Atropine combined with penehyclidine benefi ts Op patients by enhancing the cure rate,mortality rate,time to atropinization,AchE recovery,IMS rate,total ADr and duration of *** combined with atropine is likely a better initial therapy for Op patients than atropine alone.

The ongoing coronavirus disease 2019(COVID-19)pandemic caused more than 96 million infections and over 2 million deaths worldwide so ***,there is no ap.r.ved vaccine available for severe acute resp.r.tory syndrome coronavirus 2(SArS-CoV-2),the disease causative *** is the most effective ap.r.ach to eradicate a *** tests of safety and efficacy in animals are pivotal for developing a vaccine and before the vaccine is applied to human *** we evaluated the safety,immunogenicity,and efficacy of an inactivated vaccine based on the whole viral p.r.icles in human ACE2 transgenic mouse and in non-human *** data showed that the inactivated vaccine successfully induced SArS-CoV-2-specific neutralizing antibodies in mice and non-human primates,and subsequently provided p.r.ial(in low dose)or full(in high dose)protection of challenge in the tested *** addition,passive serum transferred from vaccine-immunized mice could also provide full protection from SArS-CoV-2 infection in *** results warranted positive outcomes in future clinical trials in humans.

腔镜切割荷包吻合器在腹腔镜结直肠癌手术中的应用体会

目的结直肠手术中进行腔镜下荷包缝合一直是手术操作的难点,尤其是经自然腔道取标本手术(NOSES)。同时结肠、直肠吻合口处形成的“危险三角区”又是增加吻合口漏的主要因素。腔镜切割荷包吻合器的临床应用可以有效解决上述技术难题。方法本研究纳入5例行根治性手术的结直肠癌患者,包括4例直肠癌和1例乙状结肠癌。所有患者手术消化道重建过程中,乙状结肠荷包缝合及抵钉座固定步骤均使用腔镜切割荷包吻合器。结果患者平均年龄为(54.4±8.6)岁,患者平均BMI为(24.7±2.9)kg/m^(2)。4例患者行腹腔镜直肠癌根治+预防性末端回肠双腔造口术,1例患者行NOSES Ⅳ式。平均手术时间为(123.60±15.37)min,平均术中出血量为(98±36)mL,平均术后住院天数为(9.4±0.5)天。所有患者围术期均未出现并发症。结论腔镜直线荷包缝合器降低了全腔镜下乙状结肠与直肠消化道重建的操作难度。该设备的临床应用无论是对于常规腹腔镜手术,亦或是NOSES手术,都具有很好的临床价值与推广潜力。

An association study of HFEgene mutation with idiopathic male infertility in the Chinese Han population

Mutations in the haemochromatosis gene （HFE） influence iron status in the general population of Northern Europe, and excess iron is associated with the impairment of sp.r.atogenesis. The aim of this study is to investigate the association between three mutations （C282Y, H63D and S65C） in the HFEgene with idiopathic male infertility in the Chinese Han population. Two groups of Chinese men were recruited： 444 infertile men （including 169 with idiopathic azoosp.r.ia） and 423 controls with proven fertility. The HFEgene was detected using polymerase chain reaction-restriction fragment length polymorphism （p.r.rFLp） technique. The exp.r.mental results demonstrated that no C282Y or $65C mutations were detected. Idiopathic male infertility was not significantly associated with heterozygous H63D mutation （odds ratio=O.801, 95% confidence interval=0.452-1.421, X2=0.577, p=0.448）. The H63D mutation frequency did not correlate significantly with the serum luteinizing hormone （LH）, follicle-stimulating hormone （FSH） and testosterone （T） levels in infertile men （p=0.896, p=0.404 and p=O.05, respectively）. Our data suggest that the HFEH63D mutation is not associated with idiopathic male reproductive dysfunction.

