Interplay between post-translational cyclooxygenase-2 modifications and the metabolic and proteomic profile in a colorectal cancer cohort

BACKGROUND Colorectal cancer(CRC) is the second most common cause of cancer death worldwide. It is broadly described that cyclooxygenase-2(COX-2) is mainly overexpressed in CRC but less is known regarding post-translational modifications of this enzyme that may regulate its activity, intracellular localization and stability. since metabolic and proteomic profile analysis is essential for cancer prognosis and diagnosis, our hypothesis is that the analysis of correlations between these specific parameters and COX-2 state in tumors of a high number of CRC patients could be useful for the understanding of the basis of this cancer in humans.AIM To analyze COX-2 regulation in colorectal cancer and to perform a detailed analysis of their metabolic and proteomic profile.METHODs Biopsies from both healthy and pathological colorectal tissues were taken under informed consent from patients during standard colonoscopy procedure in the University Hospital of Bellvitge(Barcelona, spain) and Germans Trias i Pujol University Hospital(Campus Can Ruti)(Barcelona, spain). Western blot analysis was used to determine COX-2 levels. Deglycosylation assays were performed in both cells and tumor samples incubating each sample with peptide N-glycosidase F(PNGase F). Prostaglandin E2(PGE2) levels were determined using a specific ELIsA. 1 H high resolution magic angle spinning(HRMAs) analysis was performed using a Bruker AvIII 500 MHz spectrometer and proteomic analysis was performed in a nano-liquid chromatography-tandem mass spectrometer(nano LC-Ms/Ms) using a QExactive HF orbitrap Ms.REsULTs Our data show that COX-2 has a differential expression profile in tumor tissue of CRC patients vs the adjacent non-tumor area, which correspond to a glycosylated and less active state of the protein. This fact was associated to a lesser PGE2 production in tumors. These results were corroborated in vitro performing deglycosylation assays in HT29 cell line where COX-2 protein profile was modified after PNGase F incubation, showing higher PGE2 levels. M

Flat Belt separation system under slat in Fattening Pig Housing: Effect of Belt slope Angle

In this study, the separation and drying efficiency of the excreta, separated into solid and liquid fraction by a flat conveyor belt under a partial slatted floor in a facility for fattening pigs, was determined. Two transverse slope angles (4° and 6°) for the flat belts were used, in a trial with pigs from 20.61 to 117.83 kg of live weight (LW), and the sample size was 7 pigs per pen, 42 pigs per room. The flat belt was more efficient with 6° than with 4° slope angle (32.65% vs. 29.91% dry matter content of solid fraction during the whole fattening period, respectively;P ° than for the 4° (1.4137 vs. 1.3030, during the whole fattening period;P °, this efficiency was not improved in finishing period -83.40 to 117.83 kg LW (two extractions per day) with regard to the growth period -20.61 to 83.40 kg LW (one extraction per day). Meanwhile, with 4° it did improved. The ventilation system underneath slat seems to play a key role in the obtained results.

In this paper,the lanthanoid complexes of N_(5) and N_(6) macrocycle ligands,without pendant arms or additional heteroatoms,are surveyed.This review covers the period from 2015 to the current date,since in 2014 schiff base macrocyclic ligands incorporating the pyridine moiety,and their complexes,were revised,and in 2015 the chemistry of pentaaza macrocycle ligands with rare earth metals was also summarized.Porphyrin and phthalocyanine ligands are not included in this review,which primarily focuses on complexes with schiff bases and amines without pendant arms or any additional donor other than nitrogen.The synthetic methods,structural characterization,based on single X-ray crystal data,and properties of the lanthanoid complexes,with special attention to magneto-structural correlations,are presented herein.

