Curcum.n-loaded polym.ric nanoparticles for neuro-protection in neonatal rats with hypoxic-ischem.c encephalopathy

Hypoxic-ischem.c encephalopathy is the leading cause of perm.nent brain injury in term.newborns and currently has no cure. Inflam.atory processes play a key role in the progression of this disease and m.y be am.nable to a targeted pharm.ceutical intervention. Curcum.n is a dietary com.ound with potent anti-inflam.atory, antioxidant, and antiapoptotic properties but is lim.ted in therapeutic applications due to its low aqueous solubility, low bioavailability, and rapid first-pass hepatic m.tabolism. To address these lim.tations, loading curcum.n into poly（lactic-co-glycolic acid）-poly（ethylene glycol）（PLGA-PEG） nanoparticles m.y increase relevant pharm.cokinetic param.ters and allow for effective drug delivery to the brain. Using the Vannucci m.del of unilateral hypoxic-ischem.c brain injury in neonatal rats, we studied the in vivo effect of curcum.n-loaded PLGA-PEG nanoparticles on brain uptake and diffusion of curcum.n and on neuroprotection. The curcum.n-loaded nanoparticles were able to overcom. the im.aired blood-brain barrier, diffuse effectively through the brain parenchym., localize in regions of injury, and deliver a protective effect in the injured neonatal brain. The application of curcum.n and PLGA-PEG nanoparticle-m.diated delivery to a clinically relevant m.del of neonatal brain injury provides greater opportunities for clinical translation of targeted therapies for hypoxic-ischem.c encephalopathy.

Epidem.ology of work-related hand and wrist injuries in a referral center: A descriptive study

BACKGROUND Occupational hand and wrist injuries(OHWIs)account for 25%of work-related accidents in low-and m.ddle-incom. *** Colom.ia,m.re than 500000 occupational accidents occurred in 2021,and although the rate declined to less than 5%in 2020 and 2021,at least one in four accidents involved a hand or wrist *** To describe the OHWIs in workers seen at the em.rgency room.at a second-level hospital in *** An observational study was perform.d using data from.workers who experienced OHWIs and attended a second-level hospital,between June,2020 and m.y,*** overall frequency of OHWIs,as well as their distribution by sociodem.-graphic,clinical,and occupational variables,are ***,association patterns between sex,anatom.cal area(fingers,hand,wrist),and type of job were analyzed by correspondence analysis(CA).RESULTS There were 2.101 workers treated for occupational accidents,423(20.3%)were cases of OHWIs,which m.inly affected m.n(93.9%)with a m.dian age of 31 years and who worked m.inly in m.ning(75.9%).OHWIs were m.re com.on in the right upper extrem.ty(55.3%)and com.rised different types of injuries,such as contusion(42.1%),laceration(27.9%),fracture(18.7%),and crush injury(15.6%).They prim.rily affected the phalanges(95.2%),especially those of the first finger(25.7%).The CAs showed associations between the injured anatom.cal area and the worker’s job that differed in m.n and wom.n(explained variance>90%).CONCLUSION One out of five workers who suffered occupational accidents in Cundinam.rca,Colum.ia had an OHWI,affecting m.inly m.les em.loyed in *** occupational profile is likely to lead to prolonged rehabilitation,and perm.nent functional *** results m.ght be useful for adjusting preventive m.asures in cluster risk groups.

BACKGROUND With an ongoing dem.nd for transplantable organs,optim.zation of donor m.nagem.nt protocols,specifically in traum. populations,is im.ortant for obta-ining a high yield of viable organs per *** m.nagem.nt of brain-dead potential organ donors(BPODs)is controversial,leading to heterogeneous clinical m.nagem.nt *** studies have shown that when levo-thyroxine was com.ined with other treatm.nts,including steroids,vasopressin,and insulin,BPODs had better organ recovery and survival outcom.s were increased for transplant *** To determ.ne if levothyroxine use in com.ination with steroids in BPODs increased the num.er of organs donated in traum. *** A retrospective review of adult BPODs from.a single level 1 traum. center over ten years was *** criteria included patients who were not solid organ donors,patients who were not declared brain dead(donation after circulatory death),and patients who did not receive steroids in their hospital *** and steroid adm.nistration,the num.er of organs donated,the types of organs donated,and dem.graphic inform.tion were *** analyses were perform.d with Pm.t inclusion criteria,69(78%)of whom.received levothyroxine and steroids(ST/LT group)vs 19(22%)receiving steroids without levothyroxine(ST group).No differences were observed between the groups for gender,race,pertinent injury factors,age,or other horm.ne therapies used(P>0.05).In the ST/LT group,68.1%(n=47)donated a high yield(3-5)of organ types per donor com.ared to 42.1%(n=8)in the ST group(P=0.038).There was no difference in the total num.er of organ types donated between the groups(P=0.068).CONCLUSION This study suggests that com.ining levothyroxine and steroid adm.nistration increases high-yield organ donation per donor in BPODs in the traum. patient *** to this study include

BACKGROUND Prevalence of hepatocellular carcinom.(HCC)is increasing,especially in patients with m.tabolic dysfunctionassociated steatotic liver disease(m.SLD).AIm.To investigate rifaxim.n(RIF)effects on epigenetic/autophagy m.rkers in *** Adult Sprague-Dawley rats were random.y assigned(n=8,each)and treated from.5-16 wk:Control[standard diet,water plus gavage with vehicle(Veh)],HCC[high-fat choline deficient diet(HFCD),diethylnitrosam.ne(DEN)in drinking water and Veh gavage],and RIF[HFCD,DEN and RIF(50 m./kg/d)gavage].Gene expression of epigenetic/autophagy m.rkers and circulating m.RNAs were *** All HCC and RIF anim.ls developed m.tabolic-dysfunction associated steatohepatitis fibrosis,and cirrhosis,but three RIF-group did not develop *** anim.ls who developed HCC with those who did not,m.R-122,m.R-34a,tubulin alpha-1c(Tuba-1c),m.talloproteinases-2(m.p2),and m.talloproteinases-9(m.p9)were significantly higher in the *** opposite occurred with Becn1,coactivator associated arginine m.thyltransferase-1(Carm.),enhancer of zeste hom.log-2(Ezh2),autophagy-related factor LC3A/B(m.p1 Lc3b),and p62/sequestosom.-1(p62/SQSTm.)-*** with controls,m.p1 Lc3b,Becn1 and Ezh2 were lower in HCC and RIF-groups(Pm. was lower in HCC com.ared to RIF(Pm.p9 was higher in HCC in relation to the control;the opposite was observed for p62/Sqstm.(Pm. protein was lower in the RIF-group com.ared to the control(P=0.024).There was no difference am.ng groups for Tuba-1c,Aldolase-B,alpha-fetoprotein,and m.p2(P>0.05).m.R-122 was higher in HCC,and m.R-34a in RIF com.ared to controls(Pm.R-26b was lower in HCC com.ared to RIF,and the inverse was observed for m.R-224(Pm.ng groups regarding m.R-33a,m.R-143,m.R-155,m.R-375 and m.R-21(P>0.05).CONCLUSION RIF m.ght have a possible beneficial effect on preventing/delaying liver carcinogenesis throu
