Sulfonylurea receptor(SUR) belongs to the adenosine 5′-triphosphate(ATp)-binding cassette(ABC) transporter family;however,SUR is associated with ion channels and acts as a regulatory subunit determining the opening or closing of the ***8 and Abcc9 genes code for the proteins SUR1 and SUR2,*** SUR1 transcript encodes a protein of 1582 amino acids with a mass around 140–177 k Da expressed in the pancreas,brain,heart,and other *** is well known that SUR1 assembles with Kir6.2 and TRpM4 to establish K_(ATp) channels and non-selective cation channels,***8 and 9 are alternatively spliced,and the resulting transcripts encode different isoforms of SUR1 and SUR2,which have been detected by different experimental ***,the use of binding assays to sulfonylureas and Western blotting has allowed the detection of shorter forms of SUR(-65 k Da).Identity of the SUR1 variants has not been clarified,and some authors have suggested that the shorter forms are ***,immunoprecipitation assays have shown that SUR2 short forms are part of a functional channel even coexisting with the typical forms of the receptor in the *** evidence confirms that the structure of the short forms of the SURs is fully functional and does not lose the ability to interact with the *** structural changes in short forms of SUR modify its affinity to ATp,regulation of its expression might represent an advantage in pathologies where ATp concentrations decrease and a therapeutic target to induce ***,the expression of SUR1 variants might be induced by conditions associated to the decrease of energetic substrates in the brain(*** stroke and epilepsy).In this review,we want to contribute to the knowledge of SUR1 complexity by analyzing evidence that shows the existence of short SUR1 variants and its possible implications in brain function.

In recent years,gold nanoparticles have demonstrated excellent enzyme-mimicking activities which resemble those of peroxidase,oxidase,catalase,superoxide dismutase or ***,merged with their ease of synthesis,tunability,biocompatibility and low cost,makes them excellent candidates when compared with biological enzymes for applications in biomedicine or biochemical ***,over 200 research papers have been systematically reviewed to present the recent progress on the fundamentals of gold nanozymes and their potential *** review reveals that the morphology and surface chemistry of the nanoparticles play an important role in their catalytic properties,as well as external parameters such as pH or ***,real applications often require specific biorecognition elements to be immobilized onto the nanozymes,leading to unexpected positive or negative effects on their ***,rational design of efficient nanozymes remains a challenge of paramount *** implementation paths have already been explored,including the application of peroxidase-like nanozymes for the development of clinical diagnostics or the regulation of oxidative stress within cells via their catalase and superoxide dismutase *** review also indicates that it is essential to understand how external parameters may boost or inhibit each of these activities,as more than one of them could ***,further toxicity studies are required to ensure the applicability of gold nanozymes in *** challenges and future prospects of gold nanozymes are discussed in this review,whose significance can be anticipated in a diverse range of fields beyond biomedicine,such as food safety,environmental analyses or the chemical industry.

Effect of the Inclusion of Chestnut in the Finishing Diet on Volatile Compounds of Dry-Cured Ham from Celta pig Breed

The effect of the inclusion of chestnut in pigs ifnishing diet on volatile compounds of dry-cured Celta ham was studied. Twelve hams of each type （from three different pigs ifnishing diets：concentrate （CO）, mixed （MI） and chestnut （CH）） were used. Volatiles were extracted using a purge-and-trap method and analyzed by gas chromatography/mass spectrometry （GC/MS）. Thirty-nine volatile compounds were identified in dry-cured Celta ham samples. Most abundant volatiles in ham samples were aldehydes, which represented respectively, 53% （CO）, 51% （MI） and 46% （CH） of the total volatile composition. With the exception of 2-butenal, 2-methyl, all aldehydes were affected by feeding system. On the other hand, hydrocarbons n-alkanes were the second major group in the volatile proifle of dry-cured Celta hams and represented 28.9, 35.7 and 32.4%of the total volatile composition for CO, MI and CH groups, respectively. Ham samples from chestnut group showed a higher content of alcohols and this result could be related with the inclusion of chestnut in the ifnishing diet of pigs. principal component analysis showed a good separation among groups. The discriminant analysis selected eight variables （butanoic acid, hexanal, octanal, nonenal （E）, decenal （E）, tetradecane, decane trimethyl and pyridine 2-methyl） and calculated two discriminating functions to predict if chestnut has been included in the ifnishing diet. Thus, it was possible to discriminate between groups fed with ifnishing diets containing chestnuts in their composition （mixed and chestnut group）.

