Connexin 43 (CX43) is one of the major components of gap junctions, the structures responsible for the intercellular communication and transmission of the electrical impulse in the left ventricle. There is limited information on the histological changes of CX43 with age and their effect on electrophysiology, especially in humans. Here, we analyzed left ventricular biopsies from living donors starting at midlife to characterize age-related CX43 remodeling. We assessed its quantity, degree of lateralization, and spatial heterogeneity together with fibrotic deposition. We observed no significant age-related remodeling of CX43. Only spatial heterogeneity increased slightly with age, and this increase was better explained by biological age than by chronological age. Importantly, we found that CX43 features varied considerably among individuals in our population with no relevant relationship to age or fibrosis content, in contrast to animal species. We used our experimental results to feed computational models of human ventricular electrophysiology and to assess the effects of interindividual differences in specific features of CX43 and fibrosis on conduction velocity, action potential duration, and arrhythmogenicity. We found that larger amounts of fibrosis were associated with the highest arrhythmic risk, with this risk being increased when fibrosis deposition was combined with a reduction in CX43 amount and/or with an increase in CX43 spatial heterogeneity. These mechanisms underlying high arrhythmic risk in some individuals were not associated with age in our study population. In conclusion, our data rule out CX43 remodeling as an age-related arrhythmic substrate in the population beyond midlife, but highlight its potential as a proarrhythmic factor at the individual level, especially when combined with increased fibrosis.

A large number of anomalous extension twins,with low or even negative twinning Schmid factors,were found to nucleate and grow in a strongly textured mg-1Al alloy during tensile deformation along the extruded *** deformation mechanisms responsible for this behaviour were investigated through in-situ electron back-scattered diffraction,grain reference orientation deviation,and slip trace-modified lattice *** was found that anomalous extension twins nucleated mainly at the onset of plastic deformation at or near grain boundary triple *** were associated with the severe strain incompatibility between neighbour grains as a result from the differentbasal slip-induced lattice ***,the anomalous twins were able to grow with the applied strain due to the continuous activation ofbasal slip in different neighbour grains,which enhanced the strain *** results reveal the complexity of the deformation mechanisms in mg alloys at the local level when deformed along hard orientations.

The combination of flow elasticity and inertia has emerged as a viable tool for focusing and manipulating particles using *** there is considerable interest in the field of elasto-inertial microfluidics owing to its potential applications,research on particle focusing has been mostly limited to low Reynolds numbers(Remigration toward equilibrium positions has not been extensively *** this work,we thoroughly studied particle focusing on the dynamic range of flow rates and particle migration using straight microchannels with a single inlet high aspect *** initially explored several parameters that had an impact on particle focusing,such as the particle size,channel dimensions,concentration of viscoelastic fluid,and flow *** experimental work covered a wide range of dimensionless numbers(0.05mbermbermberm *** results showed that the particle size played a dominant role,and by tuning the parameters,particle focusing could be achieved at Reynolds numbers ranging from 0.2(1μL/min)to 85(250μL/min).Furthermore,we numerically and experimentally studied particle migration and reported differential particle migration for high-resolution separations of 5μm,7μm and 10μm particles in a sheathless flow at a throughput of 150μL/*** work elucidates the complex particle transport in elasto-inertial flows and has great potential for the development of high-throughput and high-resolution particle separation for biomedical and environmental applications.

When data privacy is imposed as a necessity,Federated learning(FL)emerges as a relevant artificial intelligence field for developing machine learning(mL)models in a distributed and decentralized *** allows mL models to be trained on local devices without any need for centralized data transfer,thereby reducing both the exposure of sensitive data and the possibility of data interception by malicious third *** paradigm has gained momentum in the last few years,spurred by the plethora of real-world applications that have leveraged its ability to improve the efficiency of distributed learning and to accommodate numerous participants with their data *** virtue of FL,models can be learned from all such distributed data sources while preserving data *** aim of this paper is to provide a practical tutorial on FL,including a short methodology and a systematic analysis of existing software ***,our tutorial provides exemplary cases of study from three complementary perspectives:i)Foundations of FL,describing the main components of FL,from key elements to FL categories;ii)Implementation guidelines and exemplary cases of study,by systematically examining the functionalities provided by existing software frameworks for FL deployment,devising a methodology to design a FL scenario,and providing exemplary cases of study with source code for different mL approaches;and iii)Trends,shortly reviewing a non-exhaustive list of research directions that are under active investigation in the current FL *** ultimate purpose of this work is to establish itself as a referential work for researchers,developers,and data scientists willing to explore the capabilities of FL in practical applications.

