Steam disinfection enhances bioaccessibility of metallic nanoparticles in nano-enabled silicone-rubber baby bottle teats,pacifiers,and teethers

Nano-enabled silicone-rubber articles for feeding or chewing could be a source of metallic nanoparticles(NPs)directly exposed to infants and young children.However,the impact of steam disinfection on release of NPs and the related potential risks to children's health are unknown.Here,we investigated contents and form of Ag and Zn in 57 nano-enabled silicone-rubber baby bottle teats,pacifiers,and teethers of seven countries and examined the impacts of steam disinfection on in vitro bioaccessibility(IVBA)of Ag and Zn in the articles.Nearly 89%articles had a mixture of Ag-and Zn-containing NPs and the teethers had relatively high Ag and Zn contents(up to 501 and 254μg/g,respectively).Steam disinfection caused rubber decomposition into micro(nano)plastics(0.54-15.7μm)and NP release from the interior of bulk rubber and micro-sized plastics,thus enhancing the IVBA of Ag and Zn by up to 5.5 times.The findings provide insights into mechanisms for NP release by steam disinfection.Though oral exposure risk assessment suggested low health concerns on individual metal release,our study points out the need to assess the potential health risks of child co-exposure to metallic NPs and micro(nano)plastics.

Bone defects and non-union are prevalent in clinical orthopedy,and the outcomes of current treatments are often suboptimal.Bone tissue engineering offers a promising approach to treating these conditions effectively.Bone morphogenetic protein 9(BMP9)can commit mesenchymal stem cells to osteogenic lineage,and a knowledge of the underlying mechanisms may help advance the field of bone tissue engineering.Leucine-rich repeats con-taining G protein-coupled receptor 4(LGR4),a member of G protein-coupled receptors,is essential for modulating bone development.This study is aimed at investigating the impact of LGR4 on BMP9-induced osteogenesis in mesenchymal stem cells as well as the underlying mechanisms.Bone marrow stromal cells from BMp9-knockout mice exhibited diminished LGR4 expression,and exogenous LGR4 clearly restored the impaired osteogenic potency of the bone marrow stromal cells.Furthermore,LGR4 expression was increased by BMP9 in C3H10T1/2 cells.LGR4 augmented the benefits of BMP9-induced osteogenic markers and bone formation,whereas LGR4 inhibition restricted these effects.Meanwhile,the BMP9-induced li-pogenic markers were increased by LGR4 inhibition.The protein levels of Raptor and p-Stat3 were elevated by BMP9.Raptor knockdown or p-Stat3 suppression attenuated the osteoblastic markers and LGR4 expression brought on by BMP9.LGR4 significantly reversed the blocking ef-fect of Raptor knockdown or p-Stat3 suppression on the BMP9-induced osteoblastic markers.Raptor interacts with p-Stat3,and p-Stat3 activates the LGR4 promoter activity.In conclusion,LGR4 boosts BMP9 osteoblastic potency in mesenchymal stem cells,and BMP9 may up-regulate LGR4 via the mTORC1/Stat3 signal activation.

Zinc glycine chelate ameliorates DSS‑induced intestinal barrier dysfunction via attenuating TLR4/NF‑κB pathway in meat ducks

Background Zinc glycine chelate(Zn-Gly)has anti-inflammation and growth-promoting properties;however,the mechanism of Zn-Gly contribution to gut barrier function in Cherry Valley ducks during intestinal inflammation is unknown.Three-hundred 1-day-old ducks were divided into 5 groups(6 replicates and 10 ducks per replicate)in a completely randomized design:the control and dextran sulfate sodium(DSS)groups were fed a corn-soybean meal basal diet,and experimental groups received supplements of 70,120 or 170 mg/kg Zn in form of Zn-Gly.The DSS and treatment groups were given 2 mL of 0.45 g/mL DSS daily during d 15–21,and the control group received normal saline.The experiment lasted 21 d.Results Compared with DSS group,70,120 and 170 mg/kg Zn significantly increased body weight(BW),villus height and the ratio of villus to crypt,and significantly decreased the crypt depth of jejunum at 21 d.The number of goblet cells in jejunal villi in the Zn-Gly group was significantly increased by periodic acid-Schiff staining.Compared with control,the content of intestinal permeability marker D-lactic acid(D-LA)and fluxes of fluorescein isothiocyanate(FITC-D)in plasma of DSS group significantly increased,and 170 mg/kg Zn supplementation significantly decreased the D-LA content and FITC-D fluxes.Compared with control,contents of plasma,jejunum endotoxin and jejunum pro-inflammatory factors IL-1β,IL-6 and TNF-αwere significantly increased in DSS group,and were significantly decreased by 170 mg/kg Zn supplementation.Dietary Zn significantly increased the contents of anti-inflammatory factors IL-10,IL-22 and sIgA and IgG in jejunum.Real-time PCR and Western blot results showed that 170 mg/kg Zn supplementation significantly increased mRNA expression levels of CLDN-1 and expression of OCLN protein in jejunum,and decreased gene and protein expression of CLDN-2 compared with DSS group.The 120 mg/kg Zn significantly promoted the expressions of IL-22 and IgA.Dietary Zn-Gly supplementation significantly decreased pro-inflammatory genes IL-8

