Ischemic preconditioning inhibits development of edematous cerulein-induced pancreatitis: Involvement of cyclooxygenases and heat shock protein 70

AIM： To determine whether ischemic preconditioning （IP） affects the development of edematous cerulein-induced pancreatitis and to assess the role of cyclooxygenase-1 （COX-l）, COX-2, and heat shock protein 70 （HSP 70） in this process. METHODS： In male wistar rats, IP was performed by damping of celiac artery （twice for 5 min at 5-min intervals）. Thirty minutes after IP or sham operation, acute pancreatitis was induced by cerulein. Activity of COX-1 or COX-2 was inhibited by resverabol or rofecoxib, respectively （10 rng/kg）. RESULTS： IP significantly reduced pancreatic damage in cerulein-induced pancreatitis as demonstrated by the improvement of pancreas histology, reduction in serum lipase and poly-C ribonuclease activity, and serum concentration of pro-inflammatory interleukin （IL）-1β. Also, IP attenuated the pancreatitis-evoked fall in pancreatic blood flow and pancreatic DNA synthesis. Serum level of anti-inflammatory IL-10 was not affected by IP. Cerulein-induced pancreatitis and IP increased the content of HSP 70 in the pancreas. Maximal increase in HSP 70 was observed when IP was combined with cerulein-induced pancreatitis. Inhibition of COXs, especially COX-2, reduced the protective effect of IP in edematous pancreatitis. CONCLUSION： Our results indicate that IP reducespancreatic damage in cerulein-induced pancreatitis and this effect, at least in part, depends on the activity of COXs and pancreatic production of HSP 70.

Effect of pegylated interferon alpha 2b plus ribavirin treatment on plasma transforming growth factor-β1,metalloproteinase-1,and tissue metalloproteinase inhibitor-1 in patients with chronic hepatitis C

AIM： To evaluate the effect of antiviral treatment on plasma levels of transforming growth factor-β1 （TGF-β1）, metalloproteinase 1 （MMP-1）, and tissue inhibitor of metaUoproteinase-1 （TIMP-1） in patients with chronic hepatitis C. METHODS： TGF-β1, MMP-1, and TIMP-1 plasma concentrations were measured by an enzyme immunoassay in 28 patients, during 48 wk of treatment with pegylated interferon-alpha 2b （PEG-IFN-α2b） plus ribavirin （RBV） and after 24 wk of follow-up. Patients were divided into two groups： responders （R） and non-responders （NR） related to achieved sustained virologic response. Normal values were evaluated in plasma samples of 13 healthy volunteers. RESULTS： Baseline plasma concentrations of TGF-β1 and TIMP-1 （30.9±3.7 and 1 506±61 ng/mL respectively） measured in all subjects significantly exceeded the normal values （TGF-β1： 18.3±1.6 ng/mL and TIMP-1： 1 102±67 ng/mL）. In contrast, pretreatment MMP-1 mean level （6.5±0.9 ng/mL） was significantly lower than normal values （11.9±0.9 ng/mL）. Response to the treatment was observed in 12 patients （43%）. TGF-β1 mean concentration measured during the treatment phase decreased to the control level in both groups. However at wk 72, values of NR patients increased and became significantly higher than in R group. TIMP-1 concentrations in R group decreased during the treatment to the level similar to normal. In NR group, TIMP-1 remained significantly elevated during treatment and follow-up phase and significant difference between both groups was demonstrated at wk 48 and 72. MMP-1 levels were significantly decreased in both groups at baseline. Treatment caused rise of its concentration only in the R group, whereas values in NR group remained on the level similar to baseline. Statistically significant difference between groups was noted at wk 48 and 72. CONCLUSION： These findings support the usefulness of TGF-β1, TIMP-1, and MMP-1 in the management of chronic hepatitis C. Low symptomatic COVID-19 in an elderly patient with follicular lymphoma treated with rituximab-based immunotherapy:A case report

BACKGROUND Follicular lymphoma is an indolent lymphoma that may progress to a highly aggressive form requiring *** regimens utilize rituximab,an anti-CD20 monoclonal antibody,which may affect the clinical course of novel coronavirus,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infections[coronavirus disease 2019(COVID-19)].Here we describe the first case of mild COVID-19 during ongoing oncological treatment without significant deterioration after rituximab *** SUMMARY A 74-year-old female with an enlargement of her right palatine tonsil was diagnosed with follicular lymphoma following tonsillectomy and started immunochemotherapy according to the rituximab,cyclophosphamide,vincristine,prednisone *** home before the fourth cycle,she developed nonspecific symptoms(excessive fatigue,loss of appetite and nausea),misdiagnosed as adverse effects of ***,interim positron emission tomography-computed tomography scan,performed shortly before rituximab administration,revealed previously nonexistent pulmonary changes,potentially of infectious ***-CoV-2 infection was confirmed by a nasopharyngeal swab(with reverse transcriptase polymerase chain reaction test)performed the following *** rituximab infusion,the patient remained oligosymptomatic and was discharged home for *** reached a negative SARS-CoV-2 status before the subsequently scheduled regimen,the patient successfully received six cycles of rituximab,cyclophosphamide,vincristine,prednisone and obtained complete remission by positron emission tomographycomputed *** Our case shows that rituximab-based immunotherapy due to follicular lymphoma may have no evident negative effect on the COVID-19 clinical course.

