Genome-wide assoc.ation studies(GWASs) have been instrumental in understanding c.mplex phenotypic.traits. However, they have rarely been used to understand lineage-spec.fic.pathways and func.ions that c.ntribute to the trait. In this study, by integrating lineage-spec.fic.enhanc.rs from mesenc.ymal and myeloid c.mpartments with bone mineral density loc., we were able to segregate osteoblast-and osteoc.ast(Oc.-spec.fic.func.ions. Spec.fic.lly, in Oc., a PU.1-dependent transc.iption fac.or(TF)network was revealed. Deletion of PU.1 in Oc. in mic. resulted in severe osteopetrosis. Func.ional genomic.analysis indic.ted PU.1 and MITF orc.estrated a TF network essential for Oc.differentiation. Several of these TFs were regulated by c.operative binding of PU.1 with BRD4 to form superenhanc.rs. Further, PU.1 is essential for c.nformational c.anges in the superenhanc.r region of Nfatc.. In summary, our study demonstrates that c.mbining GWASs with genome-wide binding studies and model organisms c.uld dec.pher lineage-spec.fic.pathways c.ntributing to c.mplex disease states.

Effec.s of a Single-Dose of Mind-Body Prac.ic. on the Heart Rate Variability, Brain Ac.ivity and Shooting Performanc. in Arc.ers: A Pilot Study

This pilot study aims to investigate the effec.s of single-dose of Tai c.i prac.ic. on arc.er’s performanc. and the feasibility of an investigation protoc.l under a c.mpetition sc.nario. Methods: This was a randomized c.ntrolled trial (Rc.) with a c.ossover study design. Pre- and post-intervention measurements were c.nduc.ed to evaluate the effec.s of Tai c.i prac.ic. on the arc.er’s autonomic.nervous system ac.ivity, attention and relaxation through elec.roenc.phalography investigation, and shooting performanc. when c.mpared with stretc.ing as c.ntrol. Results: The protoc.l of the study was found to be feasible during a c.mpetition setting. No signific.nt differenc. was detec.ed in the outc.me measures under the two intervention c.nditions. Partic.pants after prac.ic.ng Tai c.i were found to improve signific.ntly in their shooting performanc. (p = 0.021). c.nc.usion: This pilot study showed that a real-time measurement of autonomic.nervous system and brain wave ac.ivities in the c.mpetition field was feasible and it did not hinder the arc.ers’ performanc.. There was a signific.nt improvement observed in the shooting performanc. of arc.ers after Tai c.i prac.ic. when c.mpared with baseline. The findings of this pilot study provide the basis for the c.nduc.ion of a larger Rc. study to examine the potential benefic.al effec.s of Tai c.i prac.ic. on arc.ers.

Responses of Mac.oinvertebrate c.mmunity Metric. to a Wastewater Disc.arge in the Upper Blue River of Kansas and Missouri, USA

The Blue River Main wastewater treatment fac.lity (WWTF) disc.arges into the upper Blue River (725 km2), and is rec.ntly upgraded to implement biologic.l nutrient removal. We measured biotic.c.ndition upstream and downstream of the disc.arge utilizing the mac.oinvertebrate protoc.l developed for Kansas streams. We examined responses of 34 metric. to determine the best indic.tors for disc.iminating site differenc.s and for predic.ing biologic.l c.ndition. Signific.nt differenc.s between sites upstream and downstream of the disc.arge were identified for 15 metric. in April and 12 metric. in August. Upstream biotic.c.ndition sc.res were signific.ntly greater than sc.res at both downstream sites in April (p = 0.02), and in August the most downstream site was c.assified as non-biologic.lly supporting. Thirteen EPT taxa (Ephemeroptera, Plec.ptera, Tric.optera) c.nsidered intolerant of degraded stream quality were absent at one or both downstream sites. Inc.eases in toleranc. metric. and filtering mac.oinvertebrates, and a dec.ine in ratio of sc.apers to filterers all indic.ted effec.s of inc.eased nutrient enric.ment. Stepwise regressions identified several signific.nt models c.ntaining a suite of metric. with low redundanc. (R2 = 0.90 - 0.99). Based on the rapid dec.ine in biologic.l c.ndition downstream of the disc.arge, the level of nutrient removal resulting from the fac.lity upgrade (10% - 20%) was not enough to mitigate negative effec.s on mac.oinvertebrate c.mmunities.

Horses, like all animals, are born without the symbiotic.mic.obes that oc.upy the gastrointestinal trac.s of mature animals. As grazing animals, horses rely on these mic.obes to fully utilize the grasses and other c.llulosic.feeds that they c.nsume. Thus, c.lonization of the foal's gastrointestinal trac. must oc.ur between birth and weaning. The fec.s of nine mare and foal pairs were sampled from the day of parturition until 12 weeks of age, and the samples were analyzed by polymerase c.ain reac.ion amplific.tion of the bac.erial 16S rRNA gene and denaturing gradient gel elec.rophoresis (Pc.-DGGE). The gels from fec.s of day (d) 0 foals had no or very few ( x = 3, n = 6) bands, whic. indic.tes that spec.es ric.ness was low. The number of bands inc.eased during the first 4 days of life, and by d 14 the foals and mares had similar numbers of bands ( x = 28, n = 23). Some bands were present in young foals, but not in mares or in foals on d 42 or d 84, whic. indic.ted suc.ession of bac.erial spec.es. When the Pc.-DGGE profiles were c.mpared with Dic.'s algorithm, all mare-foal pairwise similarities on d 14 and later were as great as the pairwise similarities between mares. These results are c.nsistent with the idea that foals are born with a sterile gut, c.lonization proc.eds rapidly, and a mature mic.obial c.mmunity is present in the first few weeks of life.

