Angiotensin II (Ang II) is the main mediator of the Renin-Angiotensin-System acting on AT1 and other AT receptors. It is regarded as a pleiotropic agent that induces many actions, including functioning as a growth factor, and as a contractile hormone, among others. The aim of this work was to examine the impact of Ang II on the expression and function of α1-adrenergic receptors (α1-ARs) in cultured rat aorta, and aorta-derived smooth muscle cells. Isolated Wistar rat aorta was incubated for 24 h in DMEM at 37˚C, then subjected to isometric tension and to the action of added norepinephrine, in concentration-response curves. Ang II was added (1 × 10−5 M), and in some experiments, 5-Methylurapidil (α1A-AR antagonist), AH11110A (α1B-AR antagonist), or BMY-7378 (α1D-AR antagonist), were used to identify the α1-AR involved in the response. Desensitization of the contractile response to norepinephrine was observed due to incubation time, and by the Ang II action. α1D-AR was protected from desensitization by BMY-7378;while RS-100329 and prazosin partially mitigated desensitization. In another set of experiments, isolated aorta-derived smooth muscle cells were exposed to Ang II and α1-ARs proteins were evaluated. α1D-AR increased at 30 and 60 min post Ang II exposure, the α1A-AR diminished from 1 to 4 h, while α1B-AR remained unchanged over 24 h of Ang II exposure. Ang II induced an increase of α1D-AR at short times, and BMY-7378 protected α1D-AR from desensitization.

AIM:To evaluate the z-line visualization by the PillCamTM SB2 using three different ingestion protocols. METHODS:Ninety consecutive patients undergoing small bowel capsule endoscopy(SBCE)between January and May 2008 were included in the study. They swallowed the capsule in the standing(group A= 30),supine(group B=30)and right supine positions (group C=30).Baseline patient characteristics, difficulties in capsule ingestion,esophageal transit times(ETT)and z-line visualization were noted. RESULTS:No significant differences were found between the groups with regard to baseline patient characteristics,ingestion difficulties and complete SB examinations(P>0.05).At least 1 frame of the z-line was detected in 15.8%,46.7%and 90%of patients in groups A,B and C,respectively(Pge number of z-line images was 0.21±0.53,3.23 ±6.59 and 5.53±7.55 and the mean%of the z-linedetected was 71.3,25.1 and 8.3,in groups A,B and C, respectively(both Pger in the supine group followed by the right supine and the standing groups(median of 237 s vs 64 s and 39 s, respectively;Pz-line visualization in patients undergoing SBCE can be accurately achieved in most cases when the capsule is swallowed in the right supine position.

Hepatitis C virus NS5A region mutation in chronic hepatitis C genotype 1 patients who are non-responders to two or more treatments and its relationship with response to a new treatment

To determine the number of mutations in the NS5A region of the hepatitis C virus (HCV) and its relationship to the response to antiviral therapy in patients with chronic hepatitis C genotype 1 who are non-responders to two or more treatments. METHODSSequences within HCV NS5A [PKR binding domain (PKRBD) and the interferon-sensitivity-determining region (ISDR)] were analysed via direct sequencing in a selected cohort of 72 patients, with a total of 201 treatments [interferon-alpha (IFN-α), n = 49; IFN-α + ribavirin (RBV), n = 75; pegylated (peg) IFN-α + RBV, n = 47; first-generation direct-acting antivirals (DAAs), n = 13; and second-generation DAAs, n = 17]. Of these, 48/201 achieved a sustained virological response (SVR) and 153/201 achieved no virological response (NVR). RESULTSFor both regions, treatments resulting in SVR were associated with more baseline mutations than were treatments resulting in NVR (SVR vs NVR; PKRBD: 5.82 ± 3 vs 4.86 ± 2 mutations, P = 0.045; ISDR: 2.65 ± 2 vs 1.51 ± 1.7 mutations, P = 0.005). A decrease or no change in the number of mutations over time between treatments in the PKRBD or ISDR, as shown by sequencing, was associated with patients who usually failed to respond to treatment (PKRBD, P = 0.02; ISDR, P = 0.001). Moreover, patients showing a post-treatment baseline viral load > 600000 IU/mL and increased ISDR mutations with respect to the previous treatment were 9.21 times more likely to achieve SVR (P = 0.001). CONCLUSIONThe obtained results show that among patients who have shown no response to two or more antiviral treatments, the likelihood of achieving SVR increases with the genetic variability in the ISDR region (≥ 2 mutations or number of substitutions from the HCV-J and HCV-1 prototype), especially when the viral load is greater than 600000 IU/mL.