Influence of Incomplete Soil Plugs on Bearing Capacities of Bucket Foundations in Clay

Due to the uneven seabed and heaving of soil during pumping,incomplete soil plugs may occur during the installation of bucket foundations,and the impacts on the bearing capacities of bucket foundations need to be evaluated.In this paper,the contact ratio(the ratio of the top diameter of the soil plug to the diameter of the bucket)and the soil plug ratio(the ratio of the soil heave height to the skirt height)are defined to describe the shape and size of the incomplete soil plug.Then,finite element models are established to investigate the bearing capacities of bucket foundations with incomplete soil plugs and the influences of the contact ratios,and the soil plug ratios on the bearing capacities are analyzed.The results show that the vertical bearing capacity of bucket foundations in homogeneous soil continuously improves with the increase of the contact ratio.However,in normally consolidated soil,the vertical bearing capacity barely changes when the contact ratio is smaller than 0.75,while the bearing capacity suddenly increases when the contact ratio increases to 1 due to the change of failure mode.The contact ratio hardly affects the horizontal bearing capacity of bucket foundations.Moreover,the moment bearing capacity improves with the increase of the contact ratio for small aspect ratios,but hardly varies with increasing contact ratio for aspect ratios larger than 0.5.Consequently,the reduction coefficient method is proposed based on this analysis to calculate the bearing capacities of bucket foundations considering the influence of incomplete soil plugs.The comparison results show that the proposed reduction coefficient method can be used to evaluate the influences of incomplete soil plug on the bearing capacities of bucket foundations.

Defects in the grain interiors of 3 mol%yttria-stabilized tetragonal zirconia polycrystal ceramics with 0.25 wt%alumina

The presence of high-density defects is rarely observed in bulk 3 mol%yttria-stabilized tetragonal zirconia polycrystal(3Y-TZP)ceramics obtained through conventional pressureless sintering.In the present work,fine-grained dense 147 nm 3Y-TZP ceramics were prepared by pressureless sintering of commercial 0.25 wt%alumina-doped zirconia powders at 1300℃.A novel discovery was reported in which large amounts of defects were present in the grain interiors of the sample.The phenomenon was further examined using three types of powder samples,and the reasons for defect formation were investigated by microstructural characterization using high-resolution transmission electron microscopy(HRTEM)analysis and Rietveld refinement.The results confirmed the essential dependence of the defect formation on the alumina addition.The authors attributed the defect formation to the significant difference in ionic radii of the solvent and solute during the dissolution of alumina into the zirconia lattice.The sintering kinetics were proposed to be enhanced by the presence of substantial defects,which consequently favored the low-temperature sintering of the alumina-doped zirconia ceramics.

This study presents a comprehensive overview of the atmospheric pollutants including Sulfur dioxide(SO_(2)),Nitrogen dioxide(NO_(2)),Formaldehyde(HCHO),Particulate Matter PM;PM_(10):diameter≤10μm,and PM_(2.5):diameter≤2.5μm,and Ozone(O_(3)),over Dongying(Shandong Province)from March-April 2018 and September-October 2019 by employing ground-based Multiple Axis Differential Optical Absorption Spectroscopy(MAX-DOAS)observations along with the in-situ measurements attained by the national air quality monitoring platform.The concentrations of SO_(2)and NO_(2)were under the acceptable level,while both PM_(2.5),and PM_(10)were higher than the safe levels as prescribed by national and international air quality standards.The results depict that 21%of the total observation days were found to be complex polluted days(PM_(2.5)>35μg/m^(3) and O_(3)>160μg/m^(3)).The secondary HCHO was used for accurate analysis of O_(3)sensitivity.A difference of 11.40%and 10%during March-April 2018 and September-October 2019 respectively in O_(3)sensitivity was found between HCHO_(total)/NO_(2)and HCHO_(sec)/NO_(2).The results indicate that primary HCHO have significant contribution in HCHO.O_(3)formation predominantly remained to be in VOC-limited and transitional regime during March-April 2018 and September-October 2019 in Dongying.These results imply that concurrent control of both NO_(x) and VOCs would benefit in ozone reductions.Additionally,the criteria pollutants(PM,SO_(2),and NO_(2))depicted strong correlations with each other except for O_(3)for which weak correlation coefficient was obtained with all the species.This study will prove to be baseline for designing of air pollution control strategies.