Nanozymes in point-of-Care Diagnosis:An Emerging Futuristic Approach for Biosensing

Nanomaterial-based artificial enzymes(or nanozymes) have attracted great attention in the past few years owing to their capability not only to mimic functionality but also to overcome the inherent drawbacks of the natural *** advantages of nanozymes such as diverse enzyme-mimicking activities,low cost,high stability,robustness,unique surface chemistry,and ease of surface tunability and biocompatibility have allowed their integration in a wide range of biosensing applications. Several metal,metal oxide,metal–organic framework-based nanozymes have been exploited for the development of biosensing systems,which present the potential for point-of-care analysis. To highlight recent progress in the field,in this review,more than 260 research articles are discussed systematically with suitable recent examples,elucidating the role of nanozymes to reinforce,miniaturize,and improve the performance of point-of-care diagnostics addressing the ASSURED(a ordable,sensitive,specific,user-friendly,rapid and robust,equipment-free and deliverable to the end user) criteria formulated by World Health Organization. The review reveals that many biosensing strategies such as electrochemical,colorimetric,fluorescent,and immunological sensors required to achieve the ASSURED standards can be implemented by using enzyme-mimicking activities of nanomaterials as signal producing components. However,basic system functionality is still lacking. Since the enzyme-mimicking properties of the nanomaterials are dictated by their size,shape,composition,surface charge,surface chemistry as well as external parameters such as pH or temperature,these factors play a crucial role in the design and function of nanozyme-based point-of-care diagnostics. Therefore,it requires a deliberate exertion to integrate various parameters for truly ASSURED solutions to be realized. AIMTo evaluate the effectiveness of human fibrinogen-thrombin collagen patch (TachoSilp>®p>) in the reinforcement of high-risk colon *** quasi-experimental study was conducted in Wistar rats (n = 56) that all underwent high-risk anastomoses (anastomosis with only two sutures) after colectomies. The rats were divided into two randomized groups: Control group (24 rats) and treatment group (24 rats). In the treatment group, high-risk anastomosis was reinforced with TachoSilp>®p> (a piece of TachoSilp>®p> was applied over this high-risk anastomosis, covering the gap). Leak incidence, overall survival, intra-abdominal adhesions, and histologic healing of anastomoses were analyzed. Survivors were divided into two subgroups and euthanized at 15 and 30 d after intervention in order to analyze the adhesions and histologic *** survival was 71.4% and 57.14% in the TachoSilp>®p> group and control group, respectively (p = 0.29); four rats died from other causes and six rats in the treatment group and 10 in the control group experienced colonic leakage (p > 0.05). The intra-abdominal adhesion score was similar in both groups, with no differences between subgroups. We found non-significant differences in the healing process according to the histologic score used in both groups (p = 0.066).CONCLUSIONIn our study, the use of TachoSilp>®p> was associated with a non-statistically significant reduction in the rate of leakage in high-risk anastomoses. TachoSilp>®p> has been shown to be a safe product because it does not affect the histologic healing process or increase intra-abdominal adhesions.

A Differentially Expressed Gene from a High Oil producer Cultivar of Castor Bean (<i>Ricinus communis</i>) Is Involved in the Biosynthesis of Ricinoleic Acid

Ricinus communis or castor bean is a non-edible oilseed plant widely cultivated worldwide for the high content of castor oil in its seeds and the different uses the oil has in the industry. An increase in its oil content and production efficiency is difficult, making understanding the molecular mechanisms underlying the synthesis of oils in the seed necessary. Here, a combined analysis of protein-protein interaction networks was performed using public data on differential gene expression in castor bean seeds at different stages of development. From this analysis, four key enzymes were selected and analyzed in the polyunsaturated fatty acids pathways, whose gene expression was subsequently quantified during the development of the seeds in a Colombian cultivar that produces high amounts of oils and contrasted with a lower producing cultivar. The gene coding FAH12 was differentially expressed in the early stages of seed development in the high oil-producing cultivar and has differences in amino acids A242V and Q319H. The analysis presents this gene as one of those responsible for early ricinoleic acid synthesis, making it a candidate for use in crop genetic improvement programs to increase the oil content in castor bean.

