Proteome changes of porcine follicular fluid during follicle development

Background:Ovarian follicular fluid influences follicle and oocyte growth,but the fluctuation of its protein content during folliculogenesis has not been comprehensively *** we used a shotgun approach and bioinformatics analyses to investigate and compare the proteomes of porcine follicular fluid(pFF)obtained from small(mm),medium(4–6 mm)and large(>6–12 mm)***:Follicular fluid samples containing highest estrogen levels were selected as non-atretic from small(SNA:26.1±15 ng/mL),medium(mNA:162±54 ng/mL),and large(LNA:290±37 ng/mL)follicles for proteomic *** detected 1627,1699,and 1756 proteins in SNA,mNA,and LNA samples,*** 60–63%of total proteins were specific to each sample,11–13%were shared in pairwise comparisons,and 247 proteins were shared among all *** categorization indicated comparable gene ontology(GO)terms distribution per cellular component,molecular function,and biological process categories across samples;however,the ranking of highly significantly enriched GO terms per category revealed differences between *** patterns of proteinto-protein interactions varied throughout follicle development,and proteins such as serine protease inhibitor,clade E(SERPINE);plasminogen activator,urokinase(PLAU);and plasminogen activator,urokinase receptor(PLAUR)appeared stage-specific to SNA,mNA,and LNA,***“complement and coagulation cascades”was the common major ***,properdin and fibulin-1 were abundant proteins that appeared absent in LNA ***:This study provides extensive and functional analyses of the pFF proteome changes during folliculogenesis and offers the potential for novel biomarker discovery in pFF for oocyte quality assessment.

Background: Advances in nanotechnology have permitted molecular-based targeting of cells through safe and biocompatible magnetic nanoparticles(mNP). Their use to detect and remove damaged spermatozoa from semen doses could be of great interest. Here, mNP were synthesized and tested for their ability to target apoptotic(annexin V) and acrosome-reacted(lectin) boar spermatozoa, for high-throughout retrieval in a magnetic field(nanoselection). The potential impacts of nanoselection on sperm functions and performance of offspring sired by sperm subjected to nanoselection were determined. Fresh harvested and extended boar semen was mixed with various amounts(0, 87.5, and 175 μg) of mNP-conjugates(Annexin V-mNP or Lectin-mNP) and incubated(10 to15 min) for 37 °C in Exp. 1. In Exp. 2, extended semen was mixed with optimal concentrations of mNP-conjugates and incubated(0, 30, 90, or 120 min). In Exp. 3, the synergistic effects of both mNP-conjugates(87.5 μg– 30 min)on spermatozoa was evaluated, followed by sperm fertility assessments through pregnancy of inseminated gilts and performance of neonatal offspring. Sperm motion, viability, and morphology characteristics were evaluated in all ***: Transmission electron microscopy, atomic force microscopy, and hyperspectral imaging techniques were used to confirm attachment of mNP-conjugates to damaged spermatozoa. The motility of nanoselected spermatozoa was improved(P m, as assessed by the abundance of reactive oxygen species and the integrity of the acrosome, plasma membrane, and mitochondrial membrane was not different between nanoselected and control spermatozoa. C-Nitroso species are characterized by a unique nitrogen–oxygen combination located next to a carbon backbone,which confers them a unique ambiphilic and high reactivity towards nucleophilic,electrophilic but also radical *** this ambivalence can be seen as a strong asset for developing versatile synthetic aminohydroxylation and/or hydroxylamination processes mainly through nitroso Diels–Alder,nitroso ene,and nitrosoaldol reactions,it also contributes to complex optimization and rationalization arising from the many competitive pathways,that is,the occurrence of regioisomers and stereoisomers,as well as dimerization and tautomerization *** reactivity profiles are usually seen by synthetic organic chemists as major hurdles to overcome,despite the armada of analytical and purification methods available to them,and hence to achieve selective and exploitable developments of such *** rise of computational chemistry and resources has certainly changed their perspectives,since it provides a very different angle to gather insights on intrinsic properties,reactivity,and *** silico chemistry also provides a robust alternative to time and resource-consuming synthetic work and can therefore contribute to alleviate wasteful preparations by guiding the chemist toward the best combination of reagents to achieve high selectivity and *** synergistic combination of synthetic organic chemistry and computational chemistry,within the specific context of the complex chemistry of C-nitroso species,is discussed in this *** review aims at giving an overview of the molecular and chemical properties obtained through computational chemistry as an enabling support for the rationalization and optimization of reactions relying on ambiphilic C-nitroso species over the 3 last *** provides clear,concise,and illustrated guidelines for the synthetic chemist in search of inspiration through computations.