Myelin histology:a key tool in nervous system research

The myelin sheath is a lipoprotein-rich,multilayered structure capable of increasing conduction velocity in central and peripheral myelinated nerve fibers.Due to the complex structure and composition of myelin,various histological techniques have been developed over the centuries to evaluate myelin under normal,pathological or experimental conditions.Today,methods to assess myelin integrity or content are key tools in both clinical diagnosis and neuroscience research.In this review,we provide an updated summary of the composition and structure of the myelin sheath and discuss some histological procedures,from tissue fixation and processing techniques to the most used and practical myelin histological staining methods.Considering the lipoprotein nature of myelin,the main features and technical details of the different available methods that can be used to evaluate the lipid or protein components of myelin are described,as well as the precise ultrastructural techniques.

Hypothalamic circuits and aging:keeping the circadian clock updated

Over the past century,age-related diseases,such as cancer,type-2 diabetes,obesity,and mental illness,have shown a significant increase,negatively impacting overall quality of life.studies on aged animal models have unveiled a progressive discoordination at multiple regulatory levels,including transcriptional,translational,and post-translational processes,resulting from cellular stress and circadian derangements.The circadian clock emerges as a key regulator,sustaining physiological homeostasis and promoting healthy aging through timely molecular coordination of pivotal cellular processes,such as stem-cell function,cellular stress responses,and inter-tissue communication,which become disrupted during aging.Given the crucial role of hypothalamic circuits in regulating organismal physiology,metabolic control,sleep homeostasis,and circadian rhythms,and their dependence on these processes,strategies aimed at enhancing hypothalamic and circadian function,including pharmacological and non-pharmacological approaches,offer systemic benefits for healthy aging.Intranasal brain-directed drug administration represents a promising avenue for effectively targeting specific brain regions,like the hypothalamus,while reducing side effects associated with systemic drug delivery,thereby presenting new therapeutic possibilities for diverse age-related conditions.

Highly graphitized carbon nanosheets with embedded Ni nanocrystals as anode for Li-ion batteries

A C/Ni composite was prepared via thermal decomposition of a nickel oleate complex at 700℃,yielding disperse Ni nanocrystals with an average size of 20 nm,encapsulated by carbon nanosheets as deduced from transmission electron microscopy(TEM)images and confirmed from X-ray photoelectron spectroscopy(XPs).Furthermore,the X-ray diffraction pattern revealed a good ordering of the carbon layers,forced by the Ni encapsulation to adopt a bending structure.Considering the close interaction between the graphitized framework and the metallic nanoparticles we have studied the properties of the composite as an anode for Li-ion batteries.Compared with other nanostructured synthetic carbons,this carbon composite has a low voltage hysteresis and a modest irreversible capacity value,properties that play a significant role in its behaviour as electrodes in full cell configuration.At moderate rate values,0.25 C,the electrode delivers an average capacity value around 723 mAh·g^−1 on cycling,among the highest values so far reported for this carbon type.At higher rate values,1 C,the average capacity values delivered by the cell on cycling decrease,around 205 mAh·g^−1,but it maintains good capacity retention,a coulombic efficiency close to 100%after the first cycles and recovery of the capacity values when the rate is restored from 3 to 0.1 C.

Interactivity software tools for teaching in ophthalmology

The use of interactive audience software,such as audience response systems(ARs),in medical education has become increasingly popular in recent years.This technology allows instructors to engage students in real time,encouraging active participation and promoting effective learning.The benefits of interactive audience software in medical education include increased student engagement,promotion of active learning,and enhanced learning outcomes.However,there are also several challenges to its implementation,including technical difficulties,careful planning and preparation,over-reliance on technology,and ethical concerns related to privacy and data security.The cost of implementing interactive audience software may also be a barrier for some institutions.This paper specifically reviews six interactive software platforms,including socrative,Quizizz,Pear Deck,slido,Wooclap and ClassPoint.These platforms allow for real-time assessment of student understanding,feedback,and participation.They also enable instructors to adjust their teaching strategies based on student responses and feedback.Overall,interactive audience software has shown great potential to enhance learning and engagement in medical education.It is important for instructors to carefully consider the benefits and challenges of its implementation.While the cost of implementing interactive audience software may be a barrier for some institutions,there are free and low-cost options available.

soil resilience assessment using soil profile descriptions and Analytic Hierarchy Process in Mediterranean mountains considering diverse fire occurrences

