This study assessed the quality parameters and consumer perception and behavior of butter processed with different ratios of cow and sheep milk *** milk cream butter(CMCB)showed lower moisture content and higher fat content,smaller fat globules,and higher concentrations of long-chain saturated fatty acids and monounsaturated fatty acids,resulting in improved potential health *** addition,CMCB showed yellow color,little salt,and butter taste as the main sensory characteristics,resulting in grateful and good digestibility *** milk cream addition improved the rheological properties of the products(lower viscosity,consistency index,and rigidity(J0),and higher fluidity and viscoelastic behavior(J1)),mainly due to the increased concentrations of short-chain and medium-chain saturated fatty acids and polyunsaturated fatty ***,sheep milk cream butter(SMCB)was characterized by a greasy and striking aroma,with a lower perception of good digestibility and good appearance,and a higher bitter *** association of cow and sheep milk cream could be an exciting option to obtain products with improved fatty acid profiles,health indices,and yet maintain desired rheological and sensory *** formulation with 60%sheep milk cream and 40%cow milk cream would be the suggested combination.

Barley stripe mosaic virus(BSMV) North Dakota 18(ND18),Norwich(NW),Beijing (BJ),Xinjiang(XJ),Type and CV21 strains elicit different phenotypic responses on Brachypodium distachyon, a cereal grass model for ***18 is unable to infect the Brachypodium Bd3-1 inbred line,which contains a recently mapped resistance gene designated Bsr1(Cui et *** ONe,2012, 7(6);e38333).Along with ND18,the BJ,XJ,Type and CV21 strains also failed to produce visible symptoms or virus on Bd3-1,so it is possible that Bsr1 mediates resistance against all five BSMV strains. In contrast,NW overcomes Bd3-1 resistance and is accompanied by an intense mosaic phenotype and large amounts of *** of ND18 and NW genomic RNAs,and a series of chimeric exchanges within RNAβdemonstrate that Bd3-1 phenotypes are mediated by a region within the helicase motif of *** of this region reveals that the ND18 and NW strains contain divergent amino acids at positions 390,392 and 404 within this region,and that the inability of ND18 and the BJ,XJ,Type and CV21 strains to elicit fully virulent BSMV infections on Bd3-1 correlates with a threonine(T) at position 392(T).The virulent NW strain contains K residues at both the 390 and 392 positions and a glutamate (e) residue at position *** substitutions to swap the ND18 and NW 404 residues failed to affect the respective disease ***,R390K and T392K single exchanges and a R390K/ T392K double mutant in ND18 RNA TGB1 resulted in virulent *** contrast,incorporation of single K390R or K392T mutations into the NW TGB1 protein background did not affect the susceptible disease phenotype,suggesting that the NW K and K residues compensate for each *** contrast, a K390R/K392T double mutant in the NW TGB1 background failed to develop fully systemic infections on Bd3-1,or accumulate detectable viral RNA,but prominent necrotic streaks radiating from the secondary leaf veins were *** experiments in toto

Metastases of uveal melanoma(UM)spread predominantly to the *** to low response rates to systemic therapies,liver-directed therapies(LDT)are commonly used for tumor *** impact of LDT on the response to systemic treatment is unknown.A total of 182 patients with metastatic UM treated with immune checkpoint blockade(ICB)were included in this *** were recruited from prospective skin cancer centers and the German national skin cancer registry(ADOReg)of the German Dermatologic Cooperative Oncology Group(DeCOG).Two cohorts were compared:patients with LDT(cohort A,n=78)versus those without LDT(cohort B,n=104).Data were analyzed for response to treatment,progression-free survival(PFS),and overall survival(OS).The median OS was significantly longer in cohort A than in cohort B(20.1 vs.13.8 months;P=0.0016)and a trend towards improved PFS was observed for cohort A(3.0 vs.2.5 months;P=0.054).The objective response rate to any ICB(16.7%vs.3.8%,P=0.0073)and combined ICB(14.1%vs.4.5%,P=0.017)was more favorable in cohort *** data suggest that the combination of LDT with ICB may be associated with a survival benefit and higher treatment response to ICB in patients with metastatic UM.

