Utilization of RnA sequencing to investigate olfactory dysfunction in chronic rhinosinusitis without nasal polyps:A pilot study

Objectives:Prior research on olfactory dysfunction in chronic rhinosinusitis(CRS)has focused on patients with polyps and suggests that direct inflammation of the olfactory cleft mucosa plays a contributory *** purpose of this study was to evaluate gene expression in superior turbinate mucosal specimens,comparing normosmic and dysosmic CRS patients without polyps(CRSsnP).Methods:Tissue samples were obtained from the superior turbinates of patients with CRSsnP at the time of endoscopic sinus *** subsequently underwent RnA sequencing and functional analysis to investigate biological pathways associated with differentially expressed genes between dysosmic(n=7)and normosmic(n=4)***:Differential gene expression analysis comparing dysosmic and normosmic CRSsnP patients showed upregulation of 563 genes and downregulation of 327 *** stringent criteria for multiple comparisons,one upregulated gene(Immediate Early Response 3[IER3])had an false discovery rate(FDR)correction adjustedP value considered statistically significant(Pnge 2.69).Reactome functional analysis revealed eight biological pathways significantly different between dysosmic and normosmic patients(Pn)including IL-4 and IL-13 signaling,IL-10 signaling,and rhodopsin-like ***:RnA sequencing of the superior turbinates in patients with CRSsnP can provide valuable information regarding biological pathways and genes involved in olfactory *** study supports literature suggesting that Type 2 inflammation may play a role in olfactory dysfunction in at least some patients with *** study also prompts questions regarding the role of IL-10,rhodopsin-like receptors,andIER3 in the pathogenesis of olfactory dysfunction.

We aim to evaluate the incidence of incontinence following laser endoscopic enucleation of the prostate(EEP)comparing en-bloc(Group 1)versus 2-lobe/3-lobe techniques(Group 2).We performed a retrospective review of patients undergoing EEP for benign prostaticenlargement in 12 centers between January 2020 and January *** were presented as median and interquartile range(IQR).Univariable and multivariable logistic regression analysis was performed to evaluate factors associated with stress urinary incontinence(SUI)and mixed urinary incontinence(MUI).There were 1711 patients in Group 1 and 3357 patients in Group *** in Group 2were significantly younger(68[62–73]years vs 69[63–74]years,P=0.002).Median(interquartile range)prostate volume(PV)wassimilar between the groups(70[52–92]ml in Group 1 vs 70[54–90]ml in Group 2,P=0.774).There was no difference in preoperativeInternational Prostate Symptom Score,quality of life,or maximum flow ***,morcellation,and total surgical time weresignificantly shorter in Group *** 1 month,overall incontinence rate was 6.3%in Group 1 versus 5.3%in Group 2(P=0.12),and urge incontinence was significantly higher in Group 1(55.1%vs 37.3%in Group 2,Pnths,the overall rate ofincontinence was 1.7%in Group 1 versus 2.3%in Group 2(P=0.06),and SUI was significantly higher in Group 2(55.6%vs 24.1%in Group 1,P=0.002).At multivariable analysis,PV and IPSS were factors significantly associated with higher odds of transient SUI/***,surgical time,and no early apical release technique were factors associated with higher odds of persistent SUI/MUI.

Objective:To scrutinize the definitions of minimal invasive surgical therapy(MIST)and to investigate urologists’knowledge,attitudes,and practices for benign prostatic obstruction ***:A 36-item survey was developed with a Delphi *** on definitions of MIST and attitudes and practices of benign prostatic obstruction surgeries were *** were invited globally to complete the online *** was achieved when more than or equal to 70%responses were“agree or strongly agree”and less than or equal to 15%responses were“disagree or strongly disagree”(consensus agree),or when more than or equal to 70%responses were“disagree or strongly disagree”and less than or equal to 15%responses were“agree or strongly agree”(consensus disagree).Results:The top three qualities for defining MIST were minimal blood loss(n=466,80.3%),fast post-operative recovery(n=431,74.3%),and short hospital stay(n=425,73.3%).The top three surgeries that were regarded as MIST were Urolift®(n=361,62.2%),Rezum®(n=351,60.5%),and endoscopic enucleation of the prostate(EEP)(n=332,57.2%).Consensus in the knowledge section was achieved for the superiority of Urolift®,Rezum®,and iTInD®over transurethral resection of the prostate with regard to blood loss,recovery,day surgery feasibility,and post-operative *** in the attitudes section was achieved for the superiority of Urolift®,Rezum®,and iTInD®over transurethral resection of the prostate with regard to blood loss,recovery,and day surgery *** on both sections was achieved for EEP as the option with the better symptoms and flow improvement,lower retreatment rate,and better suitable for prostate more than 80 ***:Minimal blood loss,fast post-operative recovery,and short hospital stay were the most important qualities for defining ***®,Rezum®,and EEP were regarded as MIST by most urologists.