Wildfires are complex natural phenomena that exert significant impacts on landscapes, societies, and economies. Understanding the concept of resilience is crucial in mitigating its possible negative impacts, as it involves preparing for, responding to, and recovering from wildfires. This research aims to demonstrate the utility of in situ soil profile description in assessing land use resilience using an Analytic Hierarchy Process(AHP) through an expert panel survey. The study examines a catchment located in the Balearic Islands, considering two fire occurrences(once and twice), comparing abandoned agricultural terraces and natural hillslopes. The results demonstrated that the priority ranking of variables to assess soil profile resilience against wildfires, determined by a panel of 10 experts, identified horizon depth(25.1%), slope inclination(21.5%), and hydrological connectivity(16.6%) as the most crucial factors. Other variables, such as number and size of roots, structure of pedal soil material, size class structure, and rock fragments, also contributed to resilience but to a lesser extent, with scores ranging from 5.7% to 9.6%. Analyzing the priorities established by the experts using AHP, the results showed that the least resilient soil horizon was H1 of the control hillslope, especially under high and low connectivity processes, which aligned with the loss of superficial soil horizons after one and two wildfires. Hillslopes showed greater changes in resilience after occurring wildfires compared to terraces, with the most significant alterations occurring after the second wildfire event. This study addresses a significant knowledge gap in the field by highlighting the interconnectedness of wildfires, resilience, and land use, providing insights into land management strategies for wildfire-prone regions.

Identification of Rhizobia Isolated from Nodules of Mexican Commercial soybean varieties

Rhizobia, crucial for nitrogen fixation in leguminous plants, play a vital role in soybean cultivation. This study, conducted in Mexico, a major soybean importer, aimed to identify bacteria from nodules of five soybean varieties in high-production regions. Multilocus sequence analysis (MLsA) was employed for enhanced species resolution. The study identified six Bradyrhizobium species: Bradyrhizobium japonicum UsDA 110, Bradyrhizobium japonicum UsDA 6, Bradyrhizobium elkanii UsDA 76, Bradyrhizobium neotropicale, Bradyrhizobium lablabi, and Bradyrhizobium icense. Bradyrhizobium japonicum UsDA 110 predominated in the soils, displaying symbiotic preference for the Huasteca 400 variety. However, phylogenetic analysis didn't reveal a clear association between strains, soil, and soybean variety. This research sheds light on the diversity of rhizobia in Mexican soybean cultivation, contributing to the understanding of symbiotic relationships in soybean production systems.

Liquid biopsy for gastric cancer:Techniques,applications,and future directions

After the study of circulating tumor cells in blood through liquid biopsy(LB),this technique has evolved to encompass the analysis of multiple materials originating from the tumor,such as nucleic acids,extracellular vesicles,tumor-educated platelets,and other metabolites.Additionally,research has extended to include the examination of samples other than blood or plasma,such as saliva,gastric juice,urine,or stool.LB techniques are diverse,intricate,and variable.They must be highly sensitive,and pre-analytical,patient,and tumor-related factors significantly influence the detection threshold,diagnostic method selection,and potential results.Consequently,the implementation of LB in clinical practice still faces several challenges.The potential applications of LB range from early cancer detection to guiding targeted therapy or immunotherapy in both early and advanced cancer cases,monitoring treatment response,early identification of relapses,or assessing patient risk.On the other hand,gastric cancer(GC)is a disease often diagnosed at advanced stages.Despite recent advances in molecular understanding,the currently available treatment options have not substantially improved the prognosis for many of these patients.The application of LB in GC could be highly valuable as a non-invasive method for early diagnosis and for enhancing the management and outcomes of these patients.In this comprehensive review,from a pathologist’s perspective,we provide an overview of the main options available in LB,delve into the fundamental principles of the most studied techniques,explore the potential utility of LB application in the context of GC,and address the obstacles that need to be overcome in the future to make this innovative technique a game-changer in cancer diagnosis and treatment within clinical practice.