Desensitization is a process characterized by the loss of cellular response to an agonist when this is present for a long time. α1D-adrenergic receptor (α1D-AR) desensitization is important since this receptor is involved in the contraction of large caliber arteries, such as the aorta. The aim of this research was to evaluate the desensitization of α1D-AR due to the endogenous release of norepinephrine in cultured rat aorta. Wistar rat aorta was incubated for 2 h or 24 h in DMEM at 37°C, and then subjected to isometric tension and the action of added norepinephrine, in concentration-response curve (CRC). In some experiments, BMY-7378 (α1D-AR antagonist) or 5-methylurapidil (α1A-AR antagonist) was used to identify the α1-AR involved in the response, or BMY-7378 to protect the α1D-AR from desensitization. Results showed that α1D-AR was desensitized when the aorta was incubated for 24 h, since the CRC to exogenous norepinephrine showed lower maximal contraction and the curve was displaced to the right, indicating that the receptor involved in contraction was not the α1D-AR, as compared to the aorta incubated 2 h. The receptor stimulated by norepinephrine at 24 h was neither the α1A-AR, as shown by the lack of displacement of the curve by 5-methylurapidil, but rather it seems that α1B-AR is inducing contraction. When the aorta was incubated with BMY-7378 for 24 h, the α1D-AR antagonist protected the receptor from desensitization. Endogenous norepinephrine desensitizes α1D-AR in the cultured aorta, and the α1D-AR is protected by BMY-7378.

As climate change negotiations progress,monitoring biomass and carbon stocks is becoming an important part of the current forest ***,national governments are interested in developing forest-monitoring strategies using geospatial *** statistical methods for mapping biomass,there is a nonparametric approach called k-nearest neighbor(kNN).We compared four variations of distance metrics of the kNN for the spatially-explicit estimation of aboveground biomass in a portion of the Mexican north border of the intertropical *** derived,climatic,and topographic predictor variables were combined with the Mexican National Forest Inventory(NFI)data to accomplish the *** of distance metrics applied into the kNN algorithm was evaluated using a cross validation leave-one-out *** results indicate that the Most Similar Neighbor(MSN)approach maximizes the correlation between predictor and response variables(r=0.9).Our results are in agreement with those reported in the *** findings confirm the predictive potential of the MSN approach for mapping forest variables at pixel level under the policy of Reducing Emission from Deforestation and Forest Degradation(REDD+).

Two-dimensional transition metal dichalcogenide semiconductors have emerged as promising candidates for optoelectronic devices with unprecedented properties and ultra-compact footprints. However, the high sensitivity of atomically thin materials to the surrounding dielectric media imposes severe limitations on their practical applicability. Hence, to enable the effective integration of these materials in devices, the development of reliable encapsulation procedures that preserve their physical properties is required. Here, the excitonic photoluminescence （at room temperature and 10 K） is assessed on mechanically exfoliated WSe2 monolayer flakes encapsulated with SiOx and AlxOy layers by means of chemical and physical deposition techniques. Conformal coating on untreated and non- functionalized flakes is successfully achieved by all the techniques examined, with the exception of atomic layer deposition, for which a cluster-like oxide coating is formed. No significant compositional or strain state changes in the flakes are detected upon encapsulation, independently of the technique adopted. Remarkably, our results show that the optical emission of the flakes is strongly influenced by the stoichiometry quality of the encapsulating oxide. When the encapsulation is carried out with slightly sub-stoichiometric oxides, two remarkable phenomena are observed. First, dominant trion （charged exciton） photoluminescence is detected at room temperature, revealing a clear electrical doping of the monolayers. Second, a strong decrease in the optical emission of the monolayers is observed, and attributed to non-radiative recombination processes and/or carrier transfer from the flake to the oxide. Power- and temperature-dependent photoluminescence measurements further confirm that stoichiometric oxides obtained by physical deposition lead to a successful encapsulation, opening a promising route for the development of integrated two-dimensional devices.