Using Lean manufacturing(Lm)tools in production processes is crucial for companies’economic,environmental,and social sustainability *** study shows a structural equation model(SEm)that shows the relationship between Lm Tools like Kaizen(KAI),Gemba(GEm),Value Stream mapping(VSm)and Key Performance Indicator(KPI)with Economic Sustainability(ECS).Seven hypotheses were evaluated with data from 179 responses to a questionnaire about the mexican maquiladora industry,showing that these variables are *** a 95%confidence level,the model was evaluated using the partial least squares *** findings indicate that the relationships between KAI and GEm and KAI and VSm have the strongest relationship,followed by VSm and ECS;however,VSm has the strongest effect on *** on these findings,it is recommended that managers adopt a continuous improvement(KAI)approach based on working directly on the shop floor(GEm and VSm)to support their decisions regarding economic growth(ECS).

multispecies forests have received increased scientific attention,driven by the hypothesis that biodiversity improves ecological ***,a greater species diversity presents challenges for forest management and *** study aims to develop basal area growth models for tree species *** analysis is based on a dataset of 423 permanent plots(2,500 m^(2))located in temperate forests in Durango,***,we define tree species cohorts based on individual and neighborhood-based variables using a combination of principal component and cluster ***,we estimate the basal area increment of each cohort through the generalized additive model to describe the effect of tree size,competition,stand density and site *** principal component and cluster analyses assign a total of 37 tree species to eight cohorts that differed primarily with regard to the distribution of tree size and vertical position within the *** generalized additive models provide satisfactory estimates of tree growth for the species cohorts,explaining between 19 and 53 percent of the total variation of basal area increment,and highlight the following results:i)most cohorts show a"rise-and-fall"effect of tree size on tree growth;ii)surprisingly,the competition index"basal area of larger trees"had showed a positive effect in four of the eight cohorts;iii)stand density had a negative effect on basal area increment,though the effect was minor in medium-and high-density stands,and iv)basal area growth was positively correlated with site quality except for an oak *** developed species cohorts and growth models provide insight into their particular ecological features and growth patterns that may support the development of sustainable management strategies for temperate multispecies forests.

BACKGROUND A new nomenclature consensus has emerged for liver diseases that were previously known as non-alcoholic fatty liver disease(NAFLD)and metabolic dysfunction-associated fatty liver disease(mAFLD).They are now defined as metabolic dysfunction-associated steatotic liver disease(mASLD),which includes cardiometabolic criteria in *** condition,extensively studied in obese or overweight patients,constitutes around 30%of the population,with a steady increase *** patients account for approximately 10%-15%of the mASLD ***,the pathogenesis is complex and is not well *** To systematically review the literature on the diagnosis,pathogenesis,characteristics,and prognosis in lean mASLD patients and provide an interpretation of these new *** We conducted a comprehensive database search on Pubmed and Google Scholar between January 2012 and September 2023,specifically focusing on lean NAFLD,mAFLD,or mASLD *** include original articles with patients aged 18 years or older,with a lean body mass index categorized according to the World Health Organization criteria,using a cutoff of 25 kg/m2 for the general population and 23 kg/m2 for the Asian *** We include 85 studies in our *** findings revealed that,for lean NAFLD patients,the prevalence rate varied widely,ranging from 3.8%to 34.1%.The precise pathogenesis mechanism remained elusive,with associations found in genetic variants,epigenetic modifications,and adaptative metabolic *** risk factors included metabolic syndrome,hypertension,and type 2 diabetes mellitus,but their prevalence varied based on the comparison group involving lean *** non-invasive tools,Fibrosis-4 index outperformed the NAFLD fibrosis score in lean *** modifications aided in reducing hepatic steatosis and improving cardiometabolic profiles,with some medications showing efficacy to a lesser ***,lean NAFLD patients ex

The potential of starch-chitosan blends with poloxamer for the preparation of microparticles by spray-drying

Using biopolymers as wall materials in spray drying poses challenges,particularly in attaining flowability and thermal stability among their physicochemical *** paper addresses these challenges by preparing microparticles using a blend of starch–chitosan and a poloxamer,commercially named Pluronic®*** aimed to elucidate the effects of varying poloxamer concentrations on the resulting particles through the spray drying *** with a poloxamer concentration of 3%(w/v)demonstrated a notably higher yield,especially when compared to those with 0%and 1%*** with 3%and 5%(w/v)poloxamer displayed a narrower particle size distribution,with the 3%blend showing a superior yield attributed to arrangements of blend components that improve flowability.X-ray diffraction analysis showcased the characteristic peaks of A-type starch form,with shifts suggesting enhanced interactions between *** with increased poloxamer content showed elevated thermal degradation temperatures,with the 3%blend registering a significant rise,opening avenues for encapsulating heat-sensitive *** study primarily focuses on the preparation and basic characterization of *** underscores the potential of blends with optimal poloxamer concentrations in microencapsulation,emphasizing further research to harness their capabilities thoroughly.

Luminescence constitutes a unique source of insight into hot carrier processes in metals,including those in plasmonic nanostructures used for sensing and energy ***,being weak in nature,metal luminescence remains poorly understood,its microscopic origin strongly debated,and its potential for unraveling nanoscale carrier dynamics largely ***,we reveal quantum-mechanical effects in the luminescence emanating from thin monocrystalline gold ***,we present experimental evidence,supported by first-principles simulations,to demonstrate its photoluminescence origin(i.e.,radiative emission from electron/hole recombination)when exciting in the interband *** model allows us to identify changes to the measured gold luminescence due to quantum-mechanical effects as the gold film thickness is ***,such effects are observable in the luminescence signal from flakes up to 40 nm in thickness,associated with the out-of-plane discreteness of the electronic band structure near the Fermi *** qualitatively reproduce the observations with first-principles modeling,thus establishing a unified description of luminescence in gold monocrystalline flakes and enabling its widespread application as a probe of carrier dynamics and light-matter interactions in this *** study paves the way for future explorations of hot carriers and charge-transfer dynamics in a multitude of material systems.
