Burns are a significant cause of trauma,and over the years,the focus of patient care has shifted from just survival to facilitation of improved functional ***,burn treatment,especially in the case of extensive burn injuries,involves surgical excision of injured skin and reconstruction of the burn injury with the aid of skin *** skin substitutes do not contain all skin cell types and do not facilitate recapitulation of native skin ***-dimensional(3D)bioprinting for reconstruction of burn injuries involves layer-by-layer deposition of cells along with scaffolding materials over the injured *** bioprinting can be done either in situ or in *** these approaches are similar except for the site of printing and tissue *** are technological and regulatory challenges that need to be overcome for clinical translation of bioprinted skin for burn ***,the use of bioprinting for skin reconstruction following burns is promising;bioprinting will enable accurate placement of cell types and precise and reproducible fabrication of constructs to replace the injured or damaged ***,3D bioprinting is a very transformative technology,and its use for wound reconstruction will lead to a paradigm shift in patient *** this review,we aim to introduce bioprinting,the different stages involved,in vitro and in vivo skin bioprinting,and the various clinical and regulatory challenges in adoption of this technology.

Pharmacological manipulation of transcription factor protein-protein interactions:opportunities and obstacles

Much research on transcription factor biology and their genetic pathways has been undertaken over the last30 years, especially in the field of developmental biology and cancer. Yet, very little is known about the molecularmodalities of highly dynamic interactions between transcription factors, genomic DNA, and protein *** breakthroughs such as RNA-seq (RNA-sequencing), ChIP-seq (chromatin immunoprecipitationsequencing), RIME (rapid immunoprecipitation mass spectrometry of endogenous proteins), and single-moleculeimaging will dramatically accelerate the discovery rate of their molecular mode of action in the next few *** a pharmacological viewpoint, conventional methods used to target transcription factor activity withmolecules mimicking endogenous ligands fail to achieve high specificity and are limited by a lack of identificationof new molecular targets. Protein-protein interactions are likely to represent one of the next major classes oftherapeutic targets. Transcription factors, known to act mostly via protein-protein interaction, may well be atthe forefront of this type of drug development. One hurdle in this field remains the difficulty to collate structuraldata into meaningful information for rational drug design. Another hurdle is the lack of chemical libraries meeting thestructural requirements of protein-protein interaction *** more attempts at modulating transcription factor activity are undertaken, valuable knowledge will be accumulatedon the modality of action required to modulate transcription and how these findings can be applied to developingtranscription factor drugs. Key discoveries will spawn into new therapeutic approaches not only as anticancer targetsbut also for other indications, such as those with an inflammatory component including neurodegenerative disorders,diabetes, and chronic liver and kidney diseases.

Trends in in ovo sexing technologies:insights and interpretation from papers and patents

Numerous researchers and institutions have been developing in ovo sexing technologies to improve animal welfare by identifying male embryos in an early embryonic stage and disposing of them before pain *** review gives a complete overview of the technological approaches reported in papers and patents by performing a thorough search using Web of Science and Patstat/Espacenet databases for papers and patents,*** on a total of 49 papers and 115 patent families reported until May 2023 worldwide,11 technology categories were defined:6 non-optical and 5 optical *** category was described for its characteristics while assessing its potential for ***,the dynamics of the publications of in ovo sexing techniques in both paper and patent fields were described through growth curves,and the interest or actual status was visualized using the number of paper citations and the actual legal status of the *** comparing the reported technologies in papers to those in patents,scientific gaps were observed,as some of the patented technologies were not reported in the scientific literature,e.g.,ion mobility and mass spectrometry ***,more diverse approaches in all categories were found in patents,although they do require more scientific evidence through papers or industrial adoption to prove their ***,although there is a recent trend for non-invasive techniques,invasive methods like analyzing DNA through PCR or hormones through immunosensing are still being reported(and might continue to be)in papers and *** was also observed that none of the technologies complies with all the industry requirements,although 5 companies already entered the *** the one hand,more research and harmony between consumers,industry,and governments is *** the other hand,close monitoring of the market performance of the currently available techniques will offer valuable insights into the potential an

Accuracy of sedimentgraph modeling from topography map scale and DEM mesh size

The evaluation of scale effects on modeling performance of sedimentgraphs as the ultimate outputs of the hydrological simulation is vital for adaptive watershed *** present study therefore analyzed effectability of simulated sedimentgraphs components in association with different topographic maps with various vector *** whole procedure was materialized to select the critical scale and cell size for the Galazchai Watershed,*** this end,the stormwise sedimentgraphs were modeled for 23 recorded events using the Clark's Instantaneous Unit Hydrograph (IUH) model stemmed for developing Instantaneous Unit Sedimentgraphs (IUSGs) incorporated with dimensionless sediment concentTation distribution (DSCD) based on the vector scales of 1∶25000,1∶50000,1∶100000 and 1∶250000 and cell sizes of 5,10,20 30,50,100 and 200 *** 644 direct sedimentgraphs (DSGs) were then evaluated based on Relative Errors (REs) for sediment volume,peak sediment,time to peak,base time and the Coefficient of Efficiency (CE).The results confirmed that REs for peak sediment,time to peak and CE were sensitive to cell *** results further verified that the cell sizes of 5,20,30,50 and 100 m were critical cell sizes in viewpoint of time to *** addition,the vector scales of 1∶50000 with cell size of 50 m,and 1∶100000 with cell sizes of 5 and 10 m were critical vector scales and cell sizes based on RMSE evaluation *** is concluded from the current research that the accuracy of simulation of sedimentgraph was influenced by map scales and mesh sizes.

