Trait plasticity, not values, best corresponds with woodland plant success in novel and manipulated habitats

Aims The clustering of plants with similar leaf traits along environmental gradients may arise from adaptation as well as acclimation to het-erogeneous habitat *** the forces that shape plant leaf traits requires both linking variation in trait morphology with abiotic gradients and linking that trait variation with plant per-formance under varying abiotic *** the spectrum of plant types,shade-tolerant evergreen herbs are relatively low in trait plasticity,compared to deciduous and sun-adapted *** plants employ stress-tolerant strategies for survival,which coincide with relatively static trait morphologies,slow growth and hence a lower ability to adjust to changing environmental *** We investigate how the survival of two ecologically similar under-story evergreen species,Asarum arifolium and Hepatica nobilis,cor-responds with variation in six commonly measured functional traits(leaf area,specific leaf area,plant height,leaf number,leaf length and shoot mass)along natural and experimental abiotic *** examine temporal(the period 2007-9)and spatial(100 km)variations in these traits after(i)translocating 576 plants across a span from the southerm Appalachian Mountains in NC,USA,to the Piedmont,GA,USA,which includes north-and south-facing slope habitats and(ii)the experimental manipulation of diffuse light and soil *** findings We find that when translocated into a novel habitats,with novel environmental conditions that often are more extreme than the source habitat,both species appear capable of considerable mor-phological acclimation and generally converge to similar trait *** nobilis does not exhibit mean trait values particu-larly different from those of ***,but it demonstrates much greater phenotypic *** results indicate that relatively conservative plant species nonetheless acclimate and survive across heterogeneous environmental conditions.

Expression of androgen receptor target genes in skeletal muscle

We aimed to determine the mechanisms of the anabolic actions of androgens in skeletal muscle by investigating potential androgen receptor （AR）-regulated genes in in vitro and in vivo models. The expression of the myogenic regulatory factor myogenin was significantly decreased in skeletal muscle from testosterone-treated orchidectomized male mice compared to control orchidectomized males, and was increased in muscle from male AR knockout mice that lacked DNA binding activity （AR△ZF2） versus wildtype mice, demonstrating that myogenin is repressed by the androgen/AR pathway. The ubiquitin ligase Fbxo32 was repressed by 12 h dihydrotestosterone treatment in human skeletal muscle cell myoblasts, and c-Myc expression was decreased in testosterone-treated orchidectomized male muscle compared to control orchidectomized male muscle, and increased in AR△ZF2 muscle. The expression of a group of genes that regulate the transition from myoblast proliferation to differentiation, Tceal7, p57Kip2, IEf2 and calcineurin Aa, was increased in AR△ZF2 muscle, and the expression of all but p57kip2 was also decreased in testosterone-treated orchidectomized male muscle compared to control orchidectomized male muscle. We conclude that in males, androgens act via the AR in part to promote peak muscle mass by maintaining myoblasts in the proliferative state and delaying the transition to differentiation during muscle growth and development, and by suppressing ubiquitin ligase-mediated atrophy pathways to preserve muscle mass in adult muscle.

Synthesis and characterization of fluorescence active triazole-gold complexes

Fluorescence active fused-triazole was developed as a ligand in gold(I) cation coordination. The coordination ability was evaluated using NMR and fluorescence emission. Our previously reported N-2-aryl-triazoles, though gave good emission, could not form stable complexes with gold cations. A new N-fused-triazole(NFT) derivative was prepared and the ligand indicated good fluorescence emission, and formed strong coordination with gold cations. Both ligand and complex were characterized by X-ray crystallography. Fluorescence properties of various gold complexes(with different primary ligands) were evaluated, which suggested the feasibility of using fluorescence emission as potential tools for future mechanistic investigation.

Pancreatic tuberculosis in a human immunodeficiency virus positive patient:A case report

Despite the increased incidence of tuberculosis related to human immunodeficiency virus (HIV) in recent decades, pancreatic tuberculosis has rarely been described. We report a case of pancreatic tuberculosis in a 39-year- old African man who presented with progressive dysphagia, vomiting, weight loss and productive cough, accompanied by localized epigastric pain and one episode of melena. HIV-1 testing was positive and lymphocyte subset profile showed CD4 count of 9/mm3. Abdominal computed tomography (CT) scan with contrast revealed a cystic mass in the body of the pancreas, significant portal and retroperitoneal cystic adenopathy, and multiple cystic lesions in the spleen and liver. CT guided cyst aspiration and node biopsy detected Mycobacterium tuberculosis. The patient responded well on antituberculosis and antiretroviral therapy. Tuberculosis rarely involves the pancreas, probably due to the presence of pancreatic enzymes which interfere with the seeding of Mycobacterium tuberculosis. Pancreatic tuberculosis is considered to be the result of dissemination of the infection from nearby lymphatic nodes. Endoscopic ultrasound or CT guided fine needle aspiration for cytology is the recommended diagnostic technique. Although the prognosis is good with anti-tuberculosis treatment, it could be fatal without correct diagnosis and treatment. The clinician’s high index of suspicion of pancreatic tuberculosis and application of FNAB to obtain pathological evidence are extremely important to a correct diagnosis, especially in young HIV positive patients.

