High in vitro survival rate of sheep in vitro produced blastocysts vitrified with a new method and device

Background:To advance the use of embryo vitrification in veterinary practice,we developed a system in which embryo vitrification,warming and dilution can be performed within a *** in vitro produced embryos(IVEP)were vitrified at either early(EBs:n=74)or fully expanded blastocyst stage(FEBs:n=195),using a new device named"***",composed by a 0.25-m L straw with a 50-μm pore polycarbonate grid at one *** at each stage(EBs and FEBs)were vitrified by either Two-step(TS)or multi-step(mS;6 different concentrations of vitrification solutions)***-vitrified embryos(n=102)were maintained in in vitro culture as a *** consisted of placing the straws directly into 1.5 m L tubes containing a TCm-199 solution with three decreasing concentrations of *** re-expansion,embryo survival and hatching rate were evaluated at2,24 and 48 h post *** number of apoptotic cells was determined by TUNEL ***:Blastocyst re-expansion(2 h)after warming was higher(PmS and TS methods(77.90%and 71.25%,respectively)compared with the EBs group(mS:59.38%and TS:48.50%,respectively).Survival rates of vitrified FEBs after 24 h IVC were higher(Pmethods(mS and TS)than vitrified EBs(mS:56.25%;TS:42.42%)and was higher(PmS method(94.19%)compared with those in TS(83.75%).After 48 h of culture the hatching rate for FEBs vitrified in mS system(91.86%)was similar to control(91.89%),but higher than FEB TS(77.5%)and EBs vitrified in mS(37.5%)and TS(33.33%).Number of apoptotic cells were higher in EBs,irrespective of the system used,compared to *** number of apoptotic cells in FEBs vitrified with mS was comparable to the ***:A high survival rate of IVP embryos can be achieved by the new"***"device with hatching rates in vitro comparable with control fresh *** method has the potential for use in direct embryo transfer in field conditions.

Rice production accounts for approximately half of the freshwater resources utilized in agriculture,result-ing in greenhouse gas emissions such as methane(CH4)from flooded paddy *** address this chal-lenge,environmentally friendly and cost-effective water-saving techniques have become widely adopted in rice ***,the implementation of water-saving treatments(WsTs)in paddy-field rice has been associated with a substantial yield loss of up to 50%as well as a reduction in nitrogen use efficiency(NUE).In this study,we discovered that the target of rapamycin(TOR)signaling pathway is compromised in rice under *** profiling-coupled transcriptome sequencing(polysome-seq)analysis unveiled a substantial reduction in global translation in response to WST associated with the downregulation of TOR ***,biochemical,and genetic analyses revealed new insights into the impact of the positive TOR-S6K-RPS6 and negative TOR-mAF1 modules on translation repression under ***,ammonium exhibited a greater ability to alleviate growth constraints under WsT by enhancing TOR signaling,which simultaneously promoted uptake and utilization of ammonium and nitrogen *** further demonstrated that TOR modulates the ammonium transporter AmT1;1 as well as the amino acid permease APP1 and dipeptide transporter NPF7.3 at the translational level through the 5'untranslated ***,these findings reveal that enhancing TOR signaling could mitigate rice yield penalty due to WST by regulating the processes involved in protein synthesis and *** study will contribute to the breeding of new rice varieties with increased water and fertilizer utilization efficiency.

Enhancement of Swimming Endurance by Herbal Supplement m3P

Photomultiplication-type polymer photodetectors(Pm-PPDs)were achieved with polymer P3HT as donor and PY3Se-1V as acceptor based on structure of ITO/PEDOT:PSS/active layer/*** optimal weight ratio of P3HT to PY3Se-1V is about 100:*** of isolated electron traps are formed with PY3Se-1V surrounded by P3HT due to rather less content of PY3Se-1V in active layers and about 0.94 e V energy offset between the lowest unoccupied molecular orbitals(LUmO)of P3HT and *** optimal Pm-PPDs exhibit broad spectral response from 350 to 950 nm and external quantum efficiency(EQE)values of68,200%at 360 nm,26,400%at 630 nm and 19,500%at 850 nm under-15 V *** working mechanism of Pm-PPDs is attributed to the interfacial trap-assisted hole tunneling injection from external *** performance of Pm-PPDs can be further improved by incorporating appropriate PmBBDT with high hole mobility as the third *** EQE values of optimal ternary Pm-PPDs are increased to 105,000%at 360 nm,40,000%at 630 nm and 31,800%at 850 nm under-15 V bias,benefiting from the enhanced hole transport in ternary active *** optimal ternary Pm-PPDs were successfully applied in a light-controlled circuit to turn on or turn off light emitting diode(LED).

