Dysfunctional stem and progenitor cells impair fracture healing with age

Successful fracture healing requires the simultaneous regeneration of both the bone and vasculature;mesenchymal stem cells (MSCs) are directed to replace the bone tissue, while endothelial progenitor cells (EPCs) form the new vasculature that supplies blood to the fracture site. In the elderly, the healing process is slowed, partly due to decreased regenerative function of these stem and progenitor cells. MSCs from older individuals are impaired with regard to cell number, proliferative capacity, ability to migrate, and osteochondrogenic differentiation potential. The proliferation, migration and function of EPCs are also compromised with advanced age. Although the reasons for cellular dysfunction with age are complex and multidimensional, reduced expression of growth factors, accumulation of oxidative damage from reactive oxygen species, and altered signaling of the Sirtuin-1 pathway are contributing factors to aging at the cellular level of both MSCs and EPCs. Because of these geriatric-specific issues, effective treatment for fracture repair may require new therapeutic techniques to restore cellular function. Some suggested directions for potential treatments include cellular therapies, pharmacological agents, treatments targeting age-related molecular mechanisms, and physical therapeutics. Advanced age is the primary risk factor for a fracture, due to the low bone mass and inferior bone quality associated with aging;a better understanding of the dysfunctional behavior of the aging cell will provide a foundation for new treatments to decrease healing time and reduce the development of complications during the extended recovery from fracture healing in the elderly.

Synthesis of Silver Nanoparticles from Honeybees and Its Antibacterial Potential

Honeybees (Apis mellifera) are important pollinators of flowering plants and agricultural crops contributing annually to billions of dollars in revenues to crop production. Honeybees have an average lifespan between 8 weeks to 5 years. Dead honeybees are abundantly available in beehives and can be utilized as an alternative source to synthesize nanoparticles. In recent years, biologically synthesized nanoparticles have been preferred over their chemical counterparts. However, honeybee-based-green synthesis of nanoparticles has not been explored yet. Herein, we report the biosynthesis of silver nanoparticles from honeybees and its antibacterial activity. The synthesis of silver nanoparticles was monitored visually through a gradual change in color. Furthermore, the biosynthesized nanoparticles were confirmed and characterized by UV-visible spectroscopy. Scanning Electron Microscope was utilized to analyze the average size and morphologies of the biosynthesized nanoparticles. Subsequently, the antibacterial potential of the biosynthesized silver nanoparticles was tested against selected Gram-positive and Gram-negative bacterial strains. It was found that a distinct color change from yellow to brown in the reaction solution suggested the formation of silver nanoparticles. The biosynthesized nanoparticles exhibited absorption maxima at 430 nm. The SEM analysis confirmed the spherical and cuboidal shape of the biosynthesized silver nanoparticles with a size range between 10 - 40 nm. Furthermore, the biosynthesized silver nanoparticles exhibited strong antimicrobial potential against tested Gram-positive and Gram-negative bacteria strains by aggregating on the cell surface. This study showcases the biomedical and agricultural applications of biosynthesized silver nanoparticles from honeybee wings. .

Metabolic collaboration between cells in the tumor microenvironment has a negligible effect on tumor growth

The tumor microenvironment is composed of a complex mixture of different cell types interacting under conditions of nutrient deprivation,but the metabolism therein is not fully understood due to difficulties in measuring metabolic fluxes and exchange of metabolites between different cell types in ***-scale metabolic modeling enables estimation of such exchange fluxes as well as an opportunity to gain insight into the metabolic behavior of individual cell ***,we estimated the availability of nutrients and oxygen within the tumor microenvironment using concentration measurements from blood together with a metabolite diffusion *** addition,we developed an approach to efficiently apply enzyme usage constraints in a comprehensive metabolic model of human *** combined modeling reproduced severe hypoxic conditions and the Warburg effect,and we found that limitations in enzymatic capacity contribute to cancer cells’preferential use of glutamine as a substrate to the citric acid ***,we investigated the common hypothesis that some stromal cells are exploited by cancer cells to produce metabolites useful for the cancer *** identified over 200 potential metabolites that could support collaboration between cancer cells and cancer-associated fibroblasts,but when limiting to metabolites previously identified to participate in such collaboration,no growth advantage was *** work highlights the importance of enzymatic capacity limitations for cell behaviors and exemplifies the utility of enzyme-constrained models for accurate prediction of metabolism in cells and tumor microenvironments.