Structure and properties of diffusive titanium nitride layers produced by hybrid method on AZ91D magnesium alloy

A diffusive titanium nitride layers were produced on AZ91 D magnesium alloy by a hybrid method which combines the titanium coating physical vapor deposition（PVD） with subsequent nitriding. The layers are composed of a thin surface zone of titanium nitrides and oxides, an intermediate titanium zone and Ti-Al-Mg type diffusive zone. Since the layers improve the corrosion resistance, the best corrosion resistance is obtained when the layers are produced on aluminum base sub-layer by oxynitriding.

BACKGROUND The revolution in treatment of patients with chronic hepatitis C virus(HCV)infection dates back to the introduction of direct-acting antivirals(DAAs).The increase in efficacy was most pronounced in patients infected with genotype(GT)1b,as this was the most poorly responsive population to treatment during the interferon *** To identify the most effective interferon-free therapy for GT1b-infected patients and to determine positive and negative predictors of virological *** This real-world retrospective analysis included patients chronically infected with GT1b HCV whose data were obtained from the multicenter observational EpiTer-2 *** effectiveness was evaluated for each therapeutic regimen as the percentage of sustained virological responses(SVR).Assessment of the safety was based on the evaluation of the course of therapy,the occurrence of adverse events including serious ones,deaths during treatment and in the post 12-wk follow-up *** The studied population consisted of 11385 patients with a mean age of 53±14.8 years and a female predominance(53.4%).The majority of them were treatment-naïve(74.6%)and patients with cirrhosis accounted for 24.3%.Of the DAA regimens used,76.9%were GT-specific with ombitasvir/paritaprevir/ritonavir+dasabuvir±ribavirin being the most used option(32.4%).A total of 10903 patients responded to treatment resulting in a 98.1%in the per-protocol analysis after excluding 273 patients without SVR *** effectiveness of all regimens exceeded 90%and the highest SVR of 98.9%was achieved in patients treated with a combination of glecaprevir/*** regression analyses showed that the virologic response was independently associated with female sex[odds ratio(OR)=1.67],absence of decompensated cirrhosis at baseline(OR=2.42)and higher baseline platelets(OR=1.004 per 1000/μL increase),while the presence of human immunodeficiency virus(HIV)coinfection significantly decreased the odds

本工作研究了在有石墨和碳化钨粉末催化剂存在的情况下,用氢加压还原醋酸盐缓冲溶液中的水合镍(Ⅱ)时,温度、原液的初始pH值、氢分压以及混合速率对还原过程的影响。镍的还原与镍浓度的关系遵循一级反应动力学。还原速度与氢分压的平方根成正比,而混合速率在400—700min^(-1)的范围内,还原速度与混合速率无关,用石墨粉末催化剂或用碳化钨粉末催化剂时,反应过程的表观活化能(△Ea)分别为77.6kJ/mol和80.1kJ/mol。当采用石墨粉末催化剂时,观察到了反应过程有一个非常长的诱导期。

Alternative therapies are necessary for the treatment of malaria due to emerging drug ***,many promising antimalarial compounds have poor water solubility and suffer from the lack of suitable delivery systems,which seriously limits their *** address this problem,we synthesized a series of azacarbazoles that were evaluated for antimalarial activity against D10(chloroquine-sensitive)and w2(chloroquine-resistant)strains of *** most active compound,9H-3-azacarbazole(3),was encapsulated in a novel o/w nanoemulsion consisting of ethyl esters of polyunsaturated fatty acids n-3 and n-6 obtained from flax oil as the oil phase,Smix(Tween 80 and Transcutol HP)and *** formulation was further analyzed using transmission electron microscopy,dynamic light scattering and in vitro and in vivo *** was shown that droplets of the 3-loaded nanosystem were spherical,with satisfactory stability,without cytotoxicity towards fibroblasts and intestinal cell lines at concentrations corresponding to twice the IC50 for ***,the nanoemulsion with this type of oil phase was internalized by Caco-2 ***,pharmacokinetics demonstrated rapid absorption of compound 3(tmax=5.0 min)after intragastric administration of 3-encapsulated nanoemulsion at a dose of 0.02 mg/kg in mice,with penetration of compound 3 to deep *** 3-encapsulated nanoemulsion was found to be 2.8 and 4.2 times more effective in inhibiting the D10 and w2 strains of the parasite,respectively,compared to non-encapsulated *** findings support a role for novel o/w nanoemulsions as delivery vehicles for antimalarial drugs.
