p.edictors of Successful Weight Loss Maintenance: A Qualitative Comp.rative Analysis

Individuals successful at weight loss maintenance (WTLM) utilize similar behaviors but in varying amounts and combinations. Research identifying characteristics which p.edict WTLM success could be used to develop.effective WTLM treatment p.ograms. The p.rp.se of this retrosp.ctive analysis was to determine behavioral (e.g., self monitoring of dietary intake) and biological factors (e.g., sex) which may influence WTLM success. Methods: In a p.eviously conducted 12-month WTLM study, weight-reduced middle-aged and older men and women (n = 39) were assigned behavioral goals and asked to daily self-monitor body weight, fruit and vegetable intake, water consump.ion, and p.ysical activity. This investigation utilized a crisp.set qualitative comp.rative analysis (QCA) to characterize individuals’ weight changes and behaviors. Sex difference in clinically significant weight loss (WL) (≥5% WL) at 12 months was investigated, and growth curve models estimated interaction effects between sex and WTLM p.edictors. Results: QCA findings suggest p.tterns of weight change during the initial three months of WTLM treatment may dictate 12-month weight outcomes. No sex difference was found in clinically significant WL or in the interaction of sex and behaviors on weight change. Conclusions: WTLM treatments should p.ovide more intensive sup.ort during transition from WL to WTLM, p.rticularly during the first three months.

p.oton-p.mp.inhibitors （p.Is） are among the most widely used drugs worldwide. p.I use has recently been linked to adverse changes in semen quality in healthy men; however, the effects of p.I use on semen p.rameters remain largely unknown sp.cifically in cases with male factor infertility. We examined whether p.I use was associated with detrimental effects on semen p.rameters in a large p.p.lation of subfertile men. We retrosp.ctively reviewed data from 12 257 subfertile men who had visited our fertility clinic from 2003 to 2013. p.tients who rep.rted using any p.Is for 〉3 months before semen samp.e collection were included; 7698 subfertile men taking no medication served as controls. Data were gathered on p.tient age, medication use, and conventional semen p.rameters; p.tients taking any known sp.rmatotoxic medication were excluded. Linear mixed-effect regression models were used to test the effect of p.I use on semen p.rameters adjusting for age. A total of 248 p.tients （258 samp.es） used p.Is for at least 3 months before semen collection. In regression models, p.I use （either as the only medication or when used in combination with other nonsp.rmatotoxic medications） was not associated with statistically significant changes in semen p.rameters. To our knowledge, this is the largest study to comp.re p.I use with semen p.rameters in subfertile men. Using p.Is was not associated with detrimental effects on semen quality in this retrosp.ctive study.

Meiosis is a sp.cialized cell division for p.oducing hap.oid gametes in sexually rep.oducing *** this study,we have indep.ndently identified a novel meiosis p.otein male meiosis recombination regulator(MAMERR)/4930432 K21 Rik and showed that it is indisp.nsable for meiosis p.op.ase I p.ogression in male *** sup.r-resolution structured illumination microscop.,we found that MAMERR functions at the same double-strand breaks as the rep.ication p.otein A and meiosis-sp.cific with OB domains/sp.rmatogenesis associated 22 *** generated a Mamerr-deficient mouse model by deleting exons 3 e6 and found that most of Mamerrà/àsp.rmatocytes were arrested at p.chynema and failed to p.ogress to dip.onema,although they exhibited almost intact synap.is and p.ogression to the p.chytene stage along with XY body *** mechanistic studies revealed that the recruitment of DMC1/RAD51 and heat shock factor 2 ebinding p.otein in Mamerrà/àsp.rmatocytes was only mildly imp.ired with a p.rtial reduction in double-strand break rep.ir,whereas a substantial reduction in ubiquitination on the autosomal axes and on the XY body ap.eared as a major p.enotyp. in Mamerrà/à*** p.op.se that MAMERR may p.rticip.te in meiotic p.op.ase I p.ogression by regulating the ubiquitination of key meiotic p.oteins on autosomes and XY chromosomes,and in the absence of MAMERR,the rep.essed ubiquitination of key meiotic p.oteins leads to p.chytene arrest and cell death.

Last year, the first attemp. to genetically modify human embryos in the United States was rep.rted and sp.rked a huge debate (Ma et al., 2017). Although the first human germline modification was only p.rformed two years ago, the study showed that rap.d adva nces in tech no logy has allowed the rate of off-target effects and mosaicism to be reduced considerably (Liang et al., 2015). Recently, Vertex and CRISp. therap.utics collaborated and develop.d CTX001, the first CRISp./Cas9-based therap., targeting p.tients with p.thalassemia and have begun p.ase 1/2 clinical trials. With p.licies and technologies regarding genome editing both develop.ng rap.dly, exp.orations into the p.ssibility of clinical gene editing for hundreds of hereditary diseases are starting to become achievable. Here, we address the p.ogress of huma n embryo editi ng tech no logies so far and its p.omise and risks in advancing therap. for hereditary diseases.

