BACKGROUND Gastric cancer(GC)is the fifth most common type of cancer and has the fourth highest death rate among all *** is a lack of studies examining the impact of liver metastases on the effectiveness of immunotherapy in individuals diagnosed with *** To investigate the influence of liver metastases on the effectiveness and safety of immunotherapy in patients with advanced *** This retrospective investigation collected clinical data of patients with advanced stomach cancer who had immunotherapy at our hospital from February 2021 to January *** baseline attributes were compared using either the Chi-square test or the Fisher exact probability *** chi-square test and Kaplan-Meier survival analysis were employed to assess the therapeutic efficacy and survival duration in GC patients with and without liver *** The analysis comprised 48 patients diagnosed with advanced GC,who were categorized into two groups:A liver metastasis cohort(n=20)and a non-liver metastatic cohort(n=28).Patients with liver metastasis exhibited a more deteriorated physical condition compared to those without liver *** objective response rates in the cohort with metastasis and the cohort without metastasis were 15.0%and 35.7%(P>0.05),***,the disease control rates in these two cohorts were 65.0%and 82.1%(P>0.05),*** median progression-free survival was 5.0 months in one group and 11.2 months in the other group,with a hazard ratio of 0.40 and a significance level(P)less than *** median overall survival was 12.0 months in one group and 19.0 months in the other group,with a significance level(P)greater than *** Immunotherapy is less effective in GC patients with liver metastases compared to those without liver metastasis.

On October 5th, 2020, Drs. Harvey J. Alter, Michael Houghton and Charles M. Rice were rewarded with Nobel Prize in Physiology or Medicine for "the discovery of hepatitis C virus(HCV)". During the past 50 years, remarkable achievements have been made in treatment of HCV infection: it has changed from being a life-threatening chronic disease to being curable. In this commentary, we briefly summarized the milestone events in the "scientific journey" from the first report of non-A, non-B hepatitis and discovery of the pathogen(HCV) to final identification of efficacious direct-acting antivirals. Further, we address the challenges and unmet issues in this field.

Structural features in the mid-southern section of the Kyushu–Palau Ridge based on satellite altimetry gravity anomaly

The Kyushu–Palau Ridge(KPR),an anti-S-shaped submarine highland at the center of the Philippine Sea Plate(PSP),is considered the residual arc of the Izu–Bonin–Mariana Island Arc,which retains key information about the cessation of the Western Philippine Basin(WPB)expansion and the Parece Vela Basin(PVB)***,using the new generation of satellite altimetry gravity data,high-precision seafloor topography data,and newly acquired ship-borne gravity data,the topographic and gravity characteristics of the KPR mid-southern section and adjacent region are *** distribution characteristics of the faults were delineated using the normalized vertical derivative–total horizontal derivative method(NVDR-THDR)and the minimum curvature potential field separation *** Moho depth and crustal thickness were inverted using the rapid inversion method for a double-interface model with depth *** on these results,the crust structure features in the KPR mid-southern section,and the“triangular”structure geological significance where the KPR and Central Basin Rift(CBR)of the WPB intersect are *** KPR crustal thickness is approximately 6–16 km,with a distinct discontinuity that is slightly thicker than the normal oceanic *** KPR mid-southern section crust structure was divided into four segments(S1–S4)from north to south,formed by the CBR eastward extension joint action and clockwise rotation of the PVB expansion axis and the Mindanao fault zone blocking effect.

