Convergence of the turkey gut microbiota following cohabitation under commercial settings

Background:Microbiota development is a critical aspect of turkey poult maturation,and the succession of microbes in the turkey gut has been shown to correlate with poult *** purpose of this study was to determine the fate of the microbiota in turkey poults after movement of birds first raised in an isolated hatch brood system into a more traditional commercial brood facility with pre-existing *** poults were first divided into groups raised in conventional brood pens from day-of-hatch and those raised in an experimental hatch brood *** 11 days of growth,hatch brood birds were moved into pens within the conventional brood barn and monitored for an additional 18 *** of both hatch brood and conventional pen birds was performed at multiple timepoints throughout the study,and cecal content was used to analyze the bacterial microbiota using 16S rRNA gene amplicon ***:Alpha diversity tended to be higher in samples from conventional pen birds compared to those from hatch brood birds prior to the day 11 move,but the difference between systems was not observed *** beta diversity metrics,bacterial community succession appeared delayed in the hatch brood system birds pre-move,but post-move community composition quickly converged with that of the conventional pen *** was validated through assessment of significantly different genera between hatch brood system and conventional pen birds,where numbers of significantly different taxa quickly decreased following the *** key taxa previously associated with poult performance were delayed in their appearance and relative abundance in hatch brood ***:Overall,this study demonstrates that the use of isolated hatch brood systems has an impact on the poult gut microbiota,but its impact is resolved quickly once the birds are introduced into a conventional brood ***,the benefits of pathogen reduction with hatch brood systems may out

AIM:To investigate the role of genetic polymorphisms in the progression of hepatic fibrosis in hereditary ***:A cohort of 245 well-characterised C282Y homozygous patients with haemochromatosis was studied,with all subjects having liver biopsy data and DNA available for *** study assessed the association of eight single nucleotide polymorphisms(SNPs)in a total of six genes including toll-like receptor 4(TLR4),transforming growth factor-beta(TGF-β),oxoguanine DNA glycosylase,monocyte chemoattractant protein 1,chemokine C-C motif receptor 2 and interleukin-10 with liver disease *** was performed using high resolution melt analysis and *** results were analysed in relation to the stage of hepatic fibrosis in multivariate analysis incorporating other cofactors including alcohol consumption and hepatic iron ***:There were significant associations between the cofactors of male gender(P=0.0001),increasing age(P=0.006),alcohol consumption(P=0.0001),steatosis(P=0.03),hepatic iron concentration(P<0.0001)and the presence of hepatic *** the candidate gene polymorphisms studied,none showed a significant association with hepatic fibrosis in univariate or multivariate analysis incorporating *** also specifically studied patients with hepatic iron loading above threshold levels for cirrhosis and compared the genetic polymorphisms between those with no fibrosis vs cirrhosis however there was no significant effect from any of the candidate genes ***,in this large,well characterised cohort of patients there was no association between SNPs for TGF-βor TLR4and the presence of fibrosis,cirrhosis or increasing fibrosis stage in multivariate ***:In our large,well characterised group of haemochromatosis subjects we did not demonstrate any relationship between candidate gene polymorphisms and hepatic fibrosis or cirrhosis.

Trajectory design for a solar-sail mission to asteroid 2016 HO_(3)

This paper proposes the use of solar-sail technology currently under development at NASA Langley Research Center for a CubeSat rendezvous mission with asteroid 2016 HO_(3),a quasi-satellite of ***-optimal trajectories are sought for within a 2022–202_(3) launch window,starting from an assumed launcher ejection condition in the Earth-Moon *** optimal control problem is solved through a particular implementation of a direct pseudo-spectral method for which initial guesses are generated through a relatively simple and straightforward genetic algorithm search on the optimal launch date and sail *** results show that the trajectories take 2.16–4.21 years to complete,depending on the assumed solar-sail reflectance model and solar-sail *** assess the performance of solar-sail propulsion for this mission,the trajectory is also designed assuming the use of solar electric *** resulting fuel-optimal trajectories take longer to complete than the solar-sail trajectories and require a propellant consumption that exceeds the expected propellant capacity onboard the *** comparison demonstrates the superior performance of solar-sail technology for this mission.