Experimental evidence of obesity as a risk factor for severe acute pancreatitis

The incidence of acute pancreatitis, an inflammation of the pancreas, is increasing worldwide. Pancreatic injury is mild in 80%-90% of patients who recover without complications. The remaining patients may develop a severe disease with local complications such as acinar cell necrosis, abscess and remote organ injury including lung injury. The early prediction of the severity of the disease is an important goal for physicians in management of patients with acute pancreatitis in order to optimize the therapy and to prevent organ dysfunction and local complications. For that purpose, multiple clinical scale scores have been applied to patients with acute pancreatitis. Recently, a new problem has emerged： the increased severity of the disease in obese patients. However, the mechanisms by which obesity increases the severity of acute pancreatitis are unclear. Several hypotheses have been suggested：（1） obese patients have an increased inflammation within the pancreas; （2） obese patients have an increased accumulation of fat within and around the pancreas where necrosis is often located; （3） increase in both peri- and intra-pancreatic fat and inflammatory cells explain the high incidence of pancreatic inflammation and necrosis in obese patients; （4） hepatic dysfunction associated with obesity might enhance the systemic inflammatory response by altering the detoxiflcation of inflammatory mediators; and （5） ventilation/perfusion mismatch leading to hypoxia associated with a low pancreatic flow might reduce the pancreatic oxygenation and further enhance pancreatic injury. Recent experimental investigations also show an increased mortality and morbidity in obese rodents with acute pancreatitis and the implication of the adipokines leptin and adiponectin. Such models are important to investigate whether the inflammatory response of the disease is enhanced by obesity. Ultrafast synthesis and sintering of materials in a single running experiment approach by using electric fields

Processing of materials in the form of ceramics normally involves several steps including calcination at a relatively low temperature for synthesis of the end-product powder and sintering at a high temperature for *** work we have been developing introduces a novel approach enabling synthesis plus sintering of materials in a single running experiment by using electric fields,ending with dense ceramics that display grains noticeably finer than in conventional *** new paradigm is fully illustrated with experiments conducted on amorphous CaCu3Ti4O12 precursor powder,shown to experience,on heating,crystallization through intermediate phases,followed by chemical reaction leading to synthesis of the end-product powder,plus densification depending on field *** processing time and furnace temperature are considerably reduced,demonstrating that enhanced synthesis and sintering rates applied under field *** results found in Bi2/3Cu3Ti4O12are also *** different factors that may contribute to this unique scenario,including Joule heating,defect generation,and reduction of free energy for nuclei formation promoted by the applied field,are briefly ***,the findings we bring here are exclusive as they show an exploitable way that allows rapid processing of materials with good control over particle and grain coarsening.

目的:研究仙人掌果提取液在动物体内和体外对卵巢癌的抑制作用。方法:用Crystalviolet方法测定、细胞周期分析及细胞凋亡TUNEL分析观察仙人掌果提取液在体外对卵巢癌细胞生长的抑制作用;仙人掌果提取液对人卵巢癌的动物模型的治疗效果。结果:2种卵巢癌细胞(OVCA420和SKOV3)对于不同浓度仙人掌果提取液反应敏感程度不同,IC50分别是0·8%和8%,正常卵巢上皮细胞(IOSE)的IC50为2%;仙人掌果提取液可以有效的抑制卵巢癌在动物体内的生长(Pmide(4HPR)作用相似(P>0·05)。结论:仙人掌果提取液对体外卵巢癌细胞生长和动物体内的卵巢癌生长均有明显抑制作用。

Enhancing laccase stability and activity for dyes decolorization using ZIF-8@mWCNT nanocomposite

The continuous use of chemical dyes in various industries,and their discharge into industrial effluents,results in severe problems to human life and water *** have the ability to decolorize dyes and toxic chemicals in industrial effluents as green *** possible industrial applications have been limited by poor reusability,low stability,and loss of free laccase *** this research,lac-case was immobilized on zeolitic imidazolate framework coated multi-walled carbon nanotubes(Laccase@ZIF-8@mWCNTs)via metal affinity adsorption to develop an easy separable and stable *** optimum reaction conditions for immobilized laccase are at a pH of 3.0 and a temperature of 60℃.The immobilized laccase was enhanced in storage and thermal *** results indicated that Laccase@ZIF-8@mWCNTs still maintained 68%of its original activity after 10 times of repeated *** importantly,the biocatalytic system was applied for decolorization of different dyes(20 mg·L^(-1))without a mediator,and up to 97.4%for Eriochrome black T and 95.6%Acid red 88 was achieved in 25 *** with these properties may be used in a variety of environmental and industrial applications.
