The mechanism regulating proteasomal activity under proteotoxic stress conditions remains ***,we showed that arsenite-induced proteotoxic stress resulted in upregulation of Arabidopsis homologous PUB22 and PUB23 U-boxE3 ubiquitin ligases and that pub22 pub23 double mutants displayed arsenite-insensitive seed germination and root growth ***22/PUB23 downregulated 26 S proteasome activity by promoting the dissociation of the 19 S regulatory particle from the holo-proteasome complex,resulting in intracellular accumulation of UbG76 VGFP,an artificial substrate of the proteasome complex,and insoluble poly-ubiquitinated *** results suggest that PUB22/PUB23 play a critical role in arsenite-induced proteotoxic stress response via negative regulation of 26 S proteasome integrity.

Dear Editor,Here, we report the developmental regulation of proteolytic cleavage of the FLOWERING LOCUS T （FT） protein. FT, a mem- ber of the phosphatidylethanolamine-binding protein （PEBP） family, functions as a florigen or a component of florigen. PEBP binds phosphoethanolamine, and its ligand-binding domain is evolutionarily conserved from bacteria to plants and animals （Schoentgen and Jolles, 1995; Bradley et al., 1996）. AIM: To determine the prevalence and risk factors of work-related musculoskeletal disorders in gastrointestinal endoscopists in Korea. METHODS: A survey of musculoskeletal symptoms, using a self-administered questionnaire, was conducted on 55 endoscopists practicing in general hospitals or health promotion centers. RESULTS: Forty-nine (89.1%) endoscopists reported musculoskeletal pain on at least one anatomic location and 37 (67.3%) endoscopists complained of pain at rest. Twenty-six (47.3%) endoscopists had severe musculoskeletal pain defined as a visual analogue score greater than 5.5. Factors related to the development of severe pain were (1) standing position during upper endoscopy, (2) specific posture/ habit during endoscopic procedures, and (3) multiple symptomatic areas. Finger pain was more common in beginners, whereas shoulder pain was more common in experienced endoscopists. Sixteen percent of symptomatic endoscopists have modified their practice or reduced the number of endoscopic examinations. Only a few symptomatic endoscopists had sought professional consultation with related specialists. CONCLUSION: The prevalence of musculoskeletal pain in endoscopists is very high. The location of pain was different between beginners and experienced endoscopists. Measures for the prevention and adequate management of endoscopy-related musculoskeletal symptoms are necessary.

Highly immunosuppressive tumor microenvironment containing various protumoral immune cells accelerates malignant transformation and treatment *** particular,tumor-associated macrophages(TAMs),as the predominant infiltrated immune cells in a tumor,play a pivotal role in regulating the immunosuppressive tumor *** a potential therapeutic strategy to counteract TAMs,here we explore an exosome-guided in situ direct reprogramming of tumor-supportive M2-polarized TAMs into tumor-attacking M1-type *** derived from M1-type macrophages(M1-Exo)promote a phenotypic switch from anti-inflammatory M2-like TAMs toward pro-inflammatory M1-type macrophages with high conversion *** M1 macrophages possessing protein-expression profiles similar to those of classically activated M1 macrophages display significantly increased phagocytic function and robust cross-presentation ability,potentiating antitumor immunity surrounding the ***,these M1-Exo also lead to the conversion of human patient-derived TAMs into M1-like macrophages that highly express MHC class II,offering the clinical potential of autologous and allogeneic exosome-guided direct TAM reprogramming for arming macrophages to join the fight against cancer.