AIM:To analyze the changing trends in penetrating keratoplasty(PKP)***:This retrospective study included all patients with PKP between 2006 and *** were classified using histological *** groups were as the following:pseudophakic or aphakic bullous keratopathy,regraft,acute necrotizing and ulcerative keratitis,keratoconus,Fuchs'dystrophy,corneal dystrophy other than Fuchs',corneal scar,other diagnoses and failed endothelial keratoplasty ***,two different time-periods(2006-2012 and 2013-2017)were ***:Totally 1721 histological analyses of 1214 patients were available for *** diagnoses were pseudophakic or aphakic bullous keratopathy in 487(28.3%),regraft in 443(25.7%),acute necrotizing and ulcerative keratitis in 313(18.2%),corneal scar in 153(8.9%),keratoconus in 140(8.1%).Fuchs'dystrophy in 61(3.5%),corneal dystrophy other than Fuchs'in 46(2.7%),other diagnoses in 44(2.6%)and failed endothelial keratoplasty graft in 34(2.0%)*** the first to the second analysed time-period,incidence of acute necrotizing and ulcerative keratitis,corneal scar,Fuchs'dystrophy increased(P<0.032 for all)and incidence of keratoconus significantly decreased(P=0.015).CONCLUSION:Pseudophakic or aphakic bullous keratopathy is the leading indication for PKP,followed by regraft and acute necrotizing and ulcerative keratitis.

CONTROL OF ANGIOGENESIS BY INHIBITOR OF PHOSPHOLIPASE A_2

Objective To investigate the potential effects of angiogenic process by secretory phospholipase A2 (sPLA2)inhibitor-HyPE(linking N-derivatized phosphatidyl-ethanolamine to hyaluronic acid)on human bone marrow endothelial cell line(HBME-1). Methods In order to examine the suppressing effects of HyPE on HBME-1 proliferation, migration, and capillary-like tube formation, HBME-1 were activated by angiogenic factor, specifically by basic fibroblast growth factor(b-FGF), vascular endothelial growth factor(VEGF),and oncostatin M(OSM)(at a final concentration of 25, 20, and 2.5 ng/mL, respectively), then HBME-1 proliferation, migration, and tube forma-tion were studied in the absence or presence of HyPE. HBME-1 tube formation was specially analyzed in fibrin gel. Results HyPE effectively inhibited HBME-1 proliferation and migration as a dose-dependent manner, whatever HBME-1 were grown in the control culture medium or stimulated with b-FGF, VEGF, or OSM. In fibrin, the formations of HBME-1 derived tube-like structures were enhanced by all angiogenic factors, but these were strongly suppressed by HyPE. Conclusions The results support the involvement of sPLA2 in angiogenesis. It is proposed that sPLA2 inhibitor introduces a novel approach in the control of cancer development.

Mechanisms of hepatocellular carcinoma progression

Hepatocellular carcinoma(HCC) is the most common primary malignancy of the liver. It is the second leading cause of cancer-related deaths worldwide, with a very poor prognosis. In the United States, there has been only minimal improvement in the prognosis for HCC patients over the past 15 years. Details of the molecular mechanisms and other mechanisms of HCC progression remain unclear. Consequently, there is an urgent need for better understanding of these mechanisms. HCC is often diagnosed at advanced stages, and most patients will therefore need systemic therapy, with sorafenib being the most common at the present time. However, sorafenib therapy only minimally enhances patient survival. This review provides a summary of some of the known mechanisms that either cause HCC or contribute to its progression. Included in this review are the roles of viral hepatitis, non-viral hepatitis, chronic alcohol intake, genetic predisposition and congenital abnormalities, toxic exposures, and autoimmune diseases of the liver. Well-established molecular mechanisms of HCC progression such as epithelial-mesenchymal transition, tumor-stromal interactions and the tumor microenvironment, cancer stem cells, and senescence bypass are also discussed. Additionally, we discuss the roles of circulating tumor cells,immunomodulation, and neural regulation as potential new mechanisms of HCC progression. A better understanding of these mechanisms could have implications for the development of novel and more effective therapeutic and prognostic strategies, which are critically needed.

Priority setting in health care: Attitudes of physicians and patients

Background: The opinion of physicians clearly counts in prioritizing health care, but there is little information on the rationales underlying treatment decisions and whether these rationales are accepted by patients. Objective: To compare physicians and patients regarding their understanding and use of therapeutic benefit and treatment costs as criteria for prioritizing health care. Methods: Seven physicians and twelve patients were purposefully selected to yield a heterogeneous sample. Participants were interviewed face-to-face, following a semi-structured topic guide comprising three scenarios that focused on interventions with low or unproven therapeutic benefit and high costs, respectively. For data analysis we used qualitative content analysis. Results: We found that patients and physicians differed in their understanding of therapeutic benefit, their expectations of what medicine can do and their use of costs as criteria for prioritizing health care. Physicians were less likely to assess a certain intervention as effec tive, and they less often accepted upper funding limits in health care. Unlike the physicians, patients raised non-medical aspects in decision making such as the patient’s consent and social inequalities. Conclusions: The revealed differences point toward the necessity to strengthen the doctor-patient communication, to improve information for patients about the possibilities and limits of health care and to gain a deeper understanding of their attitudes, wishes and concerns to reach an agreement by physicians and patients on the treatment to be implemented.