Extreme surges and rainfall represent major driving factors for compound flooding in estuary regions along the Chinese *** combined effect of extreme surges and rainfall(that is,compound floods)might lead to greater impacts than if the drivers occurred in ***,understanding the frequency and severity of compound flooding is important for improving flood hazard assessment and compound flood resilience in coastal *** this study,we examined the dependence between extreme surges and corresponding rainfall events in 26 catchments along the Chinese coastline during typhoon and non-typhoon seasons using copula functions,to identify where the two drivers more often occur together and the implication for flood management in these *** found that the interaction between flood drivers is statistically significant in 10catchments located around Hainan Island(south)and Shanghai,where surge peaks occur mainly during the typhoon season and around the Bohai Sea(north),where surge peaks occur mainly during the non-typhoon *** further applied the copula-based framework to model the dependence between surge peaks and associated rainfall and estimate their joint and conditional probability in two specific locations—Hainan Island and the Bohai Sea,where the correlation between flood drivers is statistically *** observed that in Hainan Island where most of the surge peaks occur during the typhoon season,extreme rainfall events during the typhoon season are generally more intense compared to annual maxima *** contrast,around the Bohai Sea where surge peaks occur mainly outside the typhoon season,rainfall is less intense than annual maxima *** results show that the interaction between extreme surges and rainfall can provide valuable insight when designing coastal and urban infrastructure,especially in highly populated urban areas prone to both coastal and pluvial flooding,such as many Chinese coastal cities.

Mitral valve(MV)tissue engineering is still in its early stage,and one major challenge in MV tissue engineering is to identify appropriate scaffold *** the potential of acellular MV scaffolds being demonstrated recently,it is important to have a full understanding of the biomechanics of the native MV components and their acellular *** this study,we have successfully characterized the structural and mechanical properties of porcine MV components,including anterior leaflet(AL),posterior leaflet(PL),strut chordae,and basal chordae,before and after *** DnA assay showed more than 90%reduction in DnA content,and Griffonia simplicifolia(GS)lectin immunohistochemistry confirmed the complete lack of porcine𝛼-Gal antigen in the acellular MV *** the acellular AL and PL,the atrialis,spongiosa,and fibrosa trilayered structure,along with its ECM constitutes,i.e.,collagen fibers,elastin fibers,and portion of GAGs,were ***-theless,the ECM of both AL and PL experienced a certain degree of disruption,exhibiting a less dense,porous ECM *** overall anatomical morphology of the strut and basal chordae were also maintained af-ter decellularization,with longitudinal morphology experiencing minimum disruption,but the cross-sectional morphology exhibiting evenly-distributed porous *** the acellular AL and PL,the nonlinear anisotropic biaxial mechanical behavior was overall preserved;however,uniaxial tensile tests showed that the removal of cellular content and the disruption of structural ECM did result in small decreases in maximum tensile modulus,tissue extensibility,failure stress,and failure strain for both MV leaflets and chordae.

The historical information of loadings on substation helps in evaluation of size of photovoltaic(PV)generation and energy storages for peak shaving and distribution system upgrade deferral. A method, based on consumption data, is proposed to separate the unusual consumption and to form the clusters of similar regular consumption. The method does optimal partition of the load pattern data into core points and border points, high and less dense regions, respectively. The local outlier factor, which does not require fixed probability distribution of data and statistical measures, ranks the unusual consumptions on only the border points, which are a few percent of the complete data. The suggested method finds the optimal or close to optimal number of clusters of similar shape of load patterns to detect regular peak and valley load demands on different days. Furthermore,identification and characterization of features pertaining to unusual consumptions in load pattern data have been done on border points only. The effectiveness of the proposed method and characterization is tested on two practical distribution systems.

Optical haze of transparent and conductive silver nanowire films

Contemporary nanostructured transparent electrodes for use in solar cells require high transmittance and high conductivity, dictating nanostructures with high aspect ratios. Optical haze is an equally important yet unstudied parameter in transparent electrodes for solar cells that is also determined by the geometry of the nanostructures that compose the electrode. In this work, the effect of the silver nanowire diameter on the optical haze values in the visible spectrum was investigated using films composed of wires with either small diameters （n60 nm） or large diameters （~150 nm）. Finite difference time domain （FDTD） simulations and experimental transmittance data confirm that smaller diameter nanowires form higher performing transparent conducting electrode （TCE） films according to the current figure of merit. While maintaining near constant transmittance and conductivity for each film, however, it was observed experimentally that films composed of silver nanowires with larger diameters have a higher haze factor than films with smaller diameters. This confirms the FDTD simulations of the haze factor for single nanowires with similarly large and small diameters. This is the first record of haze properties for Ag nWs that have been simulated or experimentally measured, and also the first evidence that the current figure of merit for TCEs is insufficient to evaluate their performance in solar cell devices.
