Survival of living organisms is fully dependent on their maintenance of genome integrity,being permanently threatened by replication stress in proliferating cells.Although the plant DNA damage response(DDR)regulator SOG1 has been demonstrated to cope with replication defects,accumulating evidence points to other pathways functioning independent of SOG1.Here,we report the roles of the Arabidopsis E2FA and EF2B transcription factors,two well-characterized regulators of DNA replication,in plant response to replication stress.Through a combination of reverse genetics and chromatin immunoprecipitation approaches,we show that E2FA and E2FB share many target genes with SOG1,providing evidence for their involvement in the DDR.Analysis of double-and triple-mutant combinations revealed that E2FB,rather than E2FA,plays the most prominent role in sustaining plant growth in the presence of replication defects,either operating antagonistically or synergistically with SOG1.Conversely,SOG1 aids in overcoming the replication defects of E2FA/E2FB-deficient plants.Collectively,our data reveal a complex transcriptional network controlling the replication stress response in which E2Fs and SOG1 act as key regulatory factors.

BACKGROUND Endoscopic treatment of pancreatic necrosis can be challenging and timeconsuming because sticky necrotic debris is sometimes difficult to remove. The over-the-scope-grasper, a new tool that has recently become available for this purpose, might also be useful for other indications. However, clinical data on the efficacy and safety of this new device are lacking.AIM To evaluate the technical success and safety of the device in a multicenter setting.METHODS The over-the-scope-grasper was used in nine selected endoscopic centers between November 2020and October 2021 for appropriate indications. Overall, 56 procedures were included in the study.We retrospectively evaluated procedural parameters of all endoscopic interventions using a predefined questionnaire, with special respect to technical success, indications, duration of intervention, type of sedation, and complications. In the case of pancreatic necrosectomy, the access route, stent type, number of necrosis pieces removed, and clinical handling were also recorded.RESULTS A total of 56 procedures were performed, with an overall technical success rate of 98%. Most of the procedures were endoscopic pancreatic necrosectomies(33 transgastric, 4 transduodenal). In 70%of the procedures, access to the necrotic cavity was established with a lumen apposing metal stent.The technical success of pancreatic necrosectomy was 97%, with a mean of 8 pieces(range, 2-25pieces) of necrosis removed in a mean procedure time of 59 min(range, 15-120 min). In addition,the device has been used to remove blood clots(n = 6), to clear insufficiency cavities before endoluminal vacuum therapy(n = 5), and to remove foreign bodies from the upper gastrointestinal tract(n = 8). In these cases, the technical success rate was 100%. No moderate or severe/fatal complications were reported in any of the 56 procedures.CONCLUSION These first multicenter data demonstrate that the over-the-scope-grasper is a promising device for endoscopic pancreatic necrosectomy, which is also appropriate for r

Scalable nanolaminated SERS multiwell cell culture assay

This paper presents a new cell culture platform enabling label-free surface-enhanced Raman spectroscopy(SERS)analysis of biological samples.The platform integrates a multilayered metal-insulator-metal nanolaminated SERS substrate and polydimethylsiloxane(PDMS)multiwells for the simultaneous analysis of cultured cells.Multiple cell lines,including breast normal and cancer cells and prostate cancer cells,were used to validate the applicability of this unique platform.The cell lines were cultured in different wells.The Raman spectra of over 100 cells from each cell line were collected and analyzed after 12h of introducing the cells to the assay.The unique Raman spectra of each cell line yielded biomarkers for identifying cancerous and normal cells.A kernel-based machine learning algorithm was used to extract the high-dimensional variables from the Raman spectra.Specifically,the nonnegative garrote on a kernel machine classifier is a hybrid approach with a mixed nonparametric model that considers the nonlinear relationships between the higher-dimension variables.The breast cancer cell lines and normal breast epithelial cells were distinguished with an accuracy close to 90%.The prediction rate between breast cancer cells and prostate cancer cells reached 94%.Four blind test groups were used to evaluate the prediction power of the SERS spectra.The peak intensities at the selected Raman shifts of the testing groups were selected and compared with the training groups used in the machine learning algorithm.The blind testing groups were correctly predicted 100% of the time,demonstrating the applicability of the multiwell SERS array for analyzing cell populations for cancer research.