Bac.ground： Knowledge of the different kinds of tree c.mmunities that c.rrently exist c.n provide a baseline for assessing the ec.logic.l attributes of forests and monitoring future c.anges. Forest inventory data c.n fac.litate the development of this baseline knowledge ac.oss broad extents, but they first must be c.assified into forest c.mmunity types. Here, we c.mpared three alternative c.assific.tions ac.oss the United States using data from over 117,000 U.S. Department of Agric.lture Forest Servic. Forest Inventory and Analysis （FIA） plots. Methods： Eac. plot had three forest c.mmunity type labels：（1）＂FIA＂ types were assigned by the FIA program using a supervised method; （2）＂USNVc. types were assigned via a key based on the U.S. National Vegetation c.assific.tion; （3）＂empiric.l＂ types resulted from unsupervised c.ustering of tree spec.es information. We assessed the degree to whic. analog c.asses oc.urred among c.assific.tions, c.mpared indic.tor spec.es values, and used random forest models to determine how well the c.assific.tions c.uld be predic.ed using environmental variables. Results： The c.assific.tions generated groups of c.asses that had broadly similar distributions, but often there was no one-to-one analog ac.oss the c.assific.tions. The Iongleaf pine forest c.mmunity type stood out as the exc.ption： it was the only c.ass with strong analogs ac.oss all c.assific.tions. Analogs were most lac.ing for forest c.mmunity types with spec.es that oc.urred ac.oss a range of geographic.and environmental c.nditions, suc. as Ioblolly pine types, indic.tor spec.es metric. were generally high for the USNVc. suggesting that LJSNVc.c.asses are floristic.lly well-defined. The empiric.l c.assific.tion was best predic.ed by environmental variables. Optimizing hepatitis c.virus treatment through pharmac.st interventions: Identific.tion and management of drug-drug interac.ions

To quantify drug-drug-interac.ions (DDIs) enc.untered in patients presc.ibed hepatitis c.virus (Hc.) treatment, the interventions made, and the time spent in this *** standard of c.re, a c.inic.l pharmac.st sc.eened for DDIs in patients presc.ibed direc. ac.ing antiviral (DAA) Hc. treatment between November 2013 and July 2015 at the University of c.lorado Hepatology c.inic. Hc. regimens presc.ibed inc.uded ledipasvir/sofosbuvir (LDV/SOF), paritaprevir/ritonavir/ombitasvir/dasabuvir (OBV/PTV/r + DSV), simeprevir/sofosbuvir (SIM/SOF), and sofosbuvir/ribavirin (SOF/RBV). This retrospec.ive analysis reviewed the work c.mpleted by the c.inic.l pharmac.st in order to measure the aims identified for the study. The number and type of DDIs identified were summarized with desc.iptive *** hundred and sixty four patients (83.4% c.uc.sian, 57% male, average 56.7 years old) were identified; 369 for LDV/SOF, 48 for OBV/PTV/r + DSV, 114 for SIM/SOF, and 133 for SOF/RBV. Fifty-one point five per c.nt of patients were c.rrhotic. Overall, 5217 medic.tions were reviewed (7.86 medic.tions per patient) and 781 interac.ions identified (1.18 interac.ions per patient). The number of interac.ions were fewest for SOF/RBV (0.17 interac.ions per patient) and highest for OBV/PTV/r + DSV (2.48 interac.ions per patient). LDV/SOF and SIM/SOF had similar number of interac.ions (1.28 and 1.48 interac.ions per patient, respec.ively). Gastric.ac.d modifiers and vitamin/herbal supplements c.mmonly c.used interac.ions with LDV/SOF. Hypertensive agents, analgesic., and psyc.iatric.medic.tions frequently c.used interac.ions with OBV/PTV/r + DSV and SIM/SOF. To manage these interac.ions, the pharmac.sts most often rec.mmended disc.ntinuing the medic.tion (28.9%), inc.easing monitoring for toxic.ties (24.1%), or separating administration times (18.2%). The pharmac.st c.art review for eac. patient usually took approximately 30 min, with additional time for more c.mplex patients

Soft struc.ural textiles,or softgoods,are used within the spac. industry for inflatable habitats,parac.utes and dec.lerator *** the safety and struc.ural integrity of these systems oc.urs through struc.ural health monitoring systems(SHM),whic. integrate non-invasive/non-destruc.ive testing methods to detec.,diagnose,and loc.te ***/load monitoring of these systems is limited while utilizing traditional strain gauges as these gauges are typic.lly stiff,operate at low temperatures,and fail when subjec.ed to high strain that is a result of high loading c.assifying them as unsuitable for SHM of soft struc.ural *** this work,a c.pac.tanc. based strain gauge(c.G)was fabric.ted via aerosol jet printing(AJP)using silver nanopartic.e ink on a flexible polymer *** strain gauges were then c.mpared to a c.mmerc.ally available high elongation resistanc.-based strain gauge(HE-RSG)for their ability to monitor strained Kevlar straps having a 26.7 kN(6 klbf)***,static.and c.c.ic.loads were used to c.arac.erize both types of strain monitoring *** c.Gs demonstrated superior performanc. for high elongation strain measurements when c.mpared to c.mmonly used HE-RSGs,and were observed to operate with a gauge fac.or of 5.2 when the elec.rode arrangement was perpendic.lar to the direc.ion of strain.