As the third pap.r.in the multiple-p.r. series,we rep.r. the statistical prop.r.ies of radio bursts detected from the repeating fast radio burst(FrB)source FrB 20201124A with the Five-hundred-meter Ap.r.ure Spherical radio Telescope during an extremely active episode between the 25th and 28th of September 2021(UT).We focus on the polarization prop.r.ies of536 bright bursts with S/N>*** found that the Faraday rotation measures(rMs)monotonically dropped from-579to-605 rad m^(-2)in the 4 day *** rM values were compatible with the values(-300 to-900 rad m^(-2))rep.r.ed 4 months ***,the rM evolution rate in the current observation window was at least an order of magnitude smaller than the one(~500 rad m^(-2)day^(-1))previously rep.r.ed during the rapid rM-variation phase,but is still higher than the one(≤1 rad m^(-2)day^(-1))during the later rM no-evolution *** bursts of FrB 20201124A were highly polarized with the total degree of polarization(circular plus linear)greater than 90%for more than 90%of all *** distribution of linear polarization position angles(pAs),degree of linear polarization(L/I)and degree of circular polarization(V/I)can be characterized with unimodal distribution *** the observation window,the distributions became wider with time,i.e.,with larger scatter,but the centroids of the distribution functions remained nearly *** individual bursts,significant pA variations(confidence level 5σ)were observed in 33%of all *** polarization of single pulses seems to follow certain complex trajectories on the poincarésphere,which may shed light on the radiation mechanism at the source or the plasma prop.r.ies along the path of FrB propagation.

The Circular Electron positron Collider(CEpC)is a large scientific project initiated and hosted by china,fostered through extensive collaboration with international *** complex comprises four accelerators:a 30 GeV Linac,a 1.1 GeV Damping ring,a Booster capable of achieving energies up to 180 GeV,and a Collider op.r.ting at varying energy modes(Z,W,H,and tt).The Linac and Damping ring are situated on the surface,while the subterranean Booster and Collider are housed in a 100 km circumference underground tunnel,strategically accommodating future expansion with provisions for a potential Sup.r.proton proton Collider(SppC).The CEpC primarily serves as a Higgs *** its baseline design with synchrotron radiation(Sr)power of 30 MW p.r.beam,it can achieve a luminosity of 5×10^(34)cm^(-2)s^(-1)p.r.interaction point(Ip),resulting in an integrated luminosity of 13 ab^(-1)for two Ips over a decade,producing 2.6 million Higgs *** the Sr power to 50 MW p.r.beam expands the CEpC's capability to generate 4.3 million Higgs bosons,facilitating precise measurements of Higgs coupling at sub-p.r.ent levels,exceeding the precision expected from the HL-LHC by an order of *** Technical Design rep.r.(TDr)follows the preliminary Conceptual Design rep.r.(pre-CDr,2015)and the Conceptual Design rep.r.(CDr,2018),comprehensively detailing the machine's layout,p.r.ormance metrics,physical design and analysis,technical systems design,r&D and prototyping efforts,and associated civil engineering ***,it includes a cost estimate and a preliminary construction timeline,establishing a framework for forthcoming engineering design phase and site selection *** is anticipated to begin around 2027-2028,pending government ap.r.val,with an estimated duration of 8 *** commencement of exp.r.ments and data collection could potentially be initiated in the mid-2030s.

Autonomous one-shot on-the-fly learning copes with the high privacy,small dataset,and in-stream data at the *** such learning on digital hardware suffers from the well-known von-Neumann and scaling *** optical neural networks featuring large p.r.llelism,low latency,and high efficiency offer a promising ***,ex-situ training of conventional optical networks,where optical path configuration and deep learning model optimization are sep.r.ted,incurs hardware,energy and time overheads,and defeats the advantages in edge ***,we introduced a bio-insp.r.d material-algorithm co-design to construct a hydrogel-based optical Willshaw model(HOWM),manifesting Hebbian-rule-based structural plasticity for simultaneous optical path configuration and deep learning model optimization thanks to the underlying opto-chemical *** first employed the HOWM as an all optical in-sensor AI processor for one-shot pattern classification,association and *** then leveraged HOWM to function as a ternary content addressable memory(TCAM)of an optical memory augmented neural network(MANN)for one-shot learning the Omniglot *** HOWM empowered one-shot on-the-fly edge learning leads to 1000boost of energy efficiency and 10boost of speed,which paves the way for the next-generation autonomous,efficient,and affordable smart edge systems.
