Background Solamargine （SM）, a steroidal glycoalkaloid isolated from the Chinese herb Solanum incanum, has been shown to inhibit the growth of some cancer cell lines and induce significant apoptosis. However, the effects of SM on multidrug-resistant （MDR） cells and the molecular mechanisms involved are poorly understood. The purpose of this study was to evaluate the anti-MDR effects of SM and the associated mechanisms in MDR K562/A02 cells. Methods The cytotoxicity of SM was measured by 3-（4,5-dimethylthiazol）-2,5-diphenyltetrazolium bromide （MTT） assay. The 14＇,6-diamidino-2-phenylindole （DAPi） nuclear staining and flow cytometry were used to detect SM-induced apoptosis. The mRNA expression of P-glycoprotein （P-gp） was investigated by real-time PCR （RT-PCR）. Western blotting was used to determine the expression of Bcl-2, Bax, and actin. The changes in the morphology of actin were examined with immunofluorescence staining. Results MTT results showed that SM effectively killed the MDR sublines K562/A02, KB/VCR, and H460/paclitaxel （Taxol）, and their parental cell lines K562, KB, and H460 to an equivalent or more sensitive degree. Based on the results by flow cytometry and immunostaining, the pro-apoptotic effects of SM were observed in MDR K562/A02 cells. Furthermore, the RT-PCR results showed that SM induced the downregulation of MDR1 mRNA. In addition, the expression of P-gp and actin was decreased in the SM-treated cells, as measured by western blotting and immunostaining. Conclusions These results demonstrate that SM effectively triggers apoptosis in MDR tumor cells, which is associated with actin disruption and downregulation of MDR1 expression. This compound may merit further investigation as a Potential therapeutic aaent that bvDasses the MDR mechanism for the treatment of MDR tumors.

The microbial transformation of glycyrrhetinic acid（1） by Cunninghamella blakesleana CGMCC 3.970 led to the production of five new metabolites（2-6）.The structures of the metabolites were determined by extensive spectroscopic（HR-ESIMS,1D and 2D NMR） data *** involved reactions exhibited specific hydroxylations at C-24,C-7,and C-15,and oxidation at ***,compounds 2,5,and 6showed significant neural anti-inflammatory activity by inhibiting lipopolysaccharide-induced NO production in mouse microglia BV2 cells with IC（50） values of 0.76,0.94,and 0.16μmol/L,respectively.

旨在制备禽腺病毒4型(FAdV-4)Fiber-2蛋白的单克隆抗体并建立Fiber-2蛋白的定量检测方法。本研究将原核表达的Fiber-2蛋白免疫BALB/c雌鼠,通过细胞融合技术制备杂交瘤细胞,并通过免疫印迹、间接免疫荧光和抗体相加试验筛选杂交瘤细胞株;以制备的单克隆抗体为捕获抗体和酶标检测抗体,通过优化捕获抗体包被浓度和酶标抗体稀释度等条件,建立特异性检测Fiber-2蛋白的双抗体夹心ELISA方法。结果:本研究成功筛选到3株能够稳定传代的杂交瘤细胞株1H2、2A9和3E1,其分泌的抗体均与FAdV-4全病毒具有良好的反应性,可识别不同抗原表位;进一步发现,以1H2为捕获抗体、2A9为酶标检测抗体,能够建立检测Fiber-2蛋白的ELISA方法;该方法具有良好的重复性,批内重复及批间重复试验变异系数均小于10%;敏感性高,Fiber-2蛋白检测限为0.078 ng/μL;特异性好,不与FAdV-4的其他结构蛋白及鸭腺病毒3型的Fiber-2蛋白发生反应。综上,本研究成功制备了3株Fiber-2蛋白单克隆抗体,并建立了定量检测Fiber-2蛋白的双抗体夹心ELISA方法,为后续FAdV-4基础研究和Fiber-2亚单位疫苗研究奠定了物质基础。

为探讨气候变化和区域人类活动对黄河上游龙羊峡至积石峡段流域植被(Normalized difference vegetation index, NDVI)的影响,本研究以位于黄河上游龙羊峡至积石峡段流域为研究区,基于MODI3Q1数据集,采用Theil Sen斜率统计法、Mann-Kendall检验及相关分析法等6种方法,评价了区内2004—2021年间NDVI的时空变化特征,并探讨了NDVI与平均气温、降雨量、蒸发量、太阳辐射量间的相关性及其驱动力。结果表明:2004—2021年间区内81.66%地区的NDVI总体呈现出逐渐增加的变化趋势;NDVI与平均气温、蒸发量、太阳辐射量之间的均呈正相关关系,正相关区域面积占总面积的96.72%,98.96%,72.73%,92.64%。此外,通过进一步分析因子探测器结果得到,区内土地利用类型为耕地、森林、灌木区域内影响植被生长的主要影响因子为平均气温,其值分别为0.216 5,0.321 5,0.326 5。本研究结果对于黄河上游流域开展植被空间分布变化监测、植被生态安全防护具有实际指导意义。

