FOXP3 protects conventional human T cells from premature restimulation-induced cell death

The adaptive immune response relies on specific apoptotic programs to maintain *** effector T cell(Tcon)expansion is constrained by both forkhead box P3(FOXP3)^(+)-regulatory T cells(Tregs)and restimulation-induced cell death(rICD),a proprlocidal apoptosis pathway triggered by repeated stimulation through the T-cell receptor(TCr).Constitutive FOXP3 expression protects Tregs from rICD by suppressing SlAM-associated protein(SAP),a key adaptor protein that amplifies TCr signaling *** role of transient FOXP3 induction in activated human CD4 and CD8 Tcons remains unresolved,but its expression is inversely correlated with acquired rICD ***,we describe a novel role for FOXP3 In protecting human Tcons from premature rICD during *** FOXP3-mediated protection from rICD in Tregs,FOXP3 protects Tcons through a distinct mechanism requiring de novo transcription that does not require SAP *** profiling and functional analyses of expanding Tcons revealed that FOXP3 enhances expression of the SlAM family receptor CD48,which in turn sustains basal autophagy and suppresses pro-apoptotic p53 *** CD48 and FOXP3 expression reduced p53 accumulation upon TCr ***,silencing FOXP3 expression or blocking CD48 decreased the mitochondrial membrane potential in expanding Tcons with a concomitant reduction in basal *** findings suggest that FOXP3 governs a distinct transcriptional program in early-stage effector Tcons that maintains rICD resistance via CD48-dependent protective autophagy and p53 suppression.

Amplitude-mode spectroscopy of chemically injected and photogenerated charge carriers in semiconducting single-walled carbon nanotubes

In one-dimensional semiconductors such as conjugated polymers and semiconducting single-walled carbon nanotubes(s-SWCNTs),injected charge carriers(electrons or holes)can have profound impacts on both electronic conductivity and optical spectra,even at low carrier *** charge-related spectral features is a key fundamental challenge with important technological ***,we employ a systematic suite of experimental and theoretical tools to understand the mid-infrared charge signatures of heavily p-type doped polymer-wrapped *** a broad range of nanotube diameters,we find that hole charge carriers induce strong Fano anti-resonances in mid-infrared transmission spectra that correspond to defect-related(D-band)and in-plane tangential(G-band)raman-active vibrational modes,along with antiresonances arising from infrared(Ir)-active polymer and SWCNT *** employ^(13)C isotope-labeled s-SWCNTs and a removable wrapping polymer to clarify the relative intensities,energies,and sources of all observed *** performed with the“amplitude mode model”are used to quantitatively reproduce raman spectra and also help to explain the outsized intensity of the D-band anti-resonance,relative to the G-band,observed for both moderately and degenerately doped *** results provide a framework for future studies of ground-and excited-state charge carriers in s-SWCNTs and a variety of low-dimensional materials.

T cell metabolism is dynamic and highly *** the intrinsic metabolic programs of T cell subsets are integral to their distinct differentiation and functional patterns,the ability of cells to acquire nutrients and cope with hostile microenvironments can limit these pathways.T cells must function in a wide variety of tissue settings,and how T cells interpret these signals to maintain an appropriate metabolic program for their demands or if metabolic mechanisms of immune suppression restrain immunity is an area of growing *** in inflamed and cancer tissues,a wide range of changes in physical conditions and nutrient availability are now acknowledged to shape *** include fever and increased temperatures,depletion of critical micro and macro-nutrients,and accumulation of inhibitory waste *** we review several of these factors and how the tissue microenvironment both shapes and constrains immunity.

Placental accommodations for transport and metabolism during intra-uterine crowding in pigs