Association between heat stress and oxidative stress in poultry;mitochondrial dysfunction and dietary interventions with phytochemicals

Heat as a stressor of poultry has been studied extensively for many decades; it affects poultry production on a worldwide basis and has significant impact on well-being and production. More recently, the involvement of heat stress in inducing oxidative stress has received much interest. Oxidative stress is defined as the presence of reactive species in excess of the available antioxidant capacity of animal cells. Reactive species can modify several biologically cellular macromolecules and can interfere with cell signaling pathways. Furthermore, during the last decade, there has been an ever-increasing interest in the use of a wide array of natural feed-delivered phytochemicals that have potential antioxidant properties for poultry. In light of this, the current review aims to（1） summarize the mechanisms through which heat stress triggers excessive superoxide radical production in the mitochondrion and progresses into oxidative stress,（2） illustrate that this pathophysiology is dependent on the intensity and duration of heat stress,（3） present different nutritional strategies for mitigation of mitochondrial dysfunction, with particular focus on antioxidant *** stress that occurs with heat exposure can be manifest in all parts of the body; however, mitochondrial dysfunction underlies oxidative stress. In the initial phase of acute heat stress, mitochondrial substrate oxidation and electron transport chain activity are increased resulting in excessive superoxide production. During the later stage of acute heat stress, down-regulation of avian uncoupling protein worsens the oxidative stress situation causing mitochondrial dysfunction and tissue damage. Typically, antioxidant enzyme activities are upregulated. Chronic heat stress, however, leads to downsizing of mitochondrial metabolic oxidative capacity, up-regulation of avian uncoupling protein, a clear alteration in the pattern of antioxidant enzyme activities, and depletion of antioxidant reserve

Changes of the glutathione redox system during the weaning transition in piglets, in relation to small intestinal morphology and barrier function

Background: Weaning is known to result in barrier dysfunction and villus atrophy in the immediate post-weaning phase, and the magnitude of these responses is hypothesized to correlate with changes in the glutathione(GSH)redox system. Therefore, these parameters were simultaneously measured throughout the weaning phase, in piglets differing in birth weight category and weaning age, as these pre-weaning factors are important determinants for the weaning transition. Low birth weight(LBW) and normal birth weight(NBW) littermates were assigned to one of three weaning treatments;i.e. weaning at 3 weeks of age(3 w), weaning at 4 weeks of age(4 w) and removal from the sow at 3 d of age and fed a milk replacer until weaning at 3 weeks of age(3 d3 w). For each of these treatments, six LBW and six NBW piglets were euthanized at 0, 2, 5, 12 or 28 d post-weaning piglets, adding up 180 ***: Weaning increased the glutathione peroxidase activity on d 5 post-weaning in plasma, and duodenal and jejunal mucosa. Small intestinal glutathione-S-transferase activity gradually increased until d 12 post-weaning, and this was combined with a progressive rise of mucosal GSH up till d 12 post-weaning. Oxidation of the GSH redox status(GSH/GSSG Eh) was only observed in the small intestinal mucosa of 3 d3 w weaned piglets at d 5 postweaning. These piglets also demonstrated increased fluorescein isothiocyanate dextran(FD4) and horseradish peroxidase fluxes in the duodenum and distal jejunum during the experiment, and specifically demonstrated increased FD4 fluxes at d 2 to d 5 post-weaning. On the other hand, profound villus atrophy was observed during the weaning transition for all weaning treatments. The efficacy and effectiveness of machine learning for weaning in mechanically ventilated patients at the intensive care unit: a systematic review

Weani ng from mechanical ventilation in the in tensive care unit (ICU) is a complex clinical problem and relevant for future organ engineering. Prolonged mechanical ventilation (MV) leads to a range of medical complications that increases length of stay and costs as well as contributes to morbidity and even mortality and long-term quality of life. The need to reduce MV is both clinical and economical. Artificial intelligence or machine learning (ML) methods are promising opportunities to positively influence patient outcomes. ML methods have been proposed to enhance clinical decisions processes by using the large amount of digital information generated in the ICU setting. There is a particular interest in empirical methods (such as ML) to improve management of "difficult-to-wean" patients, due to the associated costs and adverse events associated with this population. A systematic literature search was performed using the OVID, IEEEXplore, PubMed, and Web of Science databases. All publications that included (1) the application of ML to weaning from MV in the ICU and (2) a clinical outcome measurement were reviewed. A checklist to assess the study quality of medical ML publications was modified to suit the critical assessment of ML in MV weaning literature. The systematic search identified nine studies that used ML for weaning management from MV in critical care. The weaning management application areas included (1) prediction of successful spontaneous breathing trials (SBTs),(2) prediction of successful extubation,(3) prediction of arterial blood gases, and (4) ventilator setting and oxygenation-adjustment advisory systems. Seven of the nine studies scored seven out of eight on the quality index. The remaining two of the nine studies scored one out of eight on the quality index.
