The high effectiveness of the third dose of bNT162b2 in healthy adolescents against Omicron bA.1 has been reported in some studies,but immune responses conferring this protection are not yet *** this analysis,our study(NCT04800133)aims to evaluate the humoral and cellular responses against wild-type and Omicron(bA.1,bA.2 and/or bA.5)SARS-CoV-2 before and after a third dose of bNT162b2 in healthy *** 5 months after 2 doses,S IgG,S IgG Fc receptor-binding,and neutralising antibody responses waned significantly,yet neutralising antibodies remained detectable in all tested adolescents and S IgG avidity increased from 1 month after 2 *** antibody responses and S-specific IFN-γ^(+)and IL-2^(+)CD8^(+)T cell responses were significantly boosted in healthy adolescents after a homologous third dose of *** to adults,humoral responses for the third dose were non-inferior or superior in *** S-specific IFN-γ^(+)and IL-2^(+)CD4^(+)and CD8^(+)T cell responses in adolescents and adults were comparable or ***,after 3 doses,adolescents had preserved S IgG,S IgG avidity,S IgG FcγRIIIa-binding,against Omicron bA.2,as well as preserved cellular responses against bA.1 S and moderate neutralisation levels against bA.1,bA.2 and *** from 100 and 96%of adolescents tested at 1 and 5 months after two doses could also neutralise *** study found high antibody and T cell responses,including potent cross-variant reactivity,after three doses of bNT162b2 vaccine in adolescents in its current formulation,suggesting that current vaccines can be protective against symptomatic Omicron disease.

background: Mother to child transmission (MTCT) of HIV constitutes a major source of new pediatric infections in Cameroon. Objective: The aim of this implementation research was to assess outcomes and effectiveness of providing life-long antiretroviral therapy (ART) for HIV-positive pregnant and breastfeeding women (Option b+). Methods: From October 2013 to July 2014, HIV-positive pregnant and breastfeeding women, not on antiretroviral (ARV) prophylaxis and ART, were recruited from 22 purposefully selected health facilities in the Northwest and Southwest regions for a prospective, observational cohort evaluation. Option b+ was offered to participants and outcome indicators were measured. Results: Out of 680 women eligible for this assessment, 669 (98%) were initiated on Option b+. Retention-in-care was 90% (95% CI, 87.85 - 92.61) and 79% (95% CI, 75.20 - 81.88), and loss to follow up (LTFU) was 7% (95% CI: 4.95 - 8.90) and 15% (95% CI: 12.06 - 17.56) at 6 and 12 months respectively. Maternal mortality at 12 months after ART initiation was 2% (13). As of March 2015, 538 HIV exposed infants (HEIs) were enrolled and received postpartum nevirapine prophylaxis within 72 hours of birth and 84% (454) were on cotrimoxazole at 6 to 8 weeks. by 8 weeks of age, 498 (93%) infants had HIV DNA PCR test with 486 (97.6%) negative and 12 (2.4%) positive. Conclusion: In Cameroon, successful implementation of Option b+ increased retention-in-care to 79% at one year for pregnant and breastfeeding women and reduced MTCT rate below 5% for HEIs at 8 weeks of age. Long term retention, maternal and infant mortality and final MTCT rate after cessation of breastfeeding require further evaluation.

目的探讨肺表面活性物质相关基因功能性变异在中国人群中的携带并与国外不同人群比较。方法以756例中国新生儿为研究对象(标本来自广西新生儿疾病筛查中心的干血片),提取基因组DNA,将每例DNA按等摩尔量混合形成混合DNA(pool-DNA),本研究共建立4个pool-DNA。以pool-DNA为模板,以表面活性蛋白b(SFTPb)、表面活性蛋白C(SFTPC)、ATP连接盒转运子A3(AbCA3)为目的基因,应用PCR方法扩增全外显子及侧翼端,并将产物再次混合,建立测序文库,应用Illumina第二代测序平台对目的基因进行全外显子深度测序,应用SIFT和Polyphen预测软件筛选出功能性变异,应用TaqMan方法对预测的功能性变异再次进行基因型验证,应用collapsed(合并等位基因)统计分析方法 ,获取每个基因上功能性变异的等位基因频率并应用卡方检验方法将中国人群携带的功能性变异的基因频率与美国外显子检测项目(NHLbI GO Exome Sequencing Project,ESP)公布的欧洲裔人群及非洲裔人群数据进行比较。结果 1、本研究在三个目的基因中共检测出7种杂合稀有功能性变异且均发现于AbCA3基因,共包含10个变异的等位基因,有3种变异出现2次;SFTPb和SFTPC基因未发现变异;2、每个变异的等位基因在该携带的个体中只出现一次,未发现单个研究对象携带两种或两个变异的等位基因;3、在7种变异中,有2种(E292V,L654V)是既往国外研究中曾发现的变异类型,其余5种是本研究中发现的新变异类型;4、合并后AbCA3基因变异的最小等位基因频率为0.00661,显著低于欧洲裔人群(0.0248)和非洲裔人群(0.0193),P均bCA3是主要发生变异的基因,与国外研究相似;4、第二代测序技术是发现稀有变异的有效手段