Daytime HONO photolysis is an important source of atmospheric hydroxyl radicals(OH).Knowledge of HONO formation chemistry under typical haze conditions,however,is still limited.In the Multiphase chemistry experiment in Fogs and Aerosols in the North China Plain in 2018,we investigated the wintertime HONO formation and its atmospheric implications at a rural site Gucheng.Three different episodes based on atmospheric aerosol loading levels were classified:clean periods(CPs),moderately polluted periods(MPPs)and severely polluted periods(SPPs).Correlation analysis revealed that HONO formation via heteroge.neous conversion of NO_(2)was more efficient on aerosol surfaces than on ground,highlighting the important role of aerosols in promoting HONO formation.Daytime HONO budget analysis indicated a large missing source(with an average production rate of 0.66±0.26,0.97±0.47 and 1.45±0.55 ppbV/hr for CPs,MPPs and SPPs,respectively),which strongly correlated with photo-enhanced reactions(NO_(2)heterogeneous reaction and particulate nitrate photolysis).Average OH formation derived from HONO photolysis reached up to(0.92±0.71),(1.75±1.26)and(1.82±1.47)ppbV/hr in CPs,MPPs and SPPs respectively,much higher than that from O3 photolysis(i.e.,(0.004±0.004),(0.006±0.007)and(0.0035±0.0034)ppbV/hr).Such high OH production rates could markedly regulate the atmospheric oxidation capacity and hence promote the formation of secondary aerosols and pollutants.

目的评价舒更葡糖钠用于ASA分级Ⅲ或Ⅳ级高风险手术患者术后拮抗神经肌肉阻滞的安全性。尤其着重评估了心脏不良事件以及其他预先指定的临床关注不良事件的影响。方法符合ASA分级Ⅲ或Ⅳ级标准的患者,根据ASA分级和神经肌肉阻滞剂(罗库溴铵或维库溴铵)进行随机分组,新斯的明/格隆溴铵组:中度神经肌肉阻滞;舒更葡糖钠2 mg/kg组,中度神经肌肉阻滞;舒更葡糖钠4 mg/kg组,深度神经肌肉阻滞;舒更葡糖钠16 mg/kg组,深度神经肌肉阻滞(仅罗库溴铵)。主要研究终点包括治疗期间窦性心动过缓、窦性心动过速和其他心律失常的发生情况。结果344例患者进入随机化分配,331例患者接受了治疗[61%为男性,BMI(28.5±5.3)kg/m^(2),年龄(69±11)岁]。舒更葡糖钠2 mg/kg组治疗期间窦性心动过缓发生率显著降低,舒更葡糖钠2 mg/kg组和4 mg/kg组治疗期间窦性心动过速发生率显著降低(P0.05)。结论ASA分级Ⅲ或Ⅳ级高风险手术患者使用舒更葡糖钠拮抗罗库溴铵或维库溴铵诱导的中度和深度神经肌肉阻滞的安全性良好。

Cachexia and pancreatic cancer: Are there treatment options?

Cachexia is frequently described in patients with pancreatic ductal adenocarcinoma(PDAC)and is associated with reduced survival and quality of life.Unfortunately,the therapeutic options of this multi-factorial and complex syndrome are limited.This is due to the fact that,despite extensive preclinical and clinical research,the underlying pathological mechanisms leading to PDAC-associated cachexia are still not fully understood.Furthermore,there is still a lack of consensus on the definition of cachexia,which complicates the standardization of diagnosis and treatment as well as the analysis of the current literature.In order to provide an efficient therapy for cachexia,an early and reliable diagnosis and consistent monitoring is required,which can be challenging especially in obese patients.Although many substances have been tested in clinical and preclinical settings,so far none of them have been proven to have a long-term effect in ameliorating cancer-associated cachexia.However,recent studies have demonstrated that multidimensional therapeutic modalities are able to alleviate pancreatic cancerassociated cachexia and ultimately improve patients’outcome.In this current review,we propose a stepwise and pragmatic approach to facilitate and standardize the treatment of cachexia in pancreatic cancer patients.This strategy consists of nutritional,dietary,pharmacological,physical and psychological methods.

Aims Studies integrating phylogenetic history and large-scale community assembly are few,and many questions remain unanswered.Here,we use a global coastal dune plant data set to uncover the important factors in community assembly across scales from the local filtering processes to the global long-term diversification and dispersal dynamics.Coastal dune plant communities occur worldwide under a wide range of climatic and geologic conditions as well as in all biogeographic regions.However,global patterns in the phylogenetic composition of coastal dune plant communities have not previously been studied.Methods The data set comprised vegetation data from 18463 plots in New Zealand,South Africa,South America,North America and Europe.The phylogenetic tree comprised 2241 plant species from 149 families.We calculated phylogenetic clustering(Net Relatedness Index,NRI,and Nearest Taxon Index,NTI)of regional dune floras to estimate the amount of in situ diversification relative to the global dune species pool and evaluated the relative importance of land and climate barriers for these diversification patterns by geographic analyses of phylogenetic similarity.We then tested whether dune plant communities exhibit similar patterns of phylogenetic structure within regions.Finally,we calculated NRI for local communities relative to the regional species pool and tested for an association with functional traits(plant height and seed mass)thought to vary along sea–inland gradients.Important Findings Regional species pools were phylogenetically clustered relative to the global pool,indicating regional diversification.NTI showed stronger clustering than NRI pointing to the importance of especially recent diversifications within regions.The species pools grouped phylogenetically into two clusters on either side of the tropics suggesting greater dispersal rates within hemispheres than between hemispheres.Local NRI plot values confirmed that most communities were also phylogenetically clustered within regions.NRI values decreased with i
