Carrier-free multi-component self-assembled nano-systems have attracted widespread attention owing to their easy preparation,high drug-loading efficiency,and excellent therapeutic ***,MnAs-ICG nanospike was generated by self-assembly of indocyanine green(ICG),manganese ions(Mn^(2+)),and arsenate(AsO_(4)^(3−))based on electrostatic and coordination interactions,effectively integrating the bimodal imaging ability of magnetic resonance imaging(MRI)and fluorescence(FL)imaging-guided synergistic therapy of photothermal/chemo/chemodynamic therapy within an“all-in-one”theranostic *** as-prepared MnAs-ICG nanospike had a uniform size,well-defined nanospike morphology,and impressive loading *** MnAs-ICG nanospike exhibited sensitive responsiveness to the acidic tumor microenvironment with morphological transformation and dimensional variability,enabling deep penetration into tumor tissue and on-demand release of functional therapeutic *** vitro and in vivo results revealed that MnAs-ICG nanospike showed synergistic tumor-killing effect,prolonged blood circulation and increased tumor accumulation compared to their individual components,effectively resulting in synergistic therapy of photothermal/chemo/chemodynamic therapy with excellent anti-tumor *** together,this new strategy might hold great promise for rationally engineering multifunctional theranostic nano-platforms for breast cancer treatment.

Lacking a precise targeting strategy,castration-resistant prostate cancer(CRPC)is still hard to be treat *** treatment options that can accurately target CPRC is an important issue with urgent *** this study,a novel nanotechnologybased strategy had been developed for the precise target treatment of *** combining microwaves and photothermal therapy(PTT),this nanoplatform,cmHSP70-PL-AuNC-DOX,targets tumor tissues with outstanding precision and achieves better anti-tumor activity by simultaneously eliciting photothermal and chemotherapeutic *** nanotechnology,cmHSP70-modified and thermo-sensitive liposome-coated AuNC-DOX were prepared and used for CRPC-targeted photothermal ablation and ***(DOX)was selected as the chemotherapeutic agent for *** terms of the curative scheme,prostate tissues were firstly pre-treated with microwaves to induce the expression of heat shock protein 70(HSP70)and its migration to the cell membrane,which was then targeted by HSP70 antibody(cmHSP70)coated on the nanoparticles to achieve accurate drug *** nanoplatform then achieved precise ablation and controlled release of DOX under external near-infrared(NIR)*** the implementation,the targeting,cell killing,and safety of this therapeutical strategy had been verified in vivo and in *** work establishes an accurate,controllable,efficient,non-invasive,and safe treatment platform for targeting CRPC,provides a rational design for CRPC’s PTT,and offers new prospects for nanomedicines with great precision.

Identification of TMEM159 as a biomarker of glioblastoma progression based on immune characteristics

Background:Glioblastoma multiforme(GBM)is the most general malignancy of the primary central nervous system that is characterized by high aggressiveness and *** protein 159(TMEM159)is an endoplasmic reticulum protein that can form oligomers with *** TMEM159-seipin complex decides the site of lipid droplet(LD)formation,and the formation of LDs is a marker of ***,the role of TMEM159 in the progression of GBM has not been investigated to ***:In this study,we examined the genes that may be associated with patient prognosis in GBM by bioinformatics analyses,and identified the key genes that affect the development of GBM using single-cell RNA sequencing *** biological functions of TMEM159 in GBM cells were additionally assessed by clone formation and transwell assays as well as using a model of chick embryo chorioallantois membrane(CAM)and western *** association between TMEM159 and epidermal growth factor receptor(EGFR)was finally analyzed in GBM ***:A prognostic model was established and validated for predicting the *** curve analysis showed a critical difference in the prognosis of the high-and low-risk groups predicted by the prognostic *** results demonstrated that TMEM159 affected the proliferation and invasion of GBM *** chick embryo CAM assays demonstrated that the inhibition of TMEM159 expression reduced angiogenesis in the CAM ***:The prognostic model achieved good predictive potential for high-risk *** findings also revealed that TMEM159 might be an important prognostic factor for GBM,indicating that the protein may be a promising therapeutic target for suppressing the development of GBM.

Photothermal/matrix metalloproteinase-2 dual-responsive gelatin nanoparticles for breast cancer treatment

The chemotherapy combined with photothermal therapy has been a favorable approach for the treatment of breast *** present study,nanoparticles with the characteristics of photothermal/matrix metalloproteinase-2(MMP-2)dual-responsive,tumor targeting,and size-variability were designed for enhancing the antitumor efficacy and achieving"on-demand"drug release *** on the thermal sensitivity of gelatin,we designed a size-variable gelatin nanoparticle(GNP)to encapsulate indocyanine green(ICG)and doxorubicin(DOX).Under an 808 nm laser irradiation,GNP-DOX/ICG responded photothermally and swelled in size from 71.58?4.28 to 160.80?9.51 nm,which was beneficial for particle retention in the tumor sites and release of the loaded ***,GNP-DOX/ICG showed a size reduction of the particles to 33.24?4.11 nm and further improved drug release with the degradation of overexpressed MMP-2 in *** the subsequently performed in vitro experiments,it was confirmed that GNP-DOX/ICG could provide a therapeutic effect that was enhanced and ***,GNP-DOX/ICG could efficiently suppress the growth of 4 T1 tumor in *** conclusion,this study may provide a promising strategy in the rational design of drug delivery nanosystems based on gelatin for chemo-photothermal therapy to achieve synergistically enhanced therapeutic efficacy against breast cancer.

