10H2DA improves PCOS through autophagy-mediated mitochondrial fission dependent on the maintenance of calcium homeostasis between endoplasmic reticulum and mitochondria

Polycystic ovarian syndrome （PCOS） is the most principal reason for infertility in reproductive women， but no versatile treatment is feasible. Although trans-10-Hydroxy-2-decenoic acid （10H2DA） exhibits the anti-inflammatory， anti-cancerous and immunomodulatory properties， its role in improving PCOS remains elusive. This present study examined the effects of 10H2DA on the estrous cycle， ovarian morphology， reproductive hormone， glucose and insulin tolerances of PCOS mice， analyzed the binding between 10H2DA and free fatty acid receptor 4 （FFAR4）， and explored the regulation of 10H2DA on the c-Jun N-terminal kinase （JNK）/JUN （Jun proto-oncogene） signaling， cytosolic calcium ion (Ca2+)， endoplasmic reticulum （ER） Ca2+release， mitochondrial Ca2+， mitochondrial function， autophagy， lysosomal acidification， mitochondrial fission， secretion function and apoptosis of ovarian granulosa cells （GCs） using enzyme-linked immunosorbent assay， flow cytometry. western blotting， molecular docking， various fluorescent probes or plasmid， etc. The results found that 10H2DA alleviated the symptom of PCOS mice through improving the secretion dysfunction of GCs and protecting GCs against apoptosis. After binding to FFAR4 and inactivation of JNK/JUN signaling， 10H2DA restricted the release of ER Ca2+through targeting ryanodine receptors （RYRs） which had been identified as the downstream targets of JUN， diminished cytosolic Ca2+accumulation and prevented Ca2+influx into mitochondria via adjusting the mitochondrial calcium uptake 1 （MICU1） and mitochondrial calcium uniporter （MCU）. Concurrently， 10H2DA kept the integrity of mitochondrial function through maintaining mitochondrial Ca2+homeostasis and suppressed the discharge of mitochondrial reactive oxygen species （mtROS） into the cytosol through blocking the mitochondrial permeability transition pore opening to avoid the lipid peroxidation and ameliorate the secretory function and apoptosis of GCs. Preserved urethral plate urethroplasty for failed hypospadias repair： report of 249 cases

Hypospadias is one of the most common .abnormalities of the external male genitalia but is very difficult to repair. Hypospadias reoperation is also a common practice for pediatric urologists, though repair of hypospadias at reoperation is much more difficult. The surgical technique involves using the well-vascularized ventral penile skin strip as a neourethral plate, and choosing surgical procedures according to the penile conditions. Tubularized incised plate urethroplasty （TIP）, first presented by Snodgrass in 1994, is widely accepted due to the ease of operation and satisfactory postoperative penis appearance. It can be used in most primary hypospadias cases, as well as in reoperations following failed surgery. Many satisfactory results of TIP have been reported in failed hypospadias repair. In this study, we used TIP and the Duplay technique for failed hypospadias repair in patients with the same penile conditions, and obtained similar, satisfactory results with both methods. Under suitable conditions, it is preferable to use a well-vascularized flap to repair failed hypospadias cases, and we therefore performed some Onlay island flap urethroplasties with good results.

Numerical simulation of liquid sloshing in a spherical tank by MPS method

This paper investigates the sloshing phenomena in a spherical liquid tank using the moving particle semi-implicit(MPS)method,a crucial study in fluid *** from previous research focused on rectangular or LNG tanks,this work explores the unique motion patterns inherent to spherical *** accuracy of our in-house MPS solver MLParticle-SJTU is validated against experimental data and finite volume method(FVM).And the MPS method reveals a closer alignment with experimental outcomes,which suggests that MPS method is particularly effective for modeling complex,non-linear fluid *** the fluid’s response to excitation at its natural frequency is simulated,showcasing vigorous sloshing and rotational *** analyses of the fluid motion are conducted by drawing streamline diagrams,velocity vector diagrams,and vorticity *** fluid’s motion response is explored using both time-domain and frequency-domain curves of the fluid centroid,as well as the sloshing force.