Quantification and pharmacokinetic study of tumor-targeting agent MHI148-clorgyline amide in mouse plasma using liquid chromatography-electrospray ionization tandem mass spectrometry

A high-performance liquid chromatography-electrospray ionization tandem mass spectrometric （HPLC- ESI-MS/MS） method was developed for the quantification of MHl148-clorgyline amide （NMl-amide）, a novel tumor-targeting monoamine oxidase A inhibitor, in mouse plasma. The method was validated in terms of sensitivity, precision, accuracy, recovery and stability and then applied to a pharmacokinetic study of NMl-amide in mice following intravenous administration. NMl-amide together with the internal standard （IS）, MHI-148, was extracted by protein precipitation using acetonitrile. Multiple reaction monitoring was used for quantification of NMl-amide by detecting m/z transition of 491.2-361.9, and 685.3-258.2 for NMl-amide and the IS, respectively. The lower limit of quantification （LLOQ） of the HPLC- MS/MS method for NMl-amide was 0.005 μg/mL and the linear calibration curve was acquired with R2〉 0.99 in the concentration range of 0.005-2μg/mL. The intra- and inter-day precisions of the assay were assessed by percentage of the coefficient of variations, which was within 9.8% at LLOQ and 14.0% for other quality control samples, whereas the mean accuracy ranged from 86.8% to 113.2%. The samples were stable under storage and experimental conditions. This method was successfully applied to a pharmacokinetic study in mice following intravenous administration of 5 mg/kg NMl-amide.

Protein evolution proceeds by two distinct processes： 1） individual mutation and selection for adaptive mutations and 2） rearrangement of entire domains within proteins into novel combinations, producing new protein families that combine functional properties in ways that previously did not exist. Domain rearrangement poses a challenge to sequence alignment-based search methods, such as BLAST, in predicting homology since the methodology implicitly assumes that related proteins primarily differ from each other by individual mutations. Moreover, there is ample evidence that the evolutionary process has used （and continues to use） domains as building blocks, therefore, it seems fit to utilize computational, domain-based methods to reconstruct that process. A challenge and opportunity for computational biology is how to use knowledge of evolutionary domain recombination to characterize families of proteins whose evolutionary history includes such recombination, to discover novel proteins, and to infer protein-protein interactions. In this paper we review techniques and databases that exploit our growing knowledge of “horizontal” protein evolution, and suggest possible areas of future development. We illustrate the power of the domain-based methods and the possible directions of future development by a case history in progress aiming at facilitating a particular approach to understanding microbial pathogenicity.

Prostate cancer:a presentation of climcopathologic prognosticators among Filipino and American men at radical prostatectomy

Lower incidence and mortality rates from prostate cancer(PCa)have been shown in Asian men in general compared to *** is the first study detailing the clinicopathologic features of resected prostate cancer in Filipino men living in the Philippines(PH)_This study investigated the supposed“lower risk”Filipino and“higher risk”American PCa patients from the PH and the United States of America(USA),*** examined 348(176 from PH,172 from USA)radical prostatectomy *** clinicopathologic features of both groups(age at time of diagnosis,preoperative prostate-specific antigen[pre-op PSA]level,Gleason score[GS],Grade groups[GG],margin involvement,extra prostatic extension[EPE],seminal vesicle invasion[SVI],and regional lymph node[RLN]metastasis)were *** of seven prognosticators examined were more strongly associated with Filipinos than with *** were older at diagnosis(PH:64.32±6.56 years vs USA:58.98±8.08 years)and had higher pre-op PSA levels(PH:21.39±46.40 ng ml^(-1) vs USA:7.63±9.19 ng ml^(-1)).Filipino men had more advanced grade,GG 2 with minor pattern 5(PH:6.2%vs USA:2.9%)and GG 5(PH:14.8%vs USA:3.5%).Likewise,other adverse pathological features in margin positivity(PH:52.3%vs USA:23.8%),focal EPE(PH:14.2% vs USA:2.3%),and SVI(PH:17.1%vs USA:5.8%)were more commonly observed in *** study reveals the prognostic disadvantage of Filipinos versus Americans and highlights an important difference of Filipinos from other studied Asian ethnicities that have repeatedly been shown to have lower-risk *** study,the first on Filipino PCa patients with RP,suggests the need to modify Western-based risk stratification when employed in other countries like the PH.

Oil-water interfacial tension, wettability alteration and foaming studies of natural surfactant extracted from Vernonia Amygdalina

Surfactant flooding is an enhanced oil recovery(EOR)method for recovering residual oil in the reservoir through mechanism of interfacial tension(IFT)reduction and wettability *** to toxicity and high cost associated with conventional surfactants,recent research has focussed on developing low-cost and environmentally benign ***,a low-cost green surfactant is extracted from Vernonia Amygdalina(VA)and appraised for EOR *** extracted surfactant was characterized using Fourier Transform Infrared(FTIR)and High-Pressure Liquid Chromatography(HPLC).The IFT of the synthesized surfactant at the oil-water interface was determined using Kruss ***,the foam stability of the synthesized surfactant was ***,the wettability of the saponin based natural surfactant(SBNS)at the rock-fluid interface was analysed using Dataphysics drop shape *** result revealed that SBNS(1 wt%concentration)stabilized foam for longer periods with half-life of 1100 ***,the synthesized surfactant was effective in lowering the IFT of oil-water interface from 18 mN/m to 0.97 mN/***,SBNS altered the wettability of sandstone cores to water-wetting condition by reducing the contact angle from 118.5° to 45.7°.Overall,SBNS exhibit excellent properties desirable for EOR and thereby recommended as supplementary alternative to conventional surfactants.