pH-sensitive zwitterionic improves tumor targeting efficacy coating of gold nanocages and photothermal treatment

Stealth coating materials effectively extend a nanoparticle＇s systemic circulation lifetime yet limit its cellular internalization, which promotes and prevents tumor targeting, respectively. Here, this contradiction was resolved by using an acutely pH-sensitive zwitterionic stealth ligand capable of responding to small differences in extracellular pH between blood and tumors. Using a photothermal gold nanocage （AuNC） as a model nanotherapeutic, we found that stealth-AuNC nanoparticles showed both significantly enhanced cell uptake efficiency in acidic tumors and a markedly extended systemic circulation lifetime compared to its unaltered analogue. As a result, stealth-AuNC nanoparticles administered intravenously showed significantly enhanced accumulation within the tumor, leading to significantly improved photothermal therapeutic efficacy in mouse models. These results suggests that pH-sensitive zwitterionic ligands with sufficient sensitivity for responding to small differences in extracellular pH between blood and tumors are ideal stealth materials for simultaneously conferring both extended systemic circulation and enhanced cellular internalization, reducing the need for active targeting moieties.

Bunch-length measurement at a bunch-by-bunch rate based on time–frequency-domain joint analysis techniques and its application

This paper presents a new technique for measuring the bunch length of a high-energy electron beam at a bunch-by-bunch rate in storage *** technique uses the time–frequency-domain joint analysis of the bunch signal to obtain bunch-by-bunch and turn-by-turn longitudinal parameters,such as bunch length and synchronous *** bunch signal is obtained using a button electrode with a bandwidth of several *** data acquisition device was a high-speed digital oscilloscope with a sampling rate of more than 10 GS/s,and the single-shot sampling data buffer covered thousands of *** bunch-length and synchronous phase information were extracted via offline calculations using Python *** calibration coefficient of the system was determined using a commercial streak ***,this technique was tested on two different storage rings and successfully captured various longitudinal transient processes during the harmonic cavity debugging process at the Shanghai Synchrotron Radiation Facility(SSRF),and longitudinal instabilities were observed during the single-bunch accumulation process at Hefei Light Source(HLS).For Gaussian-distribution bunches,the uncertainty of the bunch phase obtained using this technique was better than 0.2 ps,and the bunch-length uncertainty was better than 1 *** dynamic range exceeded 10 *** technology is a powerful and versatile beam diagnostic tool that can be conveniently deployed in high-energy electron storage rings.

Macrosegregation is a critical factor that limits the mechanical properties of *** impact of equiaxed crystal sedimentation on macrosegregation has been extensively studied,as it plays a significant role in determining the distribution of alloying elements and impurities within a *** improve macrosegregation in steel connecting shafts,a multiphase solidification model that couples melt flow,heat transfer,microstructure evolution,and solute transport was established based on the volume-averaged Eulerian-Eulerian *** this model,the effects of liquid phase,equiaxed crystals,columnar dendrites,and columnar-to-equiaxed transition(CET)during solidification and evolution of microstructure can be considered *** sedimentation of equiaxed crystals contributes to negative macrosegregation,where regions between columnar dendrites and equiaxed crystals undergo significant A-type positive macrosegregation due to the ***,noticeable positive macrosegregation occurs in the area of final solidification in the *** improvement in macrosegregation is beneficial for enhancing the mechanical properties of connecting *** mitigate the thermal convection of molten steel resulting from excessive superheating,reducing the superheating during casting without employing external fields or altering the design of the ingot mold is indeed an effective approach to control macrosegregation.

Kagome magnets with diverse topological quantum responses are crucial for next-generation topological *** anisotropic magnetism and band evolution induced by ferromagnetic phase transition(FMPT)is reported in a newly discovered titanium-based kagome ferromagnet Sm Ti3Bi4,which features a distorted Ti kagome lattice and Sm atomic zig-zag ***-dependent resistivity,heat capacity,and magnetic susceptibility reveal a ferromagnetic ordering temperature Tc of23.2 K.A large magnetic anisotropy,observed by applying the magnetic field along three crystallographic axes,identifies the b axis as the easy ***-resolved photoemission spectroscopy with first-principles calculations unveils the characteristic kagome motif,including the Dirac point at the Fermi level and multiple van Hove ***,a band splitting and gap closing attributed to FMPT is observed,originating from the exchange coupling between Sm 4 f local moments and itinerant electrons of the kagome Ti atoms,as well as the time-reversal symmetry breaking induced by the long-range ferromagnetic *** the large in-plane magnetization and the evolution of electronic structure under the influence of ferromagnetic ordering,such materials promise to be a new platform for exploring the intricate electronic properties and magnetic phases based on the kagome lattice.
