Deubiquitinating enzymes(DUBs) or deubiquitinases facilitate the escape of multiple proteins from ubiquitin-proteasome degradation and are critical for regulating protein expression levels in vivo.Therefore,dissecting the underlying mechanism of DUB recognition is needed to advance the development of drugs related to DUB signaling pathways.To data,extensive studies on the ubiquitin chain specificity of DUBs have been reported,but substrate protein recognition is still not clearly understood.As a breakthrough,the scaffolding role may be significant to substrate protein selectivity.From this perspective,we systematically characterized the scaffolding proteins and complexes contributing to DUB substrate selectivity.Furthermore,we proposed a deubiquitination complex platform(DCP) as a potentially generic mechanism for DUB substrate recognition based on known examples,which might fill the gaps in the understanding of DUB substrate specificity.

To enable the experimental assessment of the seismic performance of full-scale nonstructural elements with multiple engineering parameters(EDPs),a three-layer testbed named Nonstructural Element Simulator on Shake Table(NEST)has been developed.The testbed consists of three consecutive floors of steel structure.The bottom two floors provide a space to accommodate a full-scale room.To fully explore the flexibility of NEST,we propose a novel control strategy to generate the required shake table input time histories for the testbed to track the target floor motions of the buildings of interest with high accuracy.The control strategy contains two parts:an inverse dynamic compensation via simulation of feedback control systems(IDCS)algorithm and an offline iteration procedure based on a refined nonlinear numerical model of the testbed.The key aspects of the control strategy were introduced in this paper.Experimental tests were conducted to simulate the seismic responses of a full-scale office room on the 21^(st)floor of a 42-story high-rise building.The test results show that the proposed control strategy can reproduce the target floor motions of the building of interest with less than 20%errors within the specified frequency range.

Suppose the ground field F is an algebraically closed field characteristic of p>2.In this paper,we investigate the restricted cohomology theory of restricted Lie superalgebras.Algebraic interpretations of low dimensional restricted cohomology of restricted Lie superalgebra are given.We show that there is a family of restricted model filiform Lie superalgebra Lλp,p structures parameterized by elementsλ∈Fp.We explicitly describe both the 1-dimensional ordinary and restricted cohomology superspaces of Lλp,p with coefficients in the 1-dimensional trivial module and show that these superspaces are equal.We also describe the 2-dimensional ordinary and restricted cohomology superspaces of Lλp,p with coefficients in the 1-dimensional trivial module and show that these superspaces are unequal.

A series of chemotherapeutic drugs that induce DNA damage,such as cisplatin(DDP),are standard clinical treatments for ovarian cancer,testicular cancer,and other diseases that lack effective targeted drug therapy.Drug resistance is one of the main factors limiting their application.Sensitizers can overcome the drug resistance of tumor cells,thereby enhancing the antitumor activity of chemotherapeutic drugs.In this study,we aimed to identify marketable drugs that could be potential chemotherapy sensitizers and explore the underlying mechanisms.We found that the alcohol withdrawal drug disulfiram(DSF)could significantly enhance the antitumor activity of DDP.JC-1 staining,propidium iodide(PI)staining,and western blotting confirmed that the combination of DSF and DDP could enhance the apoptosis of tumor cells.Subsequent RNA sequencing combined with Gene Set Enrichment Analysis(GSEA)pathway enrichment analysis and cell biology studies such as immunofluorescence suggested an underlying mechanism:DSF makes cells more vulnerable to DNA damage by inhibiting the Fanconi anemia(FA)repair pathway,exerting a sensitizing effect to DNA damaging agents including platinum chemotherapy drugs.Thus,our study illustrated the potential mechanism of action of DSF in enhancing the antitumor effect of DDP.This might provide an effective and safe solution for combating DDP resistance in clinical treatment.

Quantitative characterization of tight sandstone oil migration and accumulation is an emerging research frontier in the field of oil and gas exploration.In this study,a conceptual model containing multiple basic geological elements is developed,and a nonlinear seepage numerical model for tight sandstone oil migration and accumulation is established.The effects of the slip effect,overpressure driving force,buoyancy,and capillary force on the migration and accumulation of tight oil are examined.The results showed that(1)the differences in oil migration and accumulation between tight and conventional reservoirs are reflected in the growth mode of oil saturation,distribution characteristics of oil and water,and extent of the effect of the formation dip angle;(2)the slip effect has a significant impact when the average pore throat radius is less than 150 nm and the overpressure driving force and capillary force are the main mechanical mechanisms controlling oil migration and accumulation in tight sandstone,while the coupling effect of buoyancy,capillary force,and overpressure driving force controls the upper and lower limits of oil saturation.Finally,a dimensional and dimensionless identification chart for rapidly predicting the oil saturation of tight sandstone is proposed and verified using the measured data.This study provides a basis for analyzing the migration and accumulation mechanisms of tight sandstone oil and a new approach for predicting oil saturation.Additionally,we developed digital and visual analysis methods for the migration results,enriching the expression of the dynamics of hydrocarbon accumulation.

Propensity score is widely used to estimate treatment effects in observational studies.The covariate adjustment using propensity score is the most straightforward method in the literature of causal inference.In this article,we estimate the survival treatment effect with covariate adjustment using propensity score in the semiparametric accelerated failure time model.We establish the asymptotic properties of the proposed estimator by simultaneous estimating equations.We conduct simulation studies to evaluate the finite sample performance of the proposed method.A real data set from the German Breast Cancer Study Group is analyzed to illustrate the proposed method.

LnMnO_(3+σ)(Ln=La,Pr,Nd,Sm,Eu,Gd or Dy)perovskites synthesized by sol-gel method were employed for gaseous elemental mercury(Hg^(0))removal from coal-fired flue gas.Characterization results revealed the structure of the perovskites presented a phase transition process from rhombohedral system to O-and O'-orthorhombic structure with the change of A-site rare earth elements.The perovskites showed satisfactory Hg^(0) removal capacity in a narrow temperature range of 100-150℃.NdMnO_(3+σ) with an O-O'orthorhombic structure presented the best Hg^(0) removal performance,which markedly depends on four factors:crystal structure,oxygen vacancy density,Mn^(4+)/Mn^(3+) ratio and surface element segregation.The Hg^(0) removal mechanism was illustrated based on the mercury temperature programmed desorption experiment and X-ray photoelectron spectroscopy characterization.Both chemisorption and catalytic oxidation played a role in the Hg^(0) removal process.Chemisorption dominated the Hg^(0) removal,due to the slow catalytic oxidation rate at low temperature.This work preliminarily established the relation between the structure of rare earth manganese perovskite and Hg^(0) removal performance.

Neurodegenerative diseases,including Alzheimer's disease(AD),frontotemporal dementia,Parkinson's disease,and dementia with Lewy bodies,represent tremendous unmet clinical needs.A common feature of these diseases is the aberrant cerebral accumulation of pathological protein aggregates,affecting selectively vulnerable circuits in a disease-specific pattern.Earlier studies have established a relationship between abnormal aggregation and neuronal dysfunction or loss,suggesting multifactorial pathogenesis mechanisms in these neurodegenerative disorders.
