Accumulation of poly(adenosine diphosphate-ribose)by sustained supply of calcium inducing mitochondrial stress in pancreatic cancer cells

BACKGROUND The biochemical phenomenon defined as poly adenosine diphosphate(ADP)-ribosylation(PARylation)is essential for the progression of pancreatic ***,the excessive accumulation of poly ADP-ribose(PAR)induces apoptosis-inducing factor(AIF)release from mitochondria and energy deprivation resulting in the caspase-independent death of cancer *** To investigate whether sustained calcium supply could induce an anticancer effect on pancreatic cancer by PAR *** Two pancreatic cancer cell lines,AsPC-1 and CFPAC-1 were used for the *** influx and mitochondrial reactive oxygen species(ROS)were observed by fluorescence *** in enzyme levels,as well as PAR accumulation and energy metabolism,were measured using assay ***-dependent cell death was investigated followed by confirming in vivo anticancer effects by sustained calcium *** Mitochondrial ROS levels were elevated with increasing calcium influx into pancreatic cancer ***,excess PAR accumulation,decreased PAR glycohydrolase and ADP-ribosyl hydrolase 3 levels,and energy deprivation were *** vitro and in vivo antitumor effects were confirmed to accompany elevated AIF *** This study visualized the potential anticancer effects of excessive PAR accumulation by sustained calcium supply on pancreatic cancer,however elucidating a clear mode of action remains a challenge,and it should be accompanied by further studies to assess its potential for clinical application.

An Experimental Study of Condensation Heat Transfer in Sub-Millimeter Rectangular Tubes

The characteristics of local heat transfer and pressure drops were experimentally investigated using condensing R134a two-phase flow, in single rectangular tubes, with hydraulic diameter of 0.494, 0.658, and 0.972 mm. New experimental techniques were used to measure the in-tube condensation heat transfer coefficient especially for the low heat and mass flows. Tests were performed for a mass flux of 100, 200, 400, and 600 kg/m2s, a heat flux of 5 to 20 kW/m2, and a saturation temperature of 40℃. In this study, effect of heat flux, mass flux, vapor qualities, and hydraulic diameter on flow condensation were investigated and the experimental local condensation heat transfer coefficients and frictional pressure drop are shown. The experimental data of condensation Nusselt number are compared with previous correlations, most of which are proposed for the condensation of pure refrigerant in a relatively large inner diameter round tubes.

Long Short-Term Memory Recurrent Neural Network-Based Acoustic Model Using Connectionist Temporal Classification on a Large-Scale Training Corpus

A Long Short-Term Memory(LSTM) Recurrent Neural Network(RNN) has driven tremendous improvements on an acoustic model based on Gaussian Mixture Model(GMM). However, these models based on a hybrid method require a forced aligned Hidden Markov Model(HMM) state sequence obtained from the GMM-based acoustic model. Therefore, it requires a long computation time for training both the GMM-based acoustic model and a deep learning-based acoustic model. In order to solve this problem, an acoustic model using CTC algorithm is proposed. CTC algorithm does not require the GMM-based acoustic model because it does not use the forced aligned HMM state sequence. However, previous works on a LSTM RNN-based acoustic model using CTC used a small-scale training corpus. In this paper, the LSTM RNN-based acoustic model using CTC is trained on a large-scale training corpus and its performance is evaluated. The implemented acoustic model has a performance of 6.18% and 15.01% in terms of Word Error Rate(WER) for clean speech and noisy speech, respectively. This is similar to a performance of the acoustic model based on the hybrid method.

Spontaneous resolution of gallbladder hematoma in blunt traumatic injury:A case report

BACKGROUND We report a case of intragallbladder hematoma and biliary tract obstruction caused by blunt gallbladder *** report that the patient was safely treated by conservative treatment after the obstruction was resolved by endoscopic retrograde cholangiopancreatography(ERCP).CASE SUMMARY A 67-year-old man was admitted via the emergency department due to complaints of right-sided abdominal pain that started 2 d *** days prior to presentation,the patient had slipped,fallen and struck his abdomen on a motorcycle *** initial vital signs were *** physical examination,he showed right upper quadrant pain and Murphy’s sign,with decreased bowel ***,he had had a poor appetite for 4 *** had been on aspirin for 2 years due to underlying *** simple radiography revealed a slight *** laboratory findings were as follows:white blood cell count,15.5×103/µL(normal range 4.8×10^(3)–10.8×10^(3));hemoglobin,9.4 g/dL;aspartate aminotransferase/alanine transferase,423/348 U/L;total bilirubin/direct bilirubin,4.45/3.26 mg/dL;-GTP,639 U/L(normal range 5–61 U/L);and C-reactive protein,12.32 mg/dL(0–0.3).Abdominal computed tomography showed a distended gallbladder with edematous wall change and a 55 mm×40 mm *** was observed in both the intrahepatic and common bile duct *** treatment was initiated,and ERCP was performed,with hemobilia found during *** cannulation,the patient’s symptoms were relieved,and after conservative management,the patient was discharged with no further *** 1-month follow-up,the gallbladder hematoma was completely *** In the case of traumatic injury to the gallbladder,conservative treatment is feasible even in the presence of hematoma.

