Pancreatic cancer organotypics: High throughput, preclinical models for pharmacological agent evaluation

Pancreatic cancer carries a terrible prognosis,as the fourth most common cause of cancer death in the Western *** is clearly a need for new therapies to treat this *** of the reasons no effective treatment has been developed in the past decade may in part,be explained by the diverse influences exerted by the tumour *** tumour stroma cross-talk in pancreatic cancer can influence chemotherapy delivery and response ***,appropriate preclinical in vitro models which can bridge simple 2D in vitro cell based assays and complex in vivo models are required to understand the biology of pancreatic *** we discuss the evolution of 3D organotypic models,which recapitulare the morphological and functional features of pancreatic ductal adenocarcinoma(PDAC).Organotypic cultures are a valid high throughput preclinical in vitro model that maybe a useful tool to help establish new therapies for PDAC.A huge advantage of the organotypic model system is that any component of the model can be easily modulated in a short *** allows new therapies that can target the cancer,the stromal compartment or both to be tested in a model that mirrors the in vivo situation.A major challenge for the future is to expand the cellular composition of the organotypic model to further develop a system that mimics the PDAC environment more *** discuss how this challenge is being met to increase our understanding of this terrible disease and develop novel therapies that can improve the prognosis for patients.

Associations between cardiorespiratory fitness,physical activity, intraindividual variability in behavior, and cingulate cortex in younger adults

*** levels of cardiorespiratory fitness(CRF)and greater amounts of physical activity have been associated with lower intraindividual variability(IIV)in executive function in children and older *** the present study,we examined whether CRF,measured as maximal oxygen uptake(VO2max),and daily volume of moderate-to-vigorous intensity physical activity(MVPA)were associated with IIV of reaction time during performance of the incongruent condition of the Stroop task in younger ***,we examined whether the thickness of the cingulate cortex was associated with regulating variability in reaction time performance in the context of CRF or physical ***:CRF(measured as VO2max),accelerometry-measured MVPA,Stroop performance,and thickness of the rostral anterior cingulate cortex(rACC)derived from magnetic resonance imaging data were collected in 48 younger adults(age=24.58±4.95 years,mean±SD).Multiple regression was used to test associations between IIV during the Stroop task and CRF,MVPA,and rACC *** was tested using maximum likelihood estimation with ***:Consistent with our predictions,higher VO2max was associated with greater rACC thickness for the right hemisphere and greater daily amounts of MVPA were associated with greater rACC thickness for both the left and right *** thickness of the right rACC was associated with lower IIV for the incongruent condition of the Stroop *** and MVPA were not directly associated with ***,we did find that IIV and both CRF and MVPA were indirectly associated via the thickness of the right ***:These results indicate that higher CRF and greater daily volume of MVPA may be associated with lower IIV during the Stroop task via structural integrity of the *** controlled trials of MVPA would provide crucial information about the causal relations between these variables.

Classification of different sustainable flood retention basin types

Using a revised version of a previously published expert classification system, a database of potential Sustainable Flood Retention Basins has been developed for Scotland. The research shows that the majority of small and former （often old） drinking water reservoirs are kept full and their spillways are continuously in operation. Utilising some of the available capacity to contribute to flood control could significantly reduce the costs of complying with the European Union Flood Directive. Furthermore, the application of a previously developed classification model for Baden in Germany for the Scottish data set showed a lower diversity for basins in Scotland due to less developed infrastructure. The classification system appears to be robust and has the potential, with minor modifications, to be applied across Europe. The principle value of this approach is a clear and unambiguous categorisation, based on standard variables, which can help to promote communication and understanding between stakeholders.

Nuclear facilities have a regulatory requirement to measure radiation levels within Post Operational Clean Out(POCO)around nuclear facilities each year,resulting in a trend towards robotic deployments to gain an improved understanding during nuclear decommissioning *** UK Nuclear Decommissioning Authority supports the view that human-in-the-loop(HITL)robotic deployments are a solution to improve procedures and reduce risks within radiation characterisation of nuclear *** authors present a novel implementation of a Cyber-Physical System(CPS)deployed in an analogue nuclear environment,comprised of a multi-robot(MR)team coordinated by a HITL operator through a digital twin *** development of the CPS created efficient partnerships across systems including robots,digital systems and *** was presented as a multi-staged mission within an inspection scenario for the hetero-geneous Symbiotic Multi-Robot Fleet(SMuRF).Symbiotic interactions were achieved across the SMuRF where robots utilised automated collaborative governance to work together,where a single robot would face challenges in full characterisation of *** contributions include the demonstration of symbiotic autonomy and query-based learning of an autonomous mission supporting scalable autonomy and autonomy as a *** coordination of the CPS was a success and displayed further challenges and improvements related to future MR fleets.