Development and first evaluation of an attractant impregnated adhesive tape against blood-sucking flies

Stable flies are one of the most important arthropod pests of livestock that reduce cattle weight gain and milk production leading to annual economic losses in excess of$2 billion to the US cattle *** host-seeking behavior is primarily mediated by associated odors from stable fly larval development environments and host *** present paper reports the development and evaluation of attractant-impregnated adhesive tapes to reduce stable fly attacks on *** bioassays showed that only m-cresol impregnated adhesive tapes caught significantly more stable flies(16±1)than the control tape without attractant added(7±1),with a 77%fly recapture ***-impregnated adhesive tapes deployed in cattle feedlots showed significant impacts in reducing fly population,with a total of one million stable flies captured over a period of three weeks(mean catches from 57596 to 102088 stable flies per trap per week).It further relieved cattle stress with a significant reduction of biting fly avoidance behavior,(6±0.4 cows observed with tail wagging in control vs.3±0.4 from the trap-deployed).The efficacy of the developed tapes lasted up to 1-week longevity,although 70%of m-cresol was released starting from the second *** m-cresol impregnated adhesive tape provided an 80%reduction in cattle stress due to stable fly *** is the first report of a technology developed by integrating an attractant compound into an adhesive material on a plastic film with demonstrated effectiveness in trapping biting flies that attack livestock animals.

The recently reported efficient polymerized small-molecule acceptors(PSmAs)usually adopt a regioregular backbone by polymerizing small-molecule acceptors precursors with a low-reactivity 5-brominated 3-(dicyanomethylidene)indan-1-one(IC)end group or its derivatives,leading to low molecular weight,and thus reduce active layer mechanical ***,a series of newly designed chlorinated PSmAs originating from isomeric IC end groups are developed by adjusting chlorinated positions and copolymerized sites on end groups to achieve high molecular weight,favorable intermolecular interaction,and improved physicochemical *** with regioregular PY2Se-Cl-o and PY2Se-Cl-m,regiorandom PY2Se-Cl-ran has a similar absorption profile,moderate lowest unoccupied molecular orbital level,and favorable intermolecular packing and crystallization ***,the binary Pm6:PY2Se-Cl-ran blend achieves better ductility with a crack-onset strain of 17.5% and improved power conversion efficiency(PCE)of 16.23% in all-polymer solar cells(all-PSCs)due to the higher molecular weight of PY2Se-Cl-ran and optimized blend morphology,while the ternary Pm6:J71:PY2Se-Cl-ran blend offers an impressive PCE approaching 17% and excellent device stability,which are all crucial for potential practical applications of all-PSCs in wearable *** date,the efficiency of 16.86% is the highest value reported for the regiorandom PSmAs-based all-PSCs and is also one of the best values reported for the *** work provides a new perspective to develop efficient all-PSCs,with all high active layer ductility,impressive PCE,and excellent device stability,towards practical applications.

Kava or kava-kava （Piper methysticum） is a plant used for the production of a ritual drink with tranquilizing properties. The kavalactones are the main compounds of the kava drinks （see Table 1 for names and structures of the main kavalactones）. There are many clinical trials on the use of kava product for anti-anxiety, and a systematic review supports the use of kava for the treatment of generalized anxiety （Sarris et al., 2009）. Adverse effects on the liver have been reported, and kava is regulated in a number of countries （Teschke, 2010）.

Nutrient cycling and greenhouse gas emissions from soil amended with biochar-manure mixtures

Integrating biochar into cattle diets has recently emerged as a potential management practice for improving on-farm ***,information concerning the cycling of biochar-manure mixtures is scarce.A 70-d incubation experiment was conducted within two surface(0–15 cm)mollisols with contrasting textures,i.e.,sandy clay loam(Raymond)and clayey(Lethbridge),to evaluate the effects of biochar(3 mg ha^(-1))on cumulative greenhouse gas(GHG)emissions and related fertility attributes in the presence or absence of cattle manure(120 mg ha^(-1)).Five treatments were included:i)non-amended soil(control,CK),ii)soil amended with pinewood biochar(B),iii)soil amended with beef cattle manure(m)(manure from cattle on a control diet),iv)soil amended with biochar-manure(Bm)(manure from cattle on a control diet,with pinewood biochar added at 20 g kg^(-1)of diet dry matter),and v)soil amended with B and m at the aforementioned rates(B+m).A total of 40 soil columns were prepared and incubated at 21℃and 60%–80%water-holding *** average,total CO_(2)fluxes increased by 2.2-and 3.8-fold under manure treatments(i.e.,m,Bm,and B+m),within Raymond and Lethbridge soils,respectively,relative to CK and ***,total CH4 fluxes were the highest(Pmond soil under B+m and Bm relative to CK and B,and in Lethbridge soil under m and Bm relative to CK and *** Lethbridge soil,application of Bm increased cumulative N_(2)O emissions by 1.8-fold relative to *** 70-d incubation,amendment with Bm increased(Pmond and Lethbridge soils compared with B.A similar pattern was observed for water-extractable organic carbon in both soils,with Bm augmenting(Pmanure,and/or biochar-manure have contrasting short-term effects on the biogeochemistry of *** relatively low application rates,biochar does not necessarily counterbalance manure-derived inpu