Quantum phase transition of the Jaynes-Cummings model

Herein,we propose an experimentally feasible scheme to show the quantum phase transition of the Jaynes-Cummings(JC)model by modulating the transition frequency of a two-level system in a quantum Rabi model with strong *** tuning the modulation frequency and amplitude,the ratio of the effective coupling strength of the rotating terms to the effective cavity(atomic transition)frequency can enter the deep-strong coupling regime,while the counter-rotating terms can be ***,a deep-strong JC model is *** ratio of the coupling strength to resonance frequencies in the deep-strong JC model is two orders of magnitude larger than the corresponding ratio in the original quantum Rabi *** scheme can be employed in atom-cavity resonance and off-resonance cases,and it is valid over a broad *** nonzero average cavity photons of the ground state indicate the emergence of a quantum phase ***,we demonstrate the dependence of the phase diagram on the atom-cavity detuning and modulation *** the parameters used in our scheme are within the reach of current experimental *** scheme provides a new mechanism for investigating the critical phenomena of finite-sized systems without requiring classical field limits,thereby opening a door for studying fundamental quantum phenomena occurring in the ultrastrong and even deep-strong coupling regimes.

Structured-light(SL)based 3D sensors have been widely used in many *** SL is the most widely deployed among all SL sensors due to its light weight,compact size,fast video rate,and low *** transmitter(known as the dot projector)consists of a randomly patterned vertical-cavity surface-emitting laser(VCSEL)array multiplicated by a diffractive optical element(DOE)with a fixed repeated *** that the separation of any two speckles is only one known and fixed number(albeit random),there are no other known scales to cal-ibrate or ***,typical SL sensors require extensive in-factory calibrations,and the depth resolution is limited to 1 mm at-60 cm *** this paper,to the best of our knowledge,we propose a novel dot projector and a new addressable SL(ASL)3D sensor by using a regularly spaced,individually addressable VCSEL array,multiplicated by a metasurface-DOE(MDOE)into a random pattern of the *** turning on or off the VCSELs in the array provides multiple known distances between neighboring speckles,which is used as a“built-in caliper”to achieve higher accuracy of *** as a precise“vernier caliper,”the addressable VCSEL array enables fine control over speckle positions and high detection *** experimentally dem-onstrated that the proposed method can result in sub-hundred-micron level *** new concept opens new possibilities for applications such as 3D computation,facial recognition,and wearable devices.

Microstructure and Internal Friction Behavior of Laser 3D Printed Fe-Based Amorphous Composites

Laser 3D printing,also known as laser additive manufacturing(LAM),is favored for its ability to form bulk metallic glass(BMG)and its composite materials(BMGcs)with freeform *** this work,two different kinds of Fe_(41)Co_(7)Cr_(15)Mo_(14)C_(15)B_(6)Y_(2)amorphous coatings(A and B)were prepared by using LAM technology under air-and water-cooled conditions,respectively;meanwhile,to reduce the cracks generated due to the residual thermal stresses,coating C obtained by air-sweep annealing of B with a low energy-density *** morphology and amorphous content and microstructure of the coatings were investigated,the results show many cracks in coating B deposited under water-cooled conditions,and its microstructure shows an amorphous-crystal-nanocrystalline mixed *** was suppressed in coating C,obtained by air-sweep annealing based on coating B,but the amorphous content was reduced from 32.6 to 13.4%.And the hardness and corrosion resistance of the coating will increase with the increase in the amorphous ***,the internal friction behavior of a BMGcs was prepared on the basis of the process of sample C is compared with that of as-cast amorphous *** results show that the low temperature internal friction behavior of BMGcs is affected by the defects produced during printing,and the high temperature internal friction behavior is affected by the precipitated hard phase.

Machine learning with active pharmaceutical ingredient/polymer interaction mechanism:Prediction for complex phase behaviors of pharmaceuticals and formulations

The high throughput prediction of the thermodynamic phase behavior of active pharmaceutical ingredients(APIs)with pharmaceutically relevant excipients remains a major scientific challenge in the screening of pharmaceutical *** this work,a developed machine-learning model efficiently predicts the solubility of APIs in polymers by learning the phase equilibrium principle and using a few molecular *** the few-shot learning framework,thermodynamic theory(perturbed-chain statistical associating fluid theory)was used for data augmentation,and computational chemistry was applied for molecular descriptors'*** results showed that the developed machine-learning model can predict the API-polymer phase diagram accurately,broaden the solubility data of APIs in polymers,and reproduce the relationship between API solubility and the interaction mechanisms between API and polymer successfully,which provided efficient guidance for the development of pharmaceutical formulations.
