Background: There is evidence that sow heat stress(Hs) during gestation affects fetal development with implications for impaired muscle growth. We have previously demonstrated that maternal Hs during early to midgestation compromised muscle fibre hyperplasia in developing fetal pigs. Thus, we hypothesised these phenotypic changes are associated with a change in expression of genes regulating fetal skeletal muscle development and metabolism. To test this, at d 60 of gestation, RNA sequencing and immunohistochemistry were performed on fetal longissimus dorsi(LD) muscle biopsies collected from pregnant gilts that had experienced either thermoneutral control(CON, 20 ℃, n = 7 gilts, 18 LD samples) or controlled Hs(cyclic 28 to 33 ℃, n = 8 gilts, 23 LD samples)conditions for 3 ***: A total of 282 genes were differentially expressed between the Hs and CON groups in female LD muscles(false discovery rate(FDR) ≤ 0.05), whereas no differentially expressed genes were detected in male LD muscles between the two groups(FDR > 0.05). Gestational Hs increased the expression of genes associated with transcription corepressor activity, adipogenesis cascades, negative regulation of angiogenesis and pro-inflammatory signalling in female LD muscles. Immunohistochemical analyses revealed a decreased muscle vascularity density in fetuses from Hs group for both sexes compared to those from the CON group(P = 0.004).Conclusions: These results reveal gilt Hs during early to mid-gestation altered gene expression profiles in fetal LD muscles in a sexually dimorphic manner. The molecular responses, including transcription and angiogenesis repressions and enhanced adipogenesis cascades, were exclusively observed in females. However, the associated reductions in muscle vascularity were observed independently of sexes. Collectively this may indicate female fetal pigs are more adaptive to gestational Hs in terms of gene expression changes, and/or there may be sexually dimorphic differences

The purpose of this study was to determine whether the training responses observed with low-load resistance exercise to volitional fatigue translates into significant muscle hypertrophy, and compare that response to high-load resistance training. Nine previously untrained men (aged 25 [sD 3] years at the beginning of the study, standing height 1.73 [sD 0.07] m, body mass 68.9 [sD 8.1] kg) completed 6-week of high load-resistance training (HL-RT) (75% of one repeti-tion maximal [1RM], 3-sets, 3x/wk) followed by 12 months of detraining. Following this, subjects completed 6 weeks of low load-resistance training (LL-RT) to volitional fatigue (30% 1 RM, 4 sets, 3x/wk). Increases (p 0.05) in magnetic resonance imaging-measured triceps brachii and pectorals major muscle cross-sectional areas were similar for both HL-RT (11.9% and 17.6%, respectively) and LL-RT (9.8% and 21.1%, respectively). In addition, both groups increased (p 0.05) 1RM and maximal elbow extension strength following training;however, the percent increases in 1RM (8.6% vs. 21.0%) and elbow extension strength (6.5% vs. 13.9%) were significantly (p 0.05) lower with LL-RT. Both protocols elicited similar increases in muscle cross-sectional area, however differences were observed in strength. An explanation of the smaller relative increases in strength may be due to the fact that detraining after HL-RT did not cause strength values to return to baseline levels thereby producing smaller changes in strength. In addition, the results may also suggest that the consistent practice of lifting a heavy load is necessary to maximize gains in muscular strength of the trained movement. These results demonstrate that significant muscle hypertrophy can occur without high-load resistance training and suggests that the focus on percentage of external load as the important deciding factor on muscle hypertrophy is too simplistic and inappropriate.

A new graduate course has been developed at University of Bridgeport to establish entrepreneurship opportunities for today s challenging world. some advanced technologies, including Failure Modes and Effects Analysis (FMEA), standard Operation Procedure (sOP), and Lean Manufacturing, have been introduced in helping students to gain high level of engineering experiences. It also helps students to examine political, social and economic issues that associated to engineering project delivery processes that can lead to save product process expenses, increase manufacturing efficiency, improve product quality and shorten production period. Although this course was taught at mechanical/manufacturing engineering majors, it also can be applied to most areas of Us industrial and engineering practices.

Pseudomonas putida KT217 was grown on a complex medium comprised of co-products of the ethanol and biodiesel industries to assess the organism's capability to produce medium-chain-length polyhydroxyalkanoate (mcl-PHA). The growth phase was carried out in a medium containing 400 g/L condensed corn solubles (CCs), supplemented with ammonium hydroxide as a nitrogen source. Following the exponential phase, co-products of the biodiesel industry (soapstock and glycerin) were fed into the reactor to trigger PHA production. When glycerin was added to the bioreactor (75 g/L total addition), the final cell dry weight (CDW) and PHA content were 30 g/L and 31%, respectively. The monomeric composition in the PHA formed was relatively uniform throughout incubation with 3-hydroxydecanoate dominating. When a total of 153 g/L of sunflower soapstock was added to the bioreactor in a fed-batch manner, the final CDW and PHA content were 17 g/L and 17%, respectively. Following addition of soapstock the monomeric composition of the polymer changed dramatically, with the 3-hydroxyoctanoate monomer becoming dominant and greater unsaturation present in the PHA.

Aim： To study the effects of homocysteine and copper on type 5 phosphodiesterase （PDE5） expression in cavemosai vascular smooth muscle cells （CVsMCs） and to investigate superoxide （O2-） derived from nicotinamide adenine dinucleotide phosphate oxidase as homocysteine and copper generate O2-, and O2- upregulates PDE5 expression. Methods： CVsMCs derived from rabbit penis were incubated with homocysteine or copper chloride with or without superoxide dismutase （sOD）, catalase, sildenafil citrate, or apocynin （nicotinamide adenine dinucleotide phosphate inhibitor） for 16 h. The expression of PDE5 and of glyceraldehyde-3-phosphate dehydrogenase （internal standard） was assessed using Western blot analysis. In parallel, O2-was measured spectrophotometrically. Results： CuCl2 alone （up to 10 μmol/L） and homocysteine alone （up to 100 μmol/L） had no effect on O2- formation in CVsMCs compared to controls. In combination, however, homocysteine and CuCl2 markedly increased O2- formation, an effect blocked by sOD, catalase, apocynin, and sildenafil （1 μmol/L） when co-incubated over the same time course. PDE5 expression was also significantly increased in CVsMCs incubated with homocysteine and CuCl2, compared to controls. This effect was also negated by 16-h co-incubation with sOD, catalase, apocynin and sildenafil. Conclusion： This represents a novel pathogenic mechanism underlying ED, and indicates that the therapeutic actions of prolonged sildenafil use are mediated in part through inhibition of this pathway.