Deltas are densely populated industrialized regions,and home to important ports and navigation *** to human interferences,the Yangtze Estuary has experienced a significant reduction in sediment load caused by the Three Gorges Dam(TGD),as well as adjustment to local morphodynamics by the Deep-water Navigation Channel(DNC).While the dramatic reduction in sediment triggers the increased channel erosion,the deposition flux of the DNC located at the estuary mouth has little *** explore the physical mechanism of this phenomenon,a two-dimensional model is used to establish the relationship between sediment load and the sediment budget of channels based on the bathymetry in *** results show that the tidal reach and the inner estuary have a negative sediment *** the seasonal characteristics of water and sediment fluxes become less obvious *** analysis shows that the influence of upstream sediment load on deposition flux decreases along the channel,with a transition from deposition to erosion occurring in the tidal *** the last-level bifurcation,the annual siltation of sediment in the North Passage(NP)decreases by 4.5%with low *** is attributed to the reduction of sediment load partially mitigated by riverbed erosion and cascade *** addition,the lateral sediment supply,which accounts for 68%of the sediment input in the NP,is ***,this study strengthens the understanding of the relationship between sediment load and artificially deepened systems,thus allowing for better management of estuarine sediment and navigation channel.

Background Invasive aspergillosis （IA）, which is mainly caused by Aspergillus fumigatus （A. fumigatus）, is a major cause of morbidity and mortality in immunocompromised patients. Despite considerable progress in currently available antifungals the mortality still remains high in critically ill patients. U0126 which is a highly selective inhibitor of MEK1 and MEK2 in the RAF/MEK/ERK pathway in mammalian cells has been demonstrated to have an anti-proliferative role in cancer cells. The purpose of this study was to explore the role of U0126 on growth inhibition and activation of mitogen-activated protein kinases （MAPKs） in A. fumigatus. Methods Germination percentage and hyphae growth in A. fumigatus treated with U0126 were observed and compared with untreated controls. Western blotting analysis was used to detect changes in activation of SakA, MpkA and MpkB. Results U0126 inhibited germination and hyphae growth in A. fumigatus and enhanced the phosphorylation of SakA and MpkA under oxidative stress. U0126 at 10 pmol/L did not block the activation of MpkB during nitrogen starvation stress. Conclusion U0126 shows promise as an antifungal candidate and the MAPK pathway may be a possible antifungal drug target for A. fumigatus.

Background： Antiphospholipid syndrome （APS）-related immune factors are considered as an important cause of recurrent spontaneous abortion （RSA）. Anticoagulant and anti-inflammatory treatments are believed to effectively improve adverse pregnancy outcomes by affecting the abnormal autoimmune response of the maternal-fetal interface. The aim of this study was to observe the clinical characteristics and treatment outcomes of anticoagulant regimens and anti-inflammatory plus anticoagulation regimens for APS-related RSA. Methods： APS-related RSA cases from September 2011 to September 2016 at Peking University Third Hospital were retrospectively analyzed. The patients were assigned to study group （anti-inflammation plus anticoagulation） and control group （simple anticoagulation）, The incidence of repeat abortion, the incidence of placental dysfunction, the gestational weeks of pregnancy, and the mean weight of the fetus were observed. Results： The pregnancy and neonatal outcome indicators of the repeat pregnancy loss rate （ 11. 11% vs. 22.70%）, placental dysfunction-related diseases （6.35% vs. 15.60%）, the mean birth weight of infants born after 24 weeks gestation （3152.4 ± 844.67 g vs. 2765.76 ± 816.40 g）, full-ternl delivery weight （3456.28 ±419.79 g vs. 3076.18±518.79 g）, the proportions of low birth weight infants （ 12.70% vs. 21.98%）, and small for gestational age （6.35% vs. 14.18%） differed significantly between the study and control groups （all P 〈 0.05）. The incidence of preterm delivery, term delivery, and stillbirth was not significantly different between the two groups, and there was no significant difference between the study and control groups in gestational age at birth （37.6 ± 3.3 weeks vs. 36.9 ± 3.2 weeks： P 〉 0.05）. Conclusion： The anti-inflammatory and anticoagulation regimen is more effective than the simple anticoagulation regimen in the treatlnent of APS recurrent abortion.

Diabetic nephropathy, one of the major causes of death of diabetes patients, is diagnosed as the thickening of glomerular basement membrane and progressive expansion of the glomerular mesangium and tubulointerstitium. Intensive studies have shown that hyperglycemia is the key factor for renal sclerosis which can lead to end-stage renal disease for diabetic patients.^1,2 Our previous studies demonstrated that Xiaoke granule can inhibit the progression of diabetic nephropathy. However, its mechanisms remain unknown.^3,4 In this study, we found that Xiaoke granule coincidently depresses transforming growth factor-beta1 （TGF-β1） expression and inhibits the effect of high glucose on mesangial cell proliferation. This might suggest that the effect of Xiaoke granule on inhibiting progression of diabetic nephropathy through down-regulating TGF-β1 expression.