Antimicrobial silicone skin adhesives facilitated by controlled octenidine release from glycerol compartments

For improved wound healing,antimicrobial adhesives are one path ***,with the current challenge of bacterial resistance,it is essential to choose the included drug *** is an obvious choice due to its broad antimicrobial efficacy and no reported bacterial *** its pure form,octenidine complexes efficiently with the platinum catalyst in the silicone composition,inhibiting the targeted hydrosilyla-tion reaction and hindering *** obstacle is overcome by screening octenidine with cyclodextrins in homogeneously dispersed glycerol droplets,suppressing Pt inhibition in the silicone *** efficiency is demonstrated using rheol-ogy,which shows that it is possible to incorporate one wt%of octenidine into glycerol–silicone adhesives in the presence of(2-hydroxypropyl)-β-cyclodextrin without affecting the adhe-sives’mechanical *** interaction between octeni-dine and(2-hydroxypropyl)-β-cyclodextrin through an inclusion complex is confirmed by ROESY *** this screening,octenidine is still released efficiently from the glycerol–silicone adhesives upon contact with water,and the resulting antimicrobial action is subsequently *** new technology constitutes a simple and efficient method for preparing wound care adhesives that actively inhibit the growth of four bacteria strains and one fungus.

The methodological basis and procedures for determination of Chinese normalization references and weighting factors according to the EDIP-method is described. According to Chinese industrial development intensity and population density, China was divided into three regions and the normalization references for each region were calculated on the basis of an inventory of all of the region's environmental emissions in 1990. The normalization reference was determined as the total environmental impact potential for the area in question in 1990(EP(j)90) divided by the population. The weighting factor was determined as the normalization reference (ER(j)90) divided by society's target contribution in the year 2000 based on Chinese political reduction plans, ER(j)T2000. This paper presents and discuss results obtained for eight different environmental impact categories relevant for China: global warming, stratospheric ozone depletion, acidification, nutrient enrichment, photochemical ozone formation and generation of bulk waste, hazardous waste and slag and ashes.

Satisfaction of Patients Examined with Mobile X-Ray vs. X-Ray at the Hospital—A Randomized Controlled Trial

Background: In the Municipality of Aarhus, Denmark, mobile X-ray is offered to selected patients when a transfer to the Department of Radiology may be an obstacle. To our knowledge, no studies have examined patient’s satisfaction with mobile X-ray in a randomized controlled trial, but international qualitative and quantitative studies report a high level of patient satisfaction with mobile X-ray. Purpose: The purpose of the study was to investigate whether patients in aged care facilities who were offered mobile X-ray were more satisfied with the X-ray examination compared to patients examined with X-ray at the Department of Radiology, Aarhus University Hospital. Design: A part of a randomized controlled trial. Methods: Satisfaction was measured using a self-developed questionnaire, containing 13 questions measuring satisfaction from different perspectives. Participants: Due to patient’s fragility, healthcare staff members answered questions concerning satisfaction on behalf of the patients, who had been examined with mobile X-ray (n = 66) or X-ray at the hospital (n = 63). The patients were living in nursing homes and homes for the elderly in Aarhus Municipality. Data: Data were collected and stored using the computer program REDCap. Data were statistically analyzed using Fisher’s exact test. Results: Patients examined with mobile X-ray had a significantly higher satisfaction rate than those examined with X-ray at the hospital. Conclusion: Satisfaction of patients examined with X-ray was reported by healthcare staff to be in favor of mobile X-ray.
