The.me.be. of Rho family of small GTPase. Cdc42 plays important and conse.ve. role. in ce.l polarity and motility. The.Cdc42e. family prote.ns have.be.n ide.tifie. to bind to Cdc42, ye. how the. inte.act with Cdc42 to re.ulate.ce.l migration re.ains to be.e.ucidate.. In this study, we.focus on Cdc42e.l, which is e.pre.se. pre.ominantly in the.highly migratory ne.ral cre.t ce.ls in frog e.bryos. Through morpholino-me.iate. knockdown, we.show that Cdc42e.l is re.uire. for the.migration of cranial ne.ral cre.t ce.ls. Loss of Cdc42e.l le.ds to rounde. ce.l shape. and the.formation of me.brane.ble.s, consiste.t with the.obse.ve. disruption in actin organization and focal adhe.ion alignme.t. As a re.ult, Cdc42e.1 is critical for ne.ral cre.t ce.ls to apply traction force. at the.corre.t place.to migrate.e.ficie.tly. We.furthe. show that Cdc42e.1 is localize. to two are.s in ne.ral cre.t ce.ts： in me.brane.protrusions toge.he. with Cdc42 and in pe.inucie.r patche. whe.e.Cdc42 is abse.t.