The emergence of SARS-CoV-2 variants of concern and repeated outbreaks of coronavirus epidemics in the past two decades emphasize the need for next-generation pan-coronaviral *** the multi-functional papain-like protease(PLpro)domain of the viral nsp3 holds ***,none of the known coronavirus PLpro inhibitors has been shown to be in vivo ***,we screened a structurally diverse library of 50,080 compounds for potential coronavirus PLpro inhibitors and identified a noncovalent lead inhibitor F0213 that has broad-spectrum anti-coronaviral activity,including against the Sarbecoviruses(SARSCoV-1 and SARS-CoV-2),Merbecovirus(MeRS-CoV),as well as the Alphacoronavirus(hCoV-229e and hCoVOC43).Importantly,F0213 confers protection in both SARS-CoV-2-infected hamsters and MeRS-CoV-infected human DPP4-knockin mice.F0213 possesses a dual therapeutic functionality that suppresses coronavirus replication via blocking viral polyprotein cleavage,as well as promoting antiviral immunity by antagonizing the PLpro deubiquitinase *** the significant difference of substrate recognition,mode of inhibition studies suggest that F0213 is a competitive inhibitor against SARS2-PLpro via binding with the 157K amino acid residue,whereas an allosteric inhibitor of MeRSPLpro interacting with its 271e *** proof-ofconcept findings demonstrated that PLpro is a valid target for the development of broad-spectrum anticoronavirus *** orally administered F0213 may serve as a promising lead compound for combating the ongoing COVID-19 pandemic and future coronavirus outbreaks.

The advantage of using plants in nanoparticles synthesis is that they are easily available, safe to handle and possess a broad variability of metabolites such as antioxidants, nucleotides and vitamins. The aim of this study was to investigate the effects of Green and Zimbro tea and also Green coconut water as a reducing and stabilizer agent in gold nanoparticle synthesis. The gold nanoparticles were characterized by UV-Vis absorption spectroscopy, X-ray diffraction (XRD), Dynamic light scattering (DLS) and Transmission electron microscopy (TeM) analysis. Their physical stability was determined using a UV-Vis spectrophotometer over several days during storage at room temperature. We observed that green chemical process to obtain gold nanoparticles did not require any external chemicals reagent for stabilization of nanoparticulate. Absorption measurements indicated that the plasmon resonance wavelength appears around 530 nm. X-ray diffracto-grams of gold nanoparticles evidenced the presence of Au-rich (fcc) phases. TeM analysis showed a homogeneous dispersion of nanoparticles and some agglomerates. Differences in size and shape of the nanoparticles were observed. Zeta potential of AuNPs synthetized in presence of Green tea was -33 mV indicating stability of the synthesized nanoparticles.

目的考察不同型号大孔树脂纯化的刘寄奴总黄酮对IFN-γ和LPS协同诱导RAW264.7细胞中NO表达的影响。方法应用溶剂萃取和AB-8、D101、ADS-17大孔树脂分别制备刘寄奴总黄酮,分光光度法测定总黄酮的含量;IFN-γ和LPS协同诱导RAW264.7造成炎症模型;Griessreaction测定总黄酮对细胞上清液中亚硝酸盐含量的影响;RT-PCR考察总黄酮对细胞内诱导型一氧化氮合酶(iNOS)mRNA的影响;Westernblot考察总黄酮对细胞iNOS蛋白表达的影响。结果3种刘寄奴总黄酮含量均达到50%以上,且以ADS-17型大孔树脂富集所得的总黄酮含量最高,达到67.78%。3种刘寄奴总黄酮均呈剂量和时间依赖性抑制细胞上清液中亚硝酸盐的含量,同等流程中以ADS-17型大孔树脂富集的总黄酮达到半抑制率的剂量为最小。ADS-17型刘寄奴总黄酮呈剂量依赖性抑制iNOSmRNA和蛋白的表达。结论ADS-17型大孔树脂有效富集刘寄奴黄酮类化合物,适用于刘寄奴总黄酮的提取。ADS-17型刘寄奴总黄酮呈时间和剂量依赖性抑制细胞上清液中亚硝酸盐含量,部分通过抑制iNOSmRNA和蛋白的表达达到下调NO产量的结果。刘寄奴总黄酮可能是刘寄奴抗炎药效的物质基础,有望进一步开发成为iNOS抑制剂。

Water quality of Mexican tropical lake Chapala was assessed through multivariate statistical techniques, cluster analysis (CA) and principal component analysis (PCA) at ten different monitoring sites for ten physicochemical variables and six metals. This study evaluated and interpreted complex water quality data sets and apportioned of pollution sources to get better information about water quality. From descriptive statistics results, the highest concentrations of metals occurred during the dry season, and this trend was explained by the fact that an unusual rainy event occurred during the month of February 2009 and brought metals into the lake by runoffs from nearby mountains. According to international criteria for water consumption by aquatic organisms [USePA], only Zn concentration values were below these criteria whereas the values of Ni, Pb, Cd and Fe were above the corresponding values set in these criteria (Ni: 52 μg·L-1, Pb: 2.5 μg·L-1, Cd: 0.25 μg·L-1, and Fe: 1000 μg·L-1). The correlations were observed by PCA, which were used to classify the samples by CA, based on the PCA scores. Seven significant cluster groups of sampling locations—(sites 4 and 5), (sites 3 and 9), (site 7), (site 10), (sites 2 and 6), (site 8) and (site 1)— were detected on the basis of similarity of their water quality. The results revealed that the stress exerted on the lake caused by waste sources follows the order: domestic > agricultural > industrial.