The electronic structure and intervalley coupling of artificial and genuine graphene superlattices

The so-called artificial graphene is an artificial material whose low-energy carriers are described by the massless Dirac equation. Applying a periodic potential with triangular symmetry to a two-dimensional electron gas is one approach to make such a material. According to recent experimental results, it is now possible to realize artificial graphene in the lab and to even apply an additional lateral, one-dimensional periodic potential to it. We name the latter system an artificial graphene superlattice in order to distinguish it from a genuine graphene superlaedce made from graphene. In this study, we investigate the electronic structure of artificial graphene superlattices, which exhibit the emergence of energy band gaps, merging and splitting of the Dirac points, etc. Then, from a similar investigation on genuine graphene superlattices, we show that many of these features originate from the coupling between Dirac fermions residing in two different valleys--the intervaUey coupling. Furthermore, contrary to previous studies, we find that the effects of intervalley coupling on the electronic structure cannot be ignored, irrespective of the length of the spatial period of the superlattice.

Superiority of split dose midazolam as conscious sedation for outpatient colonoscopy

AIM: To elucidate the efficacy and safety of a split dose of midazolam in combination with meperidine for colonoscopy. METHODS: Eighty subjects undergoing outpatient colonoscopy were randomly assigned to group A or B. Group A (n = 40) received a split dose of midazolam in combination with meperidine. Group B (n = 40) received a single dose of midazolam in combination with meperidine. Outcome measurements were level of sedation, duration of sedation and recovery, degree of pain and satisfaction, procedure-related memory, controllability, and adverse events. RESULTS: Group A had a lower frequency of significant hypoxemia (P = 0.043) and a higher sedation score on withdrawal of the endoscope from the descending colon than group B (P = 0.043). Group B recovered from sedation slightly sooner than group A (P < 0.002). Scores for pain and memory, except insertion-related memory, were lower in group A one week after colonoscopic examination (P = 0.018 and P < 0.030, respectively). Poor patient controllability was noted by the endoscopist and nurse in group B (P = 0.038 and P = 0.032, respectively). CONCLUSION: Split dose midazolam in combination with meperidine resulted in a safer, more equable sedation status during colonoscopic examination and a reduction in procedure-related pain and memory, but resulted in longer recovery time.

Construction of random sheared fosmid library from Chinese cabbage and its use for Brassica rapa genome sequencing project

As a part of the Multinational Genome Sequencing Project of Brassica rapa, linkage group R9 and R3 were sequenced using a bacterial artificial chromosome （BAC） by BAC strategy. The current physical contigs are expected to cover approximately 90% euchromatins of both chromosomes. As the project progresses, BAC selection for sequence extension becomes more limited because BAC libraries are restriction enzyme-specific. To support the project, a random sheared fosmid library was constructed. The library consists of 97536 clones with average insert size of approximately 40 kb corresponding to seven genome equivalents, assuming a Chinese cabbage genome size of 550 Mb. The library was screened with primers designed at the end of sequences of nine points of scaffold gaps where BAC clones cannot be selected to extend the physical contigs. The selected positive clones were end-sequenced to check the overlap between the fosmid clones and the adjacent BAC clones. Nine fosmid clones were selected and fully sequenced. The sequences revealed two completed gap filling and seven sequence extensions, which can be used for further selection of BAC clones confirming that the fosmid library will facilitate the sequence completion of B. rapa.

Strong interlayer coupling and stable topological flat bands in twisted bilayer photonic Moiré superlattices

The moiré superlattice of misaligned atomic bilayers paves the way for designing a new class of materials with wide *** this work,we propose a photonic analog of the moiré superlattice based on dielectric resonator *** sharp contrast to van der Waals materials with weak interlayer coupling,we realize the strong coupling regime in a moiré superlattice,characterized by cascades of robust flat bands at large ***,we find that these flat bands are characterized by a non-trivial band topology,the origin of which is the moiré pattern of the resonator *** physical manifestation of the flat band topology is a robust one-dimensional conducting channel on edge,protected by the reflection symmetry of the moiré *** explicitly breaking the underlying reflection symmetry on the boundary terminations,we show that the first-order topological edge modes naturally deform into higher-order topological corner *** work pioneers the physics of topological phases in the designable platform of photonic moiré superlattices beyond the weakly coupled regime.