litter size and birth weights are limited by uterine capacity, defined as the ability of the uterus to maintain the appropriate development of some number of conceptuses. Uterine capacity is the result of the combined effects of uterine, placental and embryo/fetal function. The number of living conceptuses that the uterus is capable of supporting is greater during early gestation compared to later gestation. Plots of log fetal weight versus log placental weight also indicate that fetal weights are less sensitive to reduced placental weight （and therefore reduced intrauterine space） in early gestation compared to late gestation. However, even in late gestation, mechanisms still exist that maintain fetal growth when the size of the placenta is reduced. One such mechanism is likely to be improved development of the folded placental-epithelial/maternal-epithelial bilayer. Fold depth, and therefore the maternal fetal interactive surface, increases as gestation advances and is greater in placenta from smal fetuses. On the fetal side of the placenta, the epithelial bilayer is embedded in stromal tissue. Glycosaminoglycans are major components of stroma, including hyaluronan and heparan sulfate. Hyaluronidases and heparanases are present within placental tissues, and likely play roles in modification of stromal components to facilitate fold development. Glycosaminoglycans are polymers of forms of glucose （glucosamine, glucuronic acid, iduronic acid） suggesting that glycosaminoglycan synthesis may compete with the glucose needs of the developing fetus. Pig conceptuses are fructogenic, such that a substantial portion of glucose transferred from mother to fetus is converted to fructose. Fructose is an intermediate product in the synthesis of glucosamine from glucose, and glucosamine is linked to regulation of trophoblast cell proliferation through regulation of mTOr. Glucosamine supplementation during late gestation alters placental development and increases litter size

Background： During late gestation the placental epithelial interface becomes highly folded, which involves changes in stromal hyaluronan. Hyaluronan is composed of glucoronate and N-acetyl-glucosamine. We hypothesized that supplementing gestating dams with glucosamine during this time would support placental folded-epithelial-bilayer development and increase litter size. In Exp. 1, gilts were unilaterally hysterectomizedovariectomized（UHO）. UHO gilts were mated and then supplemented daily with 10 g glucosamine（n = 16） or glucose（control, n = 17） from d 85 of gestation until slaughter（d 105）. At slaughter, the number of live fetuses was recorded and each live fetus and its placenta was weighed. Uterine wall samples adjacent to the largest and smallest fetuses within each litter were processed for histology. In Exp. 2, pregnant sows in a commercial sow farm were supplemented with either 10 g glucosamine or glucose daily from d 85 of gestation to farrowing. Total piglets born and born alive were recorded for each litter. In Exp. 3, the same commercial farm and same protocol were used except that the dose of glucosamine and glucose was doubled to 20 g/***： In Exp. 1, the number of live fetuses tended to be greater in glucosamine-treated UHO gilts（P = 0.098）.Placental morphometry indicated that the width of the folded bilayer was greater（P = 0.05） in glucosamine-treated gilts. In Exp. 2, litter size did not differ between glucosamine-and glucose-treated sows. However in Exp. 3, the increased dose of glucosamine resulted in a significant treatment by parity interaction（P ≤ 0.01）, in which total piglets born and born alive were greater in glucosamine treated sows of later parity（5 and 6）.Conclusions： These results indicated that glucosamine supplementation increased the width of the folds of the placental bilayer and increased litter size in later parity, intact pregnant commercial sows.

Quantifying relations between altered hydrology and fish community responses for streams in Minnesota

Functionalized, carbon nanotube material for the catalytic degradation of organophosphate nerve agents

recent world events have emphasized the need to develop innovative, functional materials that will safely neutralize chemical warfare （CW） agents in situ to protect military personnel and civilians from dermal exposure. Here, we demonstrate the efficacy of a novel, proof-of-concept design for a Cu-containing catalyst, chemically bonded to a single-wall carbon nanotube （SWCNT） structural support, to effectively degrade an organophosphate simulant. SWCNTs have high tensile strength and are flexible and light-weight, which make them a desirable structural component for unique, fabric-like materials. This study aims to develop a self-decontaminating, carbon nanotube-derived material that can ultimately be incorporated into a wearable fabric or protective material to minimize dermal exposure to organophosphate nerve agents and to prevent accidental exposure during decontamination procedures. Carboxylated SWCNTs were functionalized with a polymer, which contained Cu-chelating bipyridine groups, and their catalytic activity against an organophosphate simulant was measured over time. The catalytically active, functionalized nanomaterial was characterized using X-ray fluorescence and raman spectroscopy. Assuming zeroth-order reaction kinetics, the hydrolysis rate of the organophosphate simulant, as monitored by UV-vis absorption in the presence of the catalytically active nanomaterial, was 63 times faster than the uncatalyzed hydrolysis rate for a sample containing only carboxylated SWCNTs or a control sample containing no added nanotube materials.
