Engineering of Yarrowia lipolytica for production of astaxanthin

Astaxanthin is a red-colored carotenoid,used as food and feed *** is mainly produced by chemical synthesis,however,the process is expensive and synthetic astaxanthin is not approved for human *** this study,we engineered the oleaginous yeast Yarrowia lipolytica for de novo production of astaxanthin by ***,we screened 12 different *** isolates for β-carotene production by introducing two genes for β-carotene biosynthesis:bi-functional phytoene synthase/lycopene cyclase(crtYB)and phytoene desaturase(crtI)from the red yeast Xanthophyllomyces *** best strain produced 31.1±0.5 mg/L β-***,we optimized the activities of 3-hydroxy-3-methylglutaryl-coenzyme A reductase(HMG1)and geranylgeranyl diphosphate synthase(GGS1/crtE)in the best producing strain and obtained 453.9±20.2 mg/L β-*** downregulation of the competing squalene synthase SQS1 increased the β-carotene titer to 797.1±57.2 mg/*** we introduced β-carotene ketolase(crtW)from Paracoccus sp.N81106 and hydroxylase(crtZ)from Pantoea ananatis to convert β-carotene into *** constructed strain accumulated 10.4±0.5 mg/L of astaxanthin but also accumulated astaxanthin biosynthesis intermediates,5.7±0.5 mg/L canthaxanthin,and 35.3±1.8 mg/L ***,we optimized the copy numbers of crtZ and crtW to obtain 3.5 mg/g DCW(54.6 mg/L)of astaxanthin in a microtiter plate *** study for the first time reports engineering of *** for the production of *** high astaxanthin content and titer obtained even in a small-scale cultivation demonstrates a strong potential for ***-based fermentation process for astaxanthin production.

Hurdles in achieving the goal of malaria elimination by India

India is a major contributor to the malaria burden in Southeast Asia, with 83% estimated cases and 82% estimated deaths reported in 2020[1]. As per the National Center for Vector-Borne Disease Control(NCVBDC) estimates, around 0.19 million cases and 93 deaths occurred in the country due to malaria in 2020. Since 2002, India has made impressive progress in malaria control with an approximately 90% reduction in malaria cases over the last two decades and is on the road to malaria elimination by 2030.

A coprecipitation method was used to synthesize superparamagnetic CoFe2O4 nanoparticles without using any capping agents/surfactants. The prepared nanoparticles were characterized using Fourier transform infrared spectroscopy, scanning electron microscopy, transmission electron microscopy, X-ray diffraction, a vibrating sample magnetometer (VSM), N2 adsorption and thermogravimetric/differential thermal analysis/differential thermal gravimetry techniques. The synthesized spinel CoFe2O4 nanoparticles had an average size of 2-8 nm with a high surface area (140.9 m2/g). The field-dependent magnetization, demonstrated by VSM and saturation magnetization, was found to be 1.77 emu/g. An efficient method was used for the synthesis of arylidene barbituric acid derivatives using CoFe2O4 magnetic nanoparticles as a magnetically separable and reusable catalyst in aqueous ethanol. The attractive features of this synthetic protocol were very short reaction time, high yields, high turnover frequency, simple work-up procedure, economy, a clean reaction methodology, and chemoselectivity, as well as provision of an ecofriendly and green synthesis.

Thermal decomposition and kinetics of plastic bonded explosives based on mixture of HMX and TATB with polymer matrices

This work describes thermal decomposition behaviour of plastic bonded explosives(PBXs) based on mixture of 1,3,5,7-tetranitro-1,3,5,7-tetrazocane(HMX) and 2,4,6-triamino-1,3,5-trinitrobenzene(TATB)with Viton A as polymer binder. Thermal decomposition of PBXs was undertaken by applying simultaneous thermal analysis(STA) and differential scanning calorimetry(DSC) to investigate influence of the HMX amount on thermal behavior and its kinetics. Thermogravimetric analysis(TGA) indicated that the thermal decomposition of PBXs based on mixture of HMX and TATB was occurred in a three-steps. The first step was mainly due to decomposition of HMX. The second step was ascribed due to decomposition of TATB, while the third step was occurred due to decomposition of the polymer matrices. The thermal decomposition % was increased with increasing HMX amount. The kinetics related to thermal decomposition were investigated under non-isothermal for a single heating rate measurement. The variation in the activation energy of PBXs based on mixture of HMX and TATB was observed with varying the HMX amount. The kinetics from the results of TGA data at various heating rates under non-isothermal conditions were also calculated by Flynn—Wall—Ozawa(FWO) and Kissinger-Akahira-Sunose(KAS)methods. The activation energies calculated by employing FWO method were very close to those obtained by KAS method. The mean activation energy calculated by FWO and KAS methods was also a good agreement with the activation energy obtained from single heating rate measurement in the first step decomposition.

Analysis of Geo Positioning System Using RF &DA

Indoor positioning systems (IPSs) have been intended to provide position information of persons and devices. Higher user percentage of handheld devices such as tablets or mobile phones had led to the development of a number of indoor positioning systems. In this research a work on a real time portable RFID indoor positioning device such as on smartphone will be performed. The personal networks will be designed to meet the users’ needs and interconnect users’ devices equipped with different communications technologies in various places to form one network for better result. Radio frequency identification (RFID) with directional antenna has proved its potential for locating objects in indoor environment. Hence, the proposed device idea will be used to exploit various unknown locations in an indoor environment such as college campus;this interpretation will rely on Wireless LAN, Received Signal Strength values from Access Points (AP) in specific mentioned arenas;these APs will be monitored constantly by RFID with directional antenna (DA) and handheld devices. For obtaining the better results from existing devices, algorithms of Range Estimation are proposed, which can be used on various handheld devices for locating indoor objects.
