Production of Lettuce Seedlings with Association of Diazotrophic Bacteria and Humic Acids

The objective of this work was to verify the responses in agronomic and physiological characters of lettuce seedlings when submitted to different doses of humic acids and the presence or absence of inoculation of the bacterium Azospirillum brasilense. The experiment was carried out at the Protected Cultivation Station and Biological Control Teacher Mário César Lopes, belonging to the Experimental Stations Nucleus of the State University of the West of Paraná. The experimental design was a randomized block design, in a 5 × 2 factorial scheme, with four repetitions, totaling 40 plots. The first factor consisted of five solutions containing humic acid (100%, 75%, 50%, 25% and 0%) and the second factor was the presence or absence of the diazotrophic bacterium Azospirillum brasilense obtained by commercial product. Germination tests were carried out in addition to the determination of agronomic characters such as the number of fully expanded leaves, plant height, stem diameter, main root length, shoot dry mass, root dry mass, total dry mass, and Dickson quality index. The association of diazotrophic bacteria and humic acids did not influence the emergency rate, mean emergence time, mean speed of emergency, emergence speed index, plant height, and root length of lettuce seedlings. The dilutions of humic acids, regardless of inoculation with bacteria diazotrophic favored the development of the number of leaves, stem diameter and Dickson quality index. Inoculation of lettuce seeds with diazotrophic bacteria associated with the addition of humic acids, favored the development of the dry mass of root and total dry matter.

Objective: As isolates of Cryptococcus are frequently kept collection stocks in institutions, sometimes without proper characterization, we sought to determine the genotype profiles, protease and phospholipase activities “in vitro” and the susceptibility testing for azoles and amphotericin B. Methodology: 84 isolates from several regions of Brazil (40 samples from clinical origin and 44 isolates from environmental origin) were maintained at the microorganism’s bank of the Biomedical Science Institute (ICB-USP) of the São Paulo University, in São Paulo, Brazil. This isolates was submitted fungal strains determination, DNA extraction and purification, determination of genotype by URA5 gene RFLP of CGB-positive isolates, protease and phospholipase activity and susceptibility to antifungals. Results: Of six CGB positive isolates tested by RFLP-PCR, only four presented a genomic profile consistent C. gattii species (VGII), while two other were C. neoformans (VNI and VNIII), indicating the existence of canavanine-resistante C. neoformans isolates in the culture collections. The clinical isolates secreted higher levels of phospholipase and environmental isolates but no differences were observed for the protease levels. Almost all isolates were sensible to azoles and amphotericin B. Conclusion: We point out in this research the existence of C. neoformans strains resistant to canavanine and intrinsic characteristic of C. gatti. These results demonstrate the importance to perform a detailed characterization of isolates kept in culture collections.

Potential Use of Tropical Grass for deferment in Semi-Arid Region

Management evaluation of Panicum maximum cv. massai (Massai guineagrass) under deferment contributes as a strategic reserve of forage in the Brazilian semi-arid region. The objective of this study was to evaluate biomass production and nutritive value of massai guineagrass in an area of thinned caatinga vegetation in two deferment periods (60 and 90 days after plot-leveling cut) and four use periods (30, 60, 90 and 120 days after the end of the rain season), in a random blocks design arranged in a split-plot (2 × 4), with three replications. Extending the deferment period of massai guineagrass promoted elevation of the biomass components, but compromised the pasture quality due to increasing stem biomass. The massai guineagrass deferred 30 days after raining season started and should use 90 days after raining season ended to ensure a forage supply of appropriate nutritional value. The increase in relative air humidity and the occurrence of rainfall after water restriction stimulate regrowth, resulting in improved nutritional value of the grass.

In carbon global cycle, the relationship between the terrestrial ecosystem and the atmosphere where there are, among others, gases that contribute to the greenhouse effect, has become object of relevant scientific interest. The content of organic matter in soil, expressed by its supplies as well as the organic matter degree of stability, are factors that can prevent the soil from acting as a drain and at the same time contribute for it to become a source of those gases. The variations in the way land is used in Brazil are factors responsible for the increase in emission of greenhouse effect gases. Based on these facts, this study was aimed to evaluate the CO2 and CH4 efflux using a gas retention chamber, and to associate these emissions to the organic carbon content in the soil. Two different areas were selected for the study, one in Tijuca Forest National Park, in a forest area, and the other at the Rio de Janeiro Federal Rural University campus. In the latter, the area was stratified in three sub areas according to the vegetation, use and water saturation degree. Samplings were performed during 8 months between 2013 and 2014.

AIM: To investigate the potential of bone marrow mononuclear cells (BM-MCs) in the regeneration of hepatic lesions induced by Schistosoma mansoni (***) chronic infection. METHODS: Female mice chronically infected with *** were treated with BM-MCs obtained from male green fluorescent protein (GFP) transgenic mice by intravenous or intralobular injections. Control mice received injections of saline in similar conditions. Enzyme-linked immunosorbent assay (ELISA) assay fortransforming growth factor-beta (TGF-β), polymerase chain reaction (PCR) for GFP DNA, immunofluorescence and morphometric studies were performed. RESULTS: Transplanted GFP+ cells migrated to granuloma areas and reduced the percentage of liver fibrosis. The presence of donor-derived cells was confirmed by Fluorescence in situ hybridization (FISH) analysis for detection of cells bearing Y chromosome and by PCR analysis for detection of GFP DNA. The levels of TGF-β, a cytokine associated with fibrosis deposition, in liver fragments of mice submitted to therapy were reduced. The number of oval cells in liver sections of ***-infected mice increased 3-4 fold after transplantation. A partial recovery in albumin expression, which is decreased upon infection with ***, was found in livers of infected mice after cellular therapy. CONCLUSION: In conclusion, transplanted BMCs migrate to and reduce the damage of chronic fibrotic liver lesions caused by ***.

Multifunctional and diversified agriculture can address diverging pressures and demands by simultaneously enhancing productivity,biodiversity,and the provision of ecosystem *** use of digital technologies can support this by designing and managing resource-efficient and context-specific agricultural *** present the Digital Agricultural Knowledge and Information System(DAKIS)to demonstrate an approach that employs digital technologies to enable decision-making towards diversified and sustainable *** develop the DAKIS,we specified,together with stakeholders,requirements for a knowledge-based decision-support tool and reviewed the literature to identify limitations in the current generation of *** results of the review point towards recurring challenges regarding the consideration of ecosystem services and biodiversity,the capacity to foster communication and cooperation between farmers and other actors,and the ability to link multiple spatiotemporal scales and sustainability *** overcome these challenges,the DAKIS provides a digital platform to support farmers'decision-making on land use and management via an integrative spatiotemporally explicit approach that analyses a wide range of data from various *** approach integrates remote and in situ sensors,artificial intelligence,modelling,stakeholder-stated demand for biodiversity and ecosystem services,and participatory sustainability impact assessment to address the diverse drivers affecting agricultural land use and management design,including natural and agronomic factors,economic and policy considerations,and socio-cultural preferences and ***,the DAKIS embeds the consideration of ecosystem services,biodiversity,and sustainability into farmers'decision-making and enables learning and progress towards site-adapted small-scale multifunctional and diversified agriculture while simultaneously supporting farmers'objectives and societal demands.