Coronary angiography findings in cardiac arrest patients with non-diagnostic post-resuscitation electrocardiogram:A comparison of shockable and non-shockable initial rhythms

AIM To investigate the impact of coronary artery disease in a cohort of patients resuscitated from cardiac arrest with non-diagnostic *** From March 2004 to February 2016, 203 consecutive patients resuscitated from in or out-of-hospital sudden cardiac arrest and non-diagnostic post-resuscitation electrocardiogram(defined as ST segment elevation or pre-sumably new left bundle branch block) whounderwent invasive coronary angiogram during hospitalization were included. For purpose of analysis and comparison, patients were classified in two groups: Initial shockable rhythm(ventricular tachycardia or ventricular fibrillation; n = 148, 72.9%) and initial non-shockable rhythm(n = 55, 27.1%). Baseline characteristics, coronary angiogram findings including Syntax Score and long-term survival rates were compared. RESULTS Sudden cardiac arrest was witnessed in 95.2% of cases, 66.7% were out-of-hospital patients and 72.4% were male. There were no significant differences in baseline characteristics between groups except for higher mean age(68.1 years vs 61 years, p = 0.001) in the nonshockable rhythm group. Overall 5-year mortality of the resuscitated patients was 37.4%. patients with non-shockable rhythms had higher mortality(60% vs 29.1%, p pital discharge based on cerebral performance category score(CpC 1-2: 32.7% vs 53.4%, p = 0.02). Although there were no significant differences in global burden of coronary artery disease defined by Syntax Score(mean Syntax Score: 10.2 vs 10.3, p = 0.96) there was a trend towards a higher incidence of acute coronary lesions in patients with shockable rhythm(29.7% vs 16.4%, p = 0.054). There was also a higher need for ad-hoc percutaneous coronary intervention in this group(21.9% vs 9.1%, p = 0.03). CONCLUSION Initial shockable group of patients had a trend towards higher incidence of acute coronary lesions and higher need of ad-hoc percutaneous intervention vs nonshockable

Determinants of percutaneous coronary intervention success in repeat chronic total occlusion procedures following an initial failed attempt

AIM To investigate the rates and determinants of success of repeat percutaneous coronary intervention(pCI) following an initial failed attempt at recanalising the chronic total occlusions(CTO) *** In 445 consecutive first attempt CTO-pCI procedures in our institution,procedural failure occurred in 149(33.5%). Sixty-four re-pCI procedures were performed in 58 patients(39%) all had a single CTO. procedural and outcome data in the re-pCI population was entered into the institutional database. A retrospective analysis of clinical,angiographic and procedural data was performed. RESULTS procedural success was achieved in 41(64%) procedures. Univariate analysis of clinical and angiographic characteristics showed that re-pCI success was associated with intravascular ultrasound(IVUS) guidance(19.5% vs 0%,p = 0.042),while failure was associated with severecalcification(30.4% vs 9.7%,p = 0.047) and a JCTO score > 3(56.5% vs 17.1% p = 0.003). Following multiple regression analysis the degree of lesion complexity(J-CTO score > 3),IVUS use,involvement of an experienced CTO operator and LAD CTO location were significant predictors of successful re-pCI. Overall the complication rate was low,with the only MACCE two periprocedural MI's neither of which required intervention. CONCLUSION Re-pCI substantially increases the overall success rate of CTO revascularization. predictors of re-pCI success included the use of IVUS,the involvement of an experienced CTO operator in the repeat attempt and the location of the CTO.

Severe Acute Respiratory Syndrome Coronavirus(SARS-CoV)-2 infection induces an exacerbated inflammation driven by innate immunity *** cells(DCs)play a key role in the defense against viral infections,for instance plasmacytoid DCs(pDCs),have the capacity to produce vast amounts of interferon-alpha(IFN-α).In COVID-19 there is a deficit in DC numbers and IFN-αproduction,which has been associated with disease *** this work,we described that in addition to the DC deficiency,several DC activation and homing markers were altered in acute COVID-19 patients,which were associated with multiple inflammatory ***,previously hospitalized and nonhospitalized patients remained with decreased numbers of CD1c+myeloid DCs and pDCs seven months after SARS-CoV-2 ***,the expression of DC markers such as CD86 and CD4 were only restored in previously nonhospitalized patients,while no restoration of integrinβ7 and indoleamine 2,3-dyoxigenase(IDO)levels were *** findings contribute to a better understanding of the immunological sequelae of COVID-19.

Searching for the neutrinoless double beta decay(NLDBD)is now regarded as the topmost promising technique to explore the nature of neutrinos after the discovery of neutrino masses in oscillation *** X-III(particle and astrophysical xenon experiment III)will search for the NLDBD of136Xe at the China Jin ping Underground Laboratory(CJpL).In the first phase of the experiment,a high pressure gas Time projection Chamber(TpC)will contain 200 kg,90%136Xe enriched gas operated at10 *** pitch micro-pattern gas detector(Microbulk Micromegas)will be used at both ends of the TpC for the charge readout with a cathode in the *** signals can be used to reconstruct the electron tracks of the NLDBD events and provide good energy and spatial *** detector will be immersed in a large water tank to ensure～5 m of water shielding in all *** second phase,a ton-scale experiment,will consist of five TpCs in the same water tank,with improved energy resolution and better control over backgrounds.
