Monotropein Induced Apoptosis and Suppressed Cell Cycle Progression in Colorectal Cancer Cells

Objective: To determine whether monotropein has an anticancer effect and explore its potential mechanisms against colorectal cancer(CRC) through network pharmacology and molecular docking combined with experimental verification. Methods: Network pharmacology and molecular docking were used to predict potential targets of monotropein against CRC. Cell counting kit assay, plate monoclonal assay and microscopic observation were used to investigate the antiproliferative effects of monotropein on CRC cells HCT116, HT29 and LoVo. Flow cytometry and scratch assay were used to analyze apoptosis and cell cycle, as well as cell migration, respectively in HCT116, HT29, and LoVo cells. Western blotting was used to detect the expression of proteins related to apoptosis, cell cycle, and cell migration, and the expression of proteins key to the Akt pathway. Results: The Gene Ontology and Reactome enrichment analyses indicated that the anticancer potential of monotropein against CRC might be involved in multiple cancer-related signaling pathways. Among these pathways, RAC-beta serine/threonine-protein kinase(Akt1, Akt2), cyclin-dependent kinase 6(CDK6), matrix metalloproteinase-9(MMP9), epidermal growth factor receptor(EGFR), cell division control protein 42 homolog(CDC42) were shown as the potential anticancer targets of monotropein against CRC. Molecular docking suggested that monotropein may interact with the 6 targets(Akt1, Akt2, CDK6, MMP9, EGFR, CDC42). Subsequently, cell activity of HCT116, HT29 and LoVo cell lines were significantly suppressed by monotropein(PlinD1, CDK4 and CDK6, inhibited cell migration by suppressing the expressions of CDC42 and MMP9(Pling pathway. In situ modification of heavy oil catalyzed by nanosized metal-organic framework at mild temperature and its mechanism

Two catalysts, nano-sized cobalt-metal-organic framework(Co-MOF) and nickel(Ni)-MOF, were successfully prepared by the modification method. Tetralin(C10H12) was used as the hydrogen donor for the catalytic cracking and hydrogenation modification study of the dehydrated crude oil from the Shengli Oilfield. The optimal reaction conditions were determined through orthogonal experiments, and the components of the crude oil and modified oil samples were analyzed. The results revealed that the nanoMOF catalysts were successfully prepared and exhibited high catalytic activity. They could catalyze the cracking of large molecules in heavy oil at mild temperatures(<300°C), leading to the decomposition of the hydrogen donor. When the mass fraction of the catalyst was 0.2%, the mass fraction of the hydrogen donor was 1%, and the reaction temperature was 280°C, the Ni-MOF showed the best catalytic viscosity reduction effect. It could reduce the viscosity of heavy oil at 50°C from 15761.9 m Pa.s to 1266.2 m Pa.s,with a viscosity reduction rate of 91.97%. The modification effect of Co-MOF was the next best, which could reduce the viscosity of heavy oil to 2500.1 m Pa.s with a viscosity reduction rate of 84.14%. Molecular dynamics simulations revealed a strong interaction force between the MOF surface and asphaltene molecules. In the process of heavy-oil catalytic hydrogenation, the nano-MOF catalyst exhibited high catalytic activity. On the one hand, the empty d orbitals outside the metal atoms in the catalyst could polarize the carbon atoms in the organic matter, accelerating the breaking of long chains. On the other hand, the metal atoms in the catalyst could bond with the carbon σ bonds, breaking the carbon-carbon bonds. This disrupted the structure of the recombined components in the crude oil, irreversibly reducing the viscosity of the heavy oil and improving its fluidity.

SciCN:A Scientific Dataset for Chinese Named Entity Recognition

Named entity recognition(NER)is a fundamental task of information extraction(IE),and it has attracted considerable research attention in recent years.The abundant annotated English NER datasets have significantly promoted the NER research in the English field.By contrast,much fewer efforts are made to the Chinese NER research,especially in the scientific domain,due to the scarcity of Chinese NER datasets.To alleviate this problem,we present aChinese scientificNER dataset–SciCN,which contains entity annotations of titles and abstracts derived from 3,500 scientific papers.We manually annotate a total of 62,059 entities,and these entities are classified into six types.Compared to English scientific NER datasets,SciCN has a larger scale and is more diverse,for it not only contains more paper abstracts but these abstracts are derived from more research fields.To investigate the properties of SciCN and provide baselines for future research,we adapt a number of previous state-of-theart Chinese NER models to evaluate SciCN.Experimental results show that SciCN is more challenging than other Chinese NER datasets.In addition,previous studies have proven the effectiveness of using lexicons to enhance Chinese NER models.Motivated by this fact,we provide a scientific domain-specific lexicon.Validation results demonstrate that our lexicon delivers better performance gains than lexicons of other domains.We hope that the SciCN dataset and the lexicon will enable us to benchmark the NER task regarding the Chinese scientific domain and make progress for future research.The dataset and lexicon are available at:https://***/yangjingla/SciCN.git.