Background Advances in cancer treatments,particularly the development of radiation therapy,have led to improvements in survival outcomes in children with brain ***,radiation therapy is associated with significant long-term neurocognitive *** present systematic review and meta-analysis aimed to compare the neurocognitive outcomes of children and adolescents with brain tumors treated with photon radiation(XRT)or proton therapy(PBRT).Methods A systematic search was conducted(PubMed,Embase,Cochrane,and Web of Science from inception until 02/01/2022)for studies comparing the neurocognitive outcomes of children and adolescents with brain tumors treated with XRT *** pooled mean differences(expressed as z scores)were calculated using a random effects method for those endpoints analyzed by a minimum of three *** Totally 10 studies(n=630 patients,average age range:1–20 years)met the inclusion *** who had received PBRT achieved significantly higher scores(difference in z scores ranging from 0.29–0.75,all Pgnificant in sensitivity analyses)after treatment than those who had received XRT for most analyzed neurocognitive outcomes(i.e.,intelligence quotient,verbal comprehension and perceptual reasoning indices,visual motor integration,and verbal memory).No robust significant differences(P>0.05 in main analyses or sensitivity analyses)were found for nonverbal memory,verbal working memory and working memory index,processing speed index,or focused *** Pediatric brain tumor patients who receive PBRT achieve significantly higher scores on most neurocognitive outcomes than those who receive *** studies with long-term follow-ups are needed to confirm these results.

Purpose:This study was aimed to analyze the mediation role of cardiorespiratory fitness(CRF)on the association between fatness and cardiometabolic risk scores(CMRs)in European ***:A cross-sectional study was conducted in adolescents(n=525;46%boys;14.1±1.1 years old,mean ±SD)from 10 European cities involved in the Healthy Lifestyle in Europe by Nutrition in Adolescence *** was measured by means of the shuttle run test,while fatness measures included body mass index(BMI),waist to height ratio,and fat mass index estimated from skinfold thicknesses.A clustered CMRs was computed by summing the standardized values of homeostasis model assessment,systolic blood pressure,triglycerides,total cholesterol/highdensity lipoprotein cholesterol ratio,and ***:Linear regression models indicated that CRF acted as an important and partial mediator in the association between fatness and CMRs in 1217-year-old adolescents(for BMI:coefficients of the indirect role b=0.058(95%confidence interval(95%CI):0.023-0.101),Sobel test z=3.11(10.0%mediation);for waist to height ratio:b=4.279(95%CI:2.242-7.059),z=3.86(11.5%mediation);and for fat mass index:b=0.060(95%CI:0.020-0.106),z=2.85(9.4%mediation);all p<0.01).Conclusion:In adolescents,the association between fatness and CMRs could be partially decreased with improvements to fitness levels;therefore,CRF contribution both in the clinical field and public health could be important to consider and promote in adolescents independently of their fatness levels.

Background:The economic impact of schistosomiasis and the underlying tradeofs between water resources devel‑opment and public health concerns have yet to be *** exerts large health,social and fnan‑cial burdens on infected individuals and *** irrigation schemes are one of the most important policy responses designed to reduce poverty,particularly in sub-Saharan Africa,they facilitate the propagation of schistoso‑miasis and other ***:We estimate the economic impact of schistosomiasis in Burkina Faso via its efect on agricultural produc‑*** create an original dataset that combines detailed household and agricultural surveys with high-resolution geo-statistical disease *** develop new methods that use the densities of the intermediate host snails of schis‑tosomiasis as instrumental variables together with panel,spatial and machine learning ***:We estimate that the elimination of schistosomiasis in Burkina Faso would increase average crop yields by around 7%,rising to 32%for high infection *** schistosomiasis unchecked,in turn,would correspond to a loss of gross domestic product of approximately 0.8%.We identify the disease burden as a shock to the agricultural productivity of *** poorest households engaged in subsistence agriculture bear a far heavier disease burden than their wealthier counterparts,experiencing an average yield loss due to schistosomiasis of between 32 and 45%.We show that the returns to water resources development are substantially reduced once its health efects are taken into account:villages in proximity of large-scale dams sufer an average yield loss of around 20%,and this burden decreases as distance between dams and villages ***:This study provides a rigorous estimation of how schistosomiasis afects agricultural production and how it is both a driver and a consequence of *** further quantifes the tradeof between the economics of wa
