Environmental stresses profoundly altered accumulation of nonsense mRNAs including intron-retaining （IR） transcripts in Arabidopsis. Temporal patterns of stress-induced IR mRNAs were dissected using both oscillating and non-oscillating transcripts. Broad-range thermal cycles triggered a sharp increase in the long IR ccA1 isoforms and altered their phasing to different times of day. Both abiotic and biotic stresses such as drought or Pseudomonas syringae infection induced a similar increase. Thermal stress induced a time delay in accumulation of ccA1 14Rb transcripts, whereas functional mRNA showed steady oscillations. Our data favor a hypothesis that stress-induced instabilities of the central oscillator can be in part compensated through fluctuations in abundance and out-of-phase oscillations of ccA1 IR transcripts. Taken together, our results support a concept that mRNA abundance can be modulated through altering ratios between functional and nonsense/IR transcripts. SR45 protein specifically bound to the retained ccA1 intron in vitro, suggesting that this sp!icing factor could be involved in regulation of intron retention. Transcriptomes of nonsense-mediated mRNA decay （NMD）-impaired and heat-stressed plants shared a set of retained introns associated with stress- and defense-inducible transcripts. constitutive activation of certain stress response networks in an NMD mutant could be linked to disequilibrium between functional and nonsense mRNAs.

Novel cinobufagin Oxime Ether Derivatives as Potential Na＋/K＋-ATPase Inhibitors： Synthesis, Biological Screening and Molecular Docking

Some cinobufagin oxime ether derivatives as potential Na＋/K＋-ATPase inkibitors were synthesized by following the side chain of istaroxime. These compounds inhibit Na＋/K＋-ATPase in a dose-dependent manner. compound 3c with an oxyethylamine side chain that is the same as that of istaroxime showed the most potent inhibi- tion, which was stronger than compound 3a with only hydroxyoxime moiety at c3 and compound 3b with a methy-lated hydroxyoxime moiety. Molecular docking was used to explore the binding modes of the target compounds with Na＋/K＋-ATPase, which suggested that the longer ethyl amine group at c3 oxime moiety of compound 3c could make stronger interaction with Na＋/K＋-ATPase via intermolecular charge-charge and H-bond interaction as compared with other derivatives.

Objective:To evaluate medical student and attending surgeon experiences with a novel interactive virtual Otolaryngology-Head and Neck Surgery(OHNS)medical student elective during the cOVID-19 *** Design:A virtual OHNS elective was created,with three components:(1)interactive virtual operating room(OR)experience using live-stream video-conferencing,(2)telehealth clinic,(3)virtual ***:OHNS Department at the University of Pennsylvania(May 2020 to June 2020).Methods:Six medical students from the University of Pennsylvania;five attending *** surveys were designed and distributed to participating medical students and attending *** included 5-point Likert scale items,with 1 indicating"not at all"and 5 indicating"very much so".Results:Response rate was 100%for both *** on average rated the educational value of the telehealth experience as 4.2±1.2,and the virtual OR experience as 4.0±*** students(n=5,83%)indicated that they had enough exposure to faculty they met on this rotation to ask for a letter of recommendation(LOR)for residency if needed,while attending surgeons had an average response of 3.0±1.0 when asked how comfortable they would feel writing a LOR for a student they met through the rotation.A majority of students(n=4,67%)felt they connected enough with faculty during the rotation to ask for *** the students(n=5,50%)indicated that the rotation allowed them to evaluate the department's culture either"extremely well"or"somewhat well".conclusions:Overall,participating students described this innovative virtual surgical rotation as an educationally and professionally valuable *** the continued suspension of visiting student rotations due to the cOVID-19 pandemic,this virtual model may have continued relevance to medical education.

Reptile sex determination is attracting much attention because the great diversity of sex-determination and dosage compensation mechanisms permits us to approach fundamental questions about mechanisms of sex chromosome turnover. Recent studies have made significant progress in better understanding diversity and conservation of reptile sex chromosomes, with however no reptile master sex determination genes identified. Here we describe an integrated genomics and cytogenetics pipeline,combining probes generated from the microdissected sex chromosomes with transcriptome and genome sequencing to explore the sex chromosome diversity in non-model Australian reptiles. We tested our pipeline on a turtle, two species of geckos, and a monitor lizard. genes identified on sex chromosomes were compared to the chicken genome to identify homologous regions among the four species. We identified candidate sex determining genes within these regions, including conserved vertebrate sex-determining genes pdgfa,pdgfra amh and wt1, and demonstrated their testis or ovary-specific expression. All four species showed gene-by-gene rather than chromosome-wide dosage compensation. Our results imply that reptile sex chromosomes originated by independent acquisition of sex-determining genes on different autosomes,as well as translocations between different ancestral macro-and *** discuss the evolutionary drivers of the slow differentiation and turnover of reptile sex chromosomes.

It is estimated that 80% of the world population depends on traditional medicine for primary healthcare need. Trianthema portulacastrum Linn.（family： Aizoaceae） is a small perennial weed found in the Americas, Africa, India, and other regions of the world. This plant is used extensively in Indian traditional medicines and is also consumed as a vegetable throughout Asia for its perceived health benefits. Phytochemical analysis of T. portulacastrum reveals the presence of alkaloids, fl avonoids, terpenoids, saponins, and phenolic compounds. Emerging studies demonstrate that crude extracts as well as bioactive phytoconstituents of T. portulacastrum exhibit potent antioxidant, anti-infective, analgesic, and anti-infl ammatory activities. A growing number of in vitro and in vivo studies demonstrate various biological and pharmacological activities, including prevention and amelioration of hepatotoxicity, nephrotoxicity, hyperglycemia, hyperlipidemia, infectious diseases and cancer. This review aims to present and analyze available literature to understand the full potential of T. portulacastrum in health promotion and disease prevention. current limitations and future directions of research on this medicinal and dietary plant are also critically discussed.

On the Qinghai-Tibet Plateau,extreme environmental conditions have imposed intense selective pressure on the evolution of phenotypic traits of wild *** date,limited information is available on behavioral and ecological traits concerning niche differentiation among sympatric animals on the Qinghai-Tibet Plateau,especially during winter when the environments are most ***,we studied the seasonal variations in habitat occurrence,territorial behavior,and diet in two sympatric snow finches(the white-rumped snow finch,Onychostruthus taczanowskii,WRSF;and the rufous-necked snow finch,Pyrgilauda ruficollis,RNSF)to determine convergence and divergence of ecological traits in such severe climatic *** results showed that:(i)WRSF occupied rural areas as a dominant species throughout the annual cycle while RNSF occupied the rural areas in summer and then shifted to human-occupied areas in winter and spring;(ii)WRSFs exhibited robust aggressive behavior and territoriality during winter relative to RNSFs;(iii)the diets of both species varied with the season but did not vary between species except that WRSF ate significantly more seeds but RNSF consumed more starchy material derived from human food waste during ***,the separations in the spatial niche and territoriality between WRSF and RNSF,especially in winter,may contribute to alleviating the pressure of interspecific competition,and promoting the coexistence of the two sympatric snow finches in the extreme environments on the Qinghai-Tibet Plateau.
