Antibodies are employed ubiquitously in biomedical sciences,including for diagnostics and *** of the most important uses is for immunohistochemical(IHC)staining,a process that has been improving and evolving over *** is useful when properly employed,yet misuse of the method is widespread and contributes to the“reproducibility crisis”in *** report some of the common problems encountered with IHC assays,and direct readers to a wealth of literature documenting and providing some solutions to this *** also describe a series of vignettes that include our approach to analytical validation of antibodies and IHC assays that have facilitated a number of biological insights into prostate cancer and the refutation of a controversial association of a viral etiology in *** postulate that a great deal of the problem with lack of accuracy in IHC assays stems from the lack of awareness by researchers for the critical necessity for end-users to validate IHC antibodies and assays in their laboratories,regardless of manufacturer claims or past *** suggest that one reason for the pervasive lack of end-user validation for research antibodies is that researchers fail to realize that there are two general classes of antibodies employed in ***,there are antibodies that are“clinical grade”reagents used by pathologists to help render diagnoses that influence patient *** diagnostic antibodies,which tend to be highly validated prior to clinical implementation,are in the vast minority(e.g.lass of antibodies are“research grade”antibodies(now numbering>3800000),which are often not extensively validated prior to *** increased awareness of the problem,both the United States,National Institutes of Health and some journals are requiring investigators to provide evidence of specificity of their antibody-based assays.

HIIT对高脂喂养ApoE敲除鼠骨骼肌氧化应激的影响及肌肉因子BAIBA的保护作用

目的:探讨高强度间歇训练(high-intensity interval training,HIIT)对高脂喂养的载脂蛋白E(ApoE)敲除(Apo^(E-/-))小鼠骨骼肌氧化应激的改善作用以及肌肉因子β-氨基异丁酸(β-aminoisobutyic acid,BAIBA)在其中的可能调节机制。方法:9周龄健康雄性C57Bl/6J小鼠和Apo^(E-/-)小鼠分别喂食标准和高脂饲料,其中C57Bl/6J小鼠为基线对照组(WT,n=6),Apo^(E-/-)小鼠经高脂饲料喂养12周后随机分为Apo^(E-/-)安静组(Apo^(E-/-),n=10)和Apo^(E-/-)高强度间歇训练组(Apo^(E-/-)HIIT,n=10),Apo^(E-/-)HIIT组进行6周HIIT,WT和Apo^(E-/-)组在笼中自由活动。最后一次运动后48 h取小鼠两侧腿部腓肠肌进行氧化损伤、抗氧化及BAIBA生成相关指标的检测。随后采用l-BAIBA预孵育C2C12细胞,后用过氧化氢(H_(2)O_(2))诱导氧化应激,结合干预条件随机分为Control、H_(2)O_(2)和l-BAIBA+H_(2)O_(2)组。相差显微镜观察细胞形态,并检测细胞ROS水平和Nrf2介导的抗氧化通路的相关指标。结果:1)与WT组相比,Apo^(E-/-)组小鼠骨骼肌氧化损伤相关的4-HNE修饰蛋白、蛋白质羰基含量和ROS水平显著升高,而抗氧化相关的p-Nrf2蛋白表达和GSH含量显著降低;而与Apo^(E-/-)组相比,Apo^(E-/-)HIIT组小鼠骨骼肌4-HNE修饰蛋白和蛋白质羰基含量明显降低,p-Nrf2蛋白表达和GSH含量明显升高。2)Apo^(E-/-)HIIT组小鼠骨骼肌生成BAIBA的相关酶基因Acads和Hadha mRNA的表达水平显著高于Apo^(E-/-)组。3)与Control组相比,H_(2)O_(2)干预后C2C12细胞明显皱缩,活细胞数量降低,ROS水平显著增加;而与H_(2)O_(2)组相比,l-BAIBA+H_(2)O_(2)组活细胞数量增加,形态较饱满,ROS水平明显降低。4)与Control组相比,H_(2)O_(2)显著降低了细胞Nrf2、p-Nrf2及其介导的抗氧化酶SOD1、NQO1和CAT蛋白以及合成GSH的相关酶基因Gss、Gsr和Gclm mRNA的表达水平,并显著降低了细胞GSH含量;而l-BAIBA+H_(2)O_(2)组较H_(2)O_(2)组则显著增加了p-Nrf2、SOD1和NQO1蛋白以及Gss、Gsr、Gclc和Gclm mRNA的表达量,同时细胞GSH含量也显著增加。结论:6周HIIT降低了高脂喂养的Apo^(E-/-)小鼠骨骼肌的氧化损伤,其中肌肉因子BAIBA对Nrf2介导的抗氧化信号的激活可能起着一定的作用。

Background:Newly diagnosed breast cancer patients experience symptoms that may affect their quality of life,treatment outcomes,and *** and managing breast cancer-related symptoms soon after diagnosis is *** purpose of this study was to investigate the associations between health-related fitness(HRF)and patient-reported symptoms in newly diagnosed breast cancer ***:This study utilized baseline data from the Alberta Moving Beyond Breast Cancer Cohort Study that were collected within 90 days of *** measures included peak cardiopulmonary fitness(peak volume of oxygen consumption(VO_(2peak))),maximal muscular strength and endurance,flexibility,and body *** measures included depression,sleep quality,and *** multivariable logistic regression was performed for ***:Of 1458 participants,51.5%reported poor sleep quality,26.5%reported significant fatigue,and 10.4%reported moderate *** multivariable-adjusted models,lower relative VO_(2peak)was independently associated with a greater likelihood of all symptom measures,including moderate depression(pleep quality(p=0.009),significant fatigue(p=0.008),any symptom(pltiple symptoms(pld associations with all symptom measures such that all 3 lower quartiles exhibited similar elevated risk compared to the highest *** strength of the threshold associations varied by the symptom measure with odds ratios ranging from-1.5 for poor sleep quality to-3.0 for moderate depression and multiple ***,lower relative upper body muscular endurance was also independently associated with fatigue in a dose-response manner(p=0.001),and higher body weight was independently associated with poor sleep quality in an inverted U pattern(p=0.021).Conclusion:Relative VO_(2peak)appears to be a critical HRF component associated with multiple patient-reported symptoms in newly diag

COVID-19:Reducing the risk via diet and lifestyle

This review shows that relatively simple changes to diet and lifestyle can significantly,and rapidly,reduce the risks associated with coronavirus disease 2019(COVID-19)in terms of infection risk,severity of disease,and even disease-related mortality.A wide range of interventions including regular exercise,adequate sleep,plant-based diets,maintenance of healthy weight,dietary supplementation,and time in nature have each been shown to have beneficial effects for supporting more positive health outcomes with COVID-19,in addition to promoting better overall *** paper brings together literature from these areas and presents the argument that non-pharmaceutical approaches should not be overlooked in our response to *** is noted that,in several cases,interventions discussed result in risk reductions equivalent to,or even greater than,those associated with currently available *** the balance of evidence suggests benefits,and the risk is minimal to none,it is suggested that communicating the power of individual actions to the public becomes morally ***,many lives could be saved,and many harms from the vaccine mandates avoided,if we were willing to embrace this lifestyle-centred approach in our efforts to deal with COVID-19.