Micro-nanoarchitectonic of aluminum-hydrogel propellant with static stability and dynamic rheology

The aluminum-water system is a promising propellant due to high energy and low signal characteristics,and the gel form is easier to store and utilize.In this work,hydrogels of water and aluminum particles were prepared using the low-molecular-weight gellant agarose.The various physical properties of gel systems,including the water loss rate,phase transition temperature,and centrifugal stability at different gellant and aluminum contents,were examined.Rheological properties were assessed through shear thinning tests,thixotropy tests,strain sweep analysis,and frequency sweep experiments.The microstructure of the gel was obtained through scanning electron microscopy images.The results show that the aluminum-hydrogel network structure is composed of micron-scale aluminum and agarose nanosheets,and the unique micro-nanostructure endows the gel with excellent mechanical strength and thermal stability,which improve with increasing gellant and aluminum contents.Notably,the gel with 2% agarose and 20% aluminum had the best performance;the storage modulus reached 90647 Pa,which was within the linear viscoelastic region,and the maximum withstand pressure was 111.2 kPa,which was 118.8% greater than that of the pure hydrogel.Additionally,the gel demonstrates remarkable shear thinning behavior and can undergo gel-sol transformation upon shearing or heating to exceeding 114℃.

An improved installation of 2-hydroxy-4-methoxybenzyl(iHmb)method for chemical protein synthesis

The 2-hydroxy-4-methoxybenzyl(Hmb)backbone modification can prevent amide bond-mediated sidereactions(e.g.,aspartimide formation,peptide aggregation)by installing the removable Hmb group into a peptide bond,thus improving the synthesis of long and challenging peptides and proteins.However,its use is largely precluded by the limited Hmb’s installation sites.In this report,an improved installation of Hmb(iHmb)method was developed to achieve the flexible installation and the convenient removal of Hmb.The iHmb method involves two critical steps:(1)oxidative diazotization of the readily installed 2-hydroxy-4-methoxy-5-amino-benzyl(Hmab)to give 2-hydroxy-4-methoxy-5-diazonium-benzyl(Hmdab)by combining soamyl nitrite(IAN)/HBF_(4),and(2)reductive elimination of Hmdab to give the desired Hmb by 1,2-ethanedithiol(EDT).The iHmb method enables the installation of Hmb at any primary amino acid including the highly sterically hindered amino acids(e.g.,valine and isoleucine).The practicality and utility of the iHmb method was demonstrated by one-shot solid-phase synthesis of a challenging aspartimide-prone peptide,the mirror-image version of a hydrophobic peptide and a long-chain peptide up to 76-residue.Furthermore,the iHmb method can be utilized to facilitate chemical protein ligation,as exemplified by the synthesis of the single-spanning membrane protein sarcolipin.The iHmb method expands the toolkit for peptide synthesis and ligation and facilitates the preparation of peptides/proteins.

Development review and the prospect of oil shale in-situ catalysis conversion technology

As an unconventional resource, oil shale possesses abundant reserves and significant potential for industrial applications. The rational and efficient development of oil shale resources holds immense importance in reducing national energy demand. In-situ catalytic technology, characterized by its high efficiency, low pollution, and minimal energy consumption, represents a key direction for future oil shale development. This paper provides a comprehensive review of research progress in in-situ oil shale mining technology, oil shale pyrolysis catalysts, the pyrolysis mechanism of kerogen, and the compatibility of different heating processes and catalysts. Furthermore, the paper proposes future research directions and prospects for oil shale in-situ catalytic technology, including reservoir modification, highefficiency catalyst synthesis, injection processes, and high-efficiency heating technology. These insights serve as valuable technical references for the advancement of oil shale in-situ catalytic technology.

Span-based joint entity and relation extraction augmented with sequence tagging mechanism

Span-based joint extraction simultaneously conducts named entity recognition(NER) and relation extraction(RE) in a text span form. However, since previous span-based models rely on span-level classifications, they cannot benefit from token-level label information, which has been proven advantageous for the task. In this paper, we propose a sequence tagging augmented span-based network(STSN), a spanbased joint model that can make use of token-level label information. In STSN, we construct a core neural architecture by deep stacking multiple attention layers, each of which consists of three basic attention units.On the one hand, the core architecture enables our model to learn token-level label information via the sequence tagging mechanism and then uses the information in the span-based joint extraction; on the other hand, it establishes a bi-directional information interaction between NER and RE. Experimental results on three benchmark datasets show that STSN consistently outperforms the strongest baselines in terms of F1,creating new state-of-the-art results.

Photoinduced transposed Paternò-Büchi reaction for effective synthesis of high-performance jet fuel

High-energy-density fuels are important for volume-limited aerospace vehicles,but the increase in fuel energy density always leads to poor cryogenic performance.Herein,we investigated the transposed Paternò-Büchi reaction of biomass cyclic ketone and cyclic alkene to synthesize a new kind of alkyl-substituted polycyclic hydrocarbon fuel with high energy density and good cryogenic performance.The triplet-energy-quenching results and phosphorescent emission spectra reveal the sensitization mechanism of the reaction,including photosensitizer excitation,triplettriplet energy transfer,cyclization,and relaxation,and the possible reaction path was revealed by the density functional theory(DFT)calculations.The reaction conditions of photosensitizer type and addition,molar ratio of substrates,reaction temperature,and incident light intensity were optimized,with the target product yield achieving 65.5%.Moreover,the reaction dynamics of the reaction rate versus the light intensity are established.After the hydrogenation-deoxygenation reaction,three fuels with a high density of 0.864-0.938 g·ml^(-1) and a low freezing point of<-55℃ are obtained.This work provides a benign and effective approach to synthesize high-performance fuels.

In this article, we consider the existence of normalized solutions for the following nonlinear biharmonic Schr?dinger equations:■where c, ε > 0, N ≥ 5, λ ∈ R is a Lagrange multiplier and is unknown, h ∈ C(■N, [0, ∞)), f : R → R is continuous function satisfying L2-subcritical growth. When ε is small enough, we get multiple normalized solutions. Moreover, we also obtain orbital stability of the solutions.