Efficacy of early treatment with infliximab in pediatric Crohn’s disease

AIM: To investigate the effectiveness of early infliximab use for induction and maintenance therapy in pediatric Crohn’s disease. METHODS: We performed a retrospective chart review of 36 patients with Crohn’s disease. Ten patients (group A) were treated with mesalamine after induction therapy with oral prednisolone, and 13 patients (group B) were treated with azathioprine after induction therapy with oral prednisolone. Thirteen patients (group C) received infliximab and azathioprine for induction and maintenance therapy for the first year, and were treated with azathioprine after 1 year. All patients were followed for at least 24 mo. Efficacy was determined by the relapse rate using the pediatric Crohn’s disease activity index score in each group at 12 and 24 mo. RESULTS: At the 1 year follow-up, the relapse ratee, Mi jin kim, Hae Jeong Lee, Yon Ho Choe (23.1%, 3 of 13 patients) in group C was lower than that (61.5%, 8 of 13 patients) in group B (P = 0.047). At the 2 years follow-up, the relapse rate (38.5%, 5 of 13 patients) in group C was lower than that (76.9%, 10 of 13 patients) in group B (P = 0.047). Adverse events in group C were fewer than in groups A and B. CONCLUSION: Early induction with infliximab at diagnosis, known as "top-down" therapy, was effective for reducing the relapse rate compared to conventional therapies for at least 2 years.

This paper aim is to examine the optimal pricing and return policies for false failure returns in a dual-channel supply chain. Four prevailing return policies in which a manufacturer both operates an E-shop and sells its product through a brick-and-mortar retailer are analyzed, i.e. （I） the manufacturer handlings E-shop＇s returns, while the retailer addresses brick-and-mortar store＇s returns （NR）; （II） the retailer tackles the whole （both E-shop＇s and brick-and-mortar store＇s） returns （ORR）; （III） the manufacturer tackles the whole returns （ORM）; and （IV） the manufacturer and the retailer are jointly responsible for the whole returns （RRM）. Firstly, the optimal pricing and return policies comparing these four scenarios under uniform-pricing strategy are presented. Our conclusions show that the ORR is an optimal return policy. Compared with the NR, consumers will get a lower product pricing under the ORR and a higher product pricing under the ORM. With regard to the RRM, the product pricing is depended on consumer preference, return-rates of the E-shop and the brick-and-mortar store. Then, the optimal pricing and return policies are analyzed under differential-pricing strategy by conducting two-stage sequential games between the manufacturer and the retailer. The findings show that if consumers in the market prefer to purchase via the E-shop, the ORR is an optimal return policy. Otherwise, the NR is the optimal return policy. Compared with the NR, the ORR retailer＇s product pricing will rely on the retailer＇s and the manufacturer＇s return-costs; the RRM retailer＇s product pricing will depend on the return-costs of the retailer and the manufacturer, the return-rates of the E-shop and the brick-and-mortar store and so on. Finally, the influences of the manufacturer and the retailer establishing a Buy-back contract are discussed. Intracardic air embolism is uncommon, however it is a serious condition which frequently leads to catastrophic complications during or after invasive procedures resulting in significant morbidity and mortality. For a successful re- suscitation of patients with intracardiac air embolism, hy- perbaric oxygen therapy is critical.~~2~ Still, due to a hyper- baric chamber＇s unavailability at many hospitals and con- cerns about patient movement resulting in further embolism during transport, such treatment method is gradually dimin- ishing.

The effects of pre-existing {10–12} extension twins on the precipitation behavior of an extruded AZ80 material during aging and on its mechanical properties after peak aging are investigated. The material containing {10–12} twins-which are formed by compression before aging(twinned material)-has a finer grain size and higher dislocation density than the extruded material. Although the peak hardnesses of the twinned and extruded materials are almost the same, the time to reach the peak hardness is considerably shorter in the former material than in the latter(4 h and 24 h, respectively). In the twinned material, the high dislocation density of the {10–12} twins promotes continuous precipitation, which results in the formation of numerous fine Mg17Al12precipitates within the twins in the early stage of *** formation of these continuous precipitates reduces the driving force for discontinuous precipitation, which consequently suppresses the formation and growth of coarse Mg17Al12precipitates at the grain boundaries. Despite its shorter peak-aging time, the 4 h-peak-aged twinned material shows higher tensile strength and elongation than the 24 h-peak-aged extruded material. These higher mechanical properties of the former material are attributed primarily to the presence of more abundant fine continuous precipitates, which are effective in strengthening the material, and less abundant coarse discontinuous precipitates, which can act as crack initiation sites. These results demonstrate that the introduction of {10–12} twins into wrought Mg–Al-based alloys can accelerate the Mg17Al12precipitation kinetics considerably and improve the strength and ductility of the peak-aged alloys simultaneously.