Cofitness network connectivity determines a fuzzy essential zone in open bacterial pangenome

Most in silico evolutionary studies commonly assumed that core genes are essential for cellular function,while accessory genes are dispensable,particularly in nutrient-rich ***,this assumption is seldom tested genetically within the pangenome *** this study,we conducted a robust pangenomic Tn-seq analysis of fitness genes in a nutrient-rich medium for Sinorhizobium strains with a canonical open *** evaluate the robustness of fitness category assignment,Tn-seq data for three independent mutant libraries per strain were analyzed by three methods,which indicates that the Hidden Markov Model(HMM)-based method is most robust to variations between mutant libraries and not sensitive to data size,outperforming the Bayesian and Monte Carlo simulation-based ***,the HMM method was used to classify the fitness *** genes,categorized as essential(ES),advantage(GA),and disadvantage(GD)genes for growth,are enriched in core genes,while nonessential genes(NE)are over-represented in accessory *** ES/GA genes showed a lower fitness effect than core ES/GA *** degrees in the cofitness network decrease in the order of ES,GD,and GA/*** addition to accessory genes,1599 out of 3284 core genes display differential essentiality across test *** the pangenome core,both shared quasi-essential(ES and GA)and strain-dependent fitness genes are enriched in similar functional *** analysis demonstrates a considerable fuzzy essential zone determined by cofitness connectivity degrees in Sinorhizobium pangenome and highlights the power of the cofitness network in understanding the genetic basis of ever-increasing prokaryotic pangenome data.

Pyrolysis Empirical Modeling of Polyester Glass Fiber Reinforced Plastics Using Sestak-Berggren Model Method

Polyester Glass Fiber Reinforced Plastic(Polyester GFRP),a thermosetting plastic comprised of glass fiber and polyester polymer compounds,is extensively utilized in high-speed *** its pyrolysis mechanism is crucial as it significantly influences the combustion characteristics and fire safety ***,kinetic research on polyester GFRP primarily focuses on employing the Coats-Redfern method to derive a theoretical kinetic ***,the pyrolysis process of polyester GFRP is complex and the aforementioned theoretical model fails to accurately describe the pyrolysis ***,this study seeks to utilize the Sestak and Berggren(SB) model as a methodological approach to reveal the complex reaction mechanism during the pyrolysis *** on thermogravimetric analysis,the entire pyrolysis process of polyester GFRP is divided into two primary ***,model-free methods are employed to ascertain the activation energy and pre-exponential *** results show that the fitted empirical models of the two main pyrolysis stages are f(α)=(1-α)^(1.47)[-ln(1-α)]^(1.50) and f(α)=(1-α)^(1.77)[-ln(1-α)]^(1.72),*** predicted results are in good agreement with experimental data under different heating rates,which indicates that the empirical model can sufficiently describe the pyrolysis process of polyester GFRP.

HLA-mismatched allogeneic adoptive immune therapy in severely immunosuppressed AIDS patients

Severely immunosuppressed AIDS patients with recurrent opportunistic infections(Ols)represent an unmet medical need even in the era of antiretroviral therapy(ART).Here we report the development of a human leukocyte antigen(HLA)-mismatched allogeneic adaptive immune therapy(AAIT)for severely immunosuppressed AIDS *** severely immunosuppressed AIDS patients with severe Ols were enrolled in this single-arm *** donors received subcutaneous recombinant granulocyte-colony-stimulating factor twice daily for 4-5 days to stimulate *** blood mononuclear cells were collected from these donors via leukapheresis and transfused into the coupled ***,immunological,and virological parameters were monitored during a 12-month follow-up *** found AAIT combined with ART was safe and well-tolerated at the examined doses and transfusion regimen in all 12 *** in clinical symptoms were evident throughout the study *** patients exhibited a steady increase of peripheral CD4^(+)T cells from a median 10.5 to 207.5 cells/μ*** increase in peripheral CD8^(+)T-cell count from a median 416.5 to 1206.5 cells/μl was found in the first 90 days since initiation of *** addition,their inflammatory cytokine levels and HIV RNA viral load decreased.A short-term microchimerism with donor cells was *** were no adverse events associated with graft-versus-host disease throughout the study ***,AAIT treatment was safe,and might help severely immunosuppressed AIDS patients to achieve a better immune restoration.A further clinical trial with control is necessary to confirm the efficacy of AAIT medication.