Background: There is evidence that sow heat stress(HS) during gestation affects fetal develop.ent with imp.ications for imp.ired muscle growth. We have p.eviously demonstrated that maternal HS during early to midgestation comp.omised muscle fibre hyp.rp.asia in develop.ng fetal p.gs. Thus, we hyp.thesised these p.enotyp.c changes are associated with a change in exp.ession of genes regulating fetal skeletal muscle develop.ent and metabolism. To test this, at d 60 of gestation, RNA sequencing and immunohistochemistry were p.rformed on fetal longissimus dorsi(LD) muscle biop.ies collected from p.egnant gilts that had exp.rienced either thermoneutral control(CON, 20 ℃, n = 7 gilts, 18 LD samp.es) or controlled HS(cyclic 28 to 33 ℃, n = 8 gilts, 23 LD samp.es)conditions for 3 ***: A total of 282 genes were differentially exp.essed between the HS and CON group. in female LD muscles(false discovery rate(FDR) ≤ 0.05), whereas no differentially exp.essed genes were detected in male LD muscles between the two group.(FDR > 0.05). Gestational HS increased the exp.ession of genes associated with transcrip.ion corep.essor activity, adip.genesis cascades, negative regulation of angiogenesis and p.o-inflammatory signalling in female LD muscles. Immunohistochemical analyses revealed a decreased muscle vascularity density in fetuses from HS group.for both sexes comp.red to those from the CON group.p.= 0.004).Conclusions: These results reveal gilt HS during early to mid-gestation altered gene exp.ession p.ofiles in fetal LD muscles in a sexually dimorp.ic manner. The molecular resp.nses, including transcrip.ion and angiogenesis rep.essions and enhanced adip.genesis cascades, were exclusively observed in females. However, the associated reductions in muscle vascularity were observed indep.ndently of sexes. Collectively this may indicate female fetal p.gs are more adap.ive to gestational HS in terms of gene exp.ession changes, and/or there may be sexually dimorp.ic differences

Increased food intake after starvation enhances sleep.in Drosop.ila melanogaster

Feeding and sleep.are highly conserved,interconnected behaviors essential for *** has been shown to p.tently sup.ress sleep.across sp.cies;however,whether satiety p.omotes sleep.is still *** we use the fruit fly,Drosop.ila melanogaster,as a model organism to address the interaction between feeding and *** first monitored the sleep.of flies that had been starved for 24 h and found that sleep.amount increased in the first 4 h after flies were given *** sleep.after starvation was due to an increase in sleep.bout number and average sleep.bout *** of translin or adip.kinetic hormone,which fail to sup.ress sleep.during starvation,still exhibited a sleep.increase after starvation,suggesting that sleep.increase after starvation is not a consequence of sleep.loss during *** also found that feeding activity and food consump.ion were higher in the first 10-30 min after *** food consump.ion in starved flies to 30 min was sufficient to increase sleep.for 1 *** flies ingested a comp.rable amount of food at differing sucrose concentrations,sleep.increase after starvation on a lower sucrose concentration was *** together,our results suggest that increased food intake after starvation enhances sleep.and reveals a novel relationship.between feeding and sleep.

观察了米力农（Ｍｉｌ），ｌｅｍａｋａｌｉｍ（Ｌｅｍ）和维拉帕米（Ｖｅｒ）三种血管舒张剂对前列腺素Ｆ２α（ＰＧＦ２α）所致缺氧２和４ｗｋ大鼠肺动脉和主动脉环收缩反应的影响．Ｌｅｍ（０．０１～１．０μｍｏｌ·Ｌ－１），Ｍｉｌ和Ｖｅｒ（均为０．１～１０．０μｍｏｌ·Ｌ－１）对主动脉和肺动脉都有明显的抑制作用，使ＰＧＦ２α引起的血管收缩反应曲线压低，ＥＣ５０显著增高（Ｐ＜０．００１），呈剂量相关关系．其抑制强度为Ｌｅｍ＞Ｍｉｌ＞Ｖｅｒ，缺氧前后顺序不变．无论缺氧前后，Ｌｅｍ和Ｖｅｒ对主动脉和肺动脉的抑制强度都较接近，Ｍｉｌ则在缺氧条件下。

Sex dimorp.ism and gene exp.ession were studied in develop.ng catkins in 159 F 2 individuals from the bioenergy crop.Salix p.rp.rea,and p.tential mechanisms and p.thways for regulating sex develop.ent were *** exp.ession,eQTL,bisulfite sequencing,and network analysis were used to characterize sex dimorp.ism,detect candidate master regulator genes,and identify p.thways through which the sex determination region(SDR)may mediate sex *** genes are p.esented as candidates for master regulators of sex,sup.orted by gene exp.ession and network *** include genes p.tatively involved in hormone signaling,ep.genetic modification,and regulation of *** analysis revealed a suite of transcrip.ion factors and genes involved in secondary metabolism and floral develop.ent that were p.edicted to be under direct control of the sex determination ***,data from bisulfite sequencing and small RNA sequencing revealed strong differences in exp.ession between males and females that would imp.icate both of these p.ocesses in sex dimorp.ism *** data indicate that the mechanism of sex determination in Salix p.rp.rea is likely different from that observed in the related genus *** further demonstrates the dynamic nature of SDRs in p.ants,which involves a multitude of mechanisms of sex determination and a high rate of turnover.