Human activity over natural inputs determines the bacterial community in an ice core from the Muztag ata glacier

Ice core provides a valuable vertical timeline of past climates and anthropogenic activities.Environmental proxies have been widely used in these studies,but there are few biological indicators available.To address this gap,we investigated the bacterial community from a 74 m ice core of Muztag ata glacier on the Tibetan Plateau to link biological indicators with past climate and anthropogenic activities.By analyzing the portion of the ice core with environmental proxies available(corresponding to 1907 to 1991),we observed an increase in bacterial richness throughout the ice core,which was associated with higher NH_(4)^(+),an indicator of agricultural development.The bacterial community was jointly determined by human activity,natural input,and air temperature,with a strong human influence after the 1950s.Furthermore,the relative abundance of animal gut-associated bacteria,including Aerococcaceae,Nocardiaceae,Muribaculaceae,and Lachnospiraceae,was associated with livestock number changes in the Central Asian region.Together with other bacterial lineages,they jointly explained 59.8%of the livestock number changes.This study provides quantitative evidence of the associations between bacterial indicators and past climate and human activities,highlighting the potential of using bacterial proxies for ice core studies.

C1QTNF5 is a novel attachment factor that facilitates the entry of influenza A virus

Influenza A virus(IAV)binds sialic acid receptors on the cell surface to enter the host cells,which is the key step in initiating infection,transmission and pathogenesis.Understanding the factors that contribute to the highly efficient entry of IAV into human cells will help elucidate the mechanism of viral entry and pathogenicity,and provide new targets for intervention.In the present study,we reported a novel membrane protein,C1QTNF5,which binds to the hemagglutinin protein of IAV and promotes IAV infection in vitro and in vivo.We found that the HA1 region of IAV hemagglutinin is critical for the interaction with C1QTNF5 protein,and C1QTNF5 interacts with hemagglutinin mainly through its N-terminus(1–103 aa).In addition,we further demonstrated that overexpression of C1QTNF5 promotes IAV entry,while blocking the interaction between C1QTNF5 and IAV hemagglutinin greatly inhibits viral entry.However,C1QTNF5 does not function as a receptor to mediate IAV infection in sialic acid-deficient CHO-Lec2 cells,but promotes IAV to attach to these cells,suggesting that C1QTNF5 is an important attachment factor for IAV.This work reveals C1QTNF5 as a novel IAV attachment factor and provides a new perspective for antiviral strategies.

Numerical Simulation on Radon Retardation Behavior of Covering Floats in Radon-Containing Water

Objective This study aimed to efficiently reduce the release of radon from water bodies to protect the environment.Methods Based on the sizes of the experimental setup and modular float,computational fluid dynamics(CFD)was used to assess the impact of the area coverage rate,immersion depth,diffusion coefficient,and radon transfer velocity at the gas–liquid interface on radon migration and exhalation of radon-containing water.Based on the numerical simulation results,an estimation model for the radon retardation rate was constructed.The effectiveness of the CFD simulation was evaluated by comparing the experimental and simulated variation values of the radon retardation rate with the coverage area rates.Results The effect of radon transfer velocity on radon retardation in water bodies was minor and insignificant according to the appropriate value;therefore,an estimation model of the radon retardation rate of the coverage of a radon-containing water body was constructed using the synergistic impacts of three factors:area coverage rate,immersion depth,and diffusion coefficient.The deviation between the experimental and simulated results was<4.3%.Conclusion Based on the numerical simulation conditions,an estimation model of the radon retardation rate of covering floats in water bodies under the synergistic effect of multiple factors was obtained,which provides a reference for designing covering floats for radon retardation in radoncontaining water.

Interface strengthening for thermal sprayed WC-10Co4Cr coating subjected to pulsed magnetic treatment

The reliability of the coated industry components demands ideal fatigue properties of the coating,and it is mainly determined by the performance of the interfaces.In this study,pulsed magnetic treatment(PMT)was applied to the thermal sprayed WC-10Co4Cr coating,and the fatigue lifetime of the coated bolt increased by 219.82%under an imitation of the operating mode condition.Scratch tests further proved that both the adhesion and cohesion strength were improved after PMT,and they benefit from the interface strengthening effects.The formation of coherent WC/Co interfaces was characterized by in-situ transmission electron microscopy(TEM),and the molecular dynamic simulations indicate that the work of separation of these interfaces is much higher than the original disordered ones.Residual stress was relaxed and distributed more homogeneously after PMT,and it mainly contributes to the coating/substrate strengthening.This work provides a new post-treatment method focusing on the interfaces in the WC-based coating and gives insight into its mechanism so that it is hopeful to be applied to other kinds of coatings.
