MicroRNAs（miRNAs） play an important role as regulators of growth and development in *** miRNA families target genes encoding disease resistance proteins,such as nucleotide binding site-leucine-rich repeat（NBS-LRR） plant innate immune *** filamentous fungus fusarium oxysporum *** causes vascular wilt disease in susceptible tomato（Solarium lycopersicum） *** explored a possible role for miRNAs in tomato defense against *** using deep sequencing of small RNA *** miRNA profiling of sensitive（Moneymaker） and resistant（Motelle） tomato cultivars infected with *** revealed several miRNAs that correlated with disease *** particular,slmiR482 f and slmiR5300,two members of the miR482/2118 microRNA superfamily,were repressed during infection with *** analysis confirmed that four of the predicted targets of these two miRNAs exhibited increased expression in the resistant Motelle cultivar and encode nucleotide-binding site（NBS） domain-containing ***,the ability of two predicted mRNA targets of slmiR482f（Solyc08g075630 and Solyc08g076000） and two targets of slmiR5300（Solyc05g008650and Solyc09g018220） to be regulated by the miRNAs was confirmed by Agrobacterium-mediated transient co-expression in Nicotiana *** results also revealed evidence for mRNA cleavage and/or translational inhibition mechanisms during regulation of target gene expression by the microRNAs.A TRV-based virus-induced gene silencing approach in the resistant Motelle cultivar revealed a clear role in resistance to *** for one target of slmiR482f（Solyc08g075630;fusarium Resistance Protein-1;fRP-1） and two targets of slmiR5300（Solyc05g008650;fRP-2 and Solyc09g018220;fRP-3）.To our knowledge,this is the first demonstration of microRNAs that mediate resistance to *** in tomato and first evidence of a role for miR5300 in pathogen resistance in any plant system.

Background:Liver resection and local ablation are the only curative treatment for non-cirrhotic hepatocellular carcinoma(HCC).few data exist concerning the prognosis of patients resected for non-cirrhotic *** objectives of this study were to determine the prognostic factors of recurrence-free survival(RfS)and overall survival(OS)and to develop a prognostication algorithm for non-cirrhotic ***:french multicenter retrospective study including HCC patients with non-cirrhotic liver without underlying viral hepatitis:f0,f1 or f2 ***:A total of 467 patients were included in 11 centers from 2010 to ***-cirrhotic liver had a fibrosis score of f0(n=237,50.7%),f1(n=127,27.2%)or f2(n=103,22.1%).OS and RfS at 5 years were 59.2%and 34.5%,*** multivariate analysis,microvascular invasion and HCC differentiation were prognostic factors of OS and RfS and the number and size were prognostic factors of RfS(Pfication based on RfS provided an algorithm based on size(P=0.013)and number(P10 cm(n=176,Group 2);>2 HCC regardless of size Conclusions:We developed a prognostication algorithm based on the number(≤or>2)and size(≤or>10 cm),which could be used as a treatment decision support concerning the need for perioperative *** case of bifocal HCC,surgery should not be a contraindication.

Shade caused by trees or buildings can affect the quality and growth of ***,breeding for shade tolerance is an essential component of most turfgrass improvement *** objective of this study was to evaluate the performance of seashore paspalum(Paspalum vaginatum Sw.)breeding lines under shade in multi-environment *** sources were 20 lines from the University of Georgia seashore paspalum breeding program and two checks,the cultivars'SeaDwarf'and'SeaStar'.field trials were conducted from 2016 to 2019 under shade structures designed to reduce ambient sunlight at three locations:Citra(fL),Tifton(GA),and Raleigh(NC).The response variables evaluated were percent living ground cover(%GC),dark green color index(DGCI),canopy height(CH)and turfgrass quality(TQ).Data were analyzed by mixed model approaches using ASReml-R,and the t-statistics were used to group the *** genetic variances were observed in the single-location-repeated-measures analysis for CH and DGCI at Citra and Tifton,%GC at Citra,and TQ at *** correlations of the predicted values for entries between locations ranged from low to moderate,with the exception of between Citra and Tifton(0.76)for *** were a few breeding lines with superior performance to the entry average for all traits for which the entry variance was *** conclusion,genetic variability was observed for all traits and superior seashore paspalum breeding lines for multiple traits were identified under shade.

Natural antibodies serve as the body’s first line of defense against pneumococcal challenge. Polyreactive human pneumococcal polysaccharide IgG antibodies have not been extensively studied. We analyzed human polyreactive antibodies that bind multiple pneumococcal polysaccharides, including PPS14 and PPS23f. These antibodies were isolated from single pneumococcal polysaccharide specific B cells allowing for the analysis of human immunoglobulins with natively paired variable regions. Although isolated individually, these antibodies demonstrated similar characteristics. Most antibodies possessed a variable light chain with a CDR3 length made up of nine amino acids and relatively high number of flexible amino acids in combined VH/VL. While these antibodies were polyreactive and structurally alike, kinetic analysis revealed unique KD values. Variable chains are responsible for antigen recognition whereas antibody fine specificity is affected by isotype structure. To investigate the contribution of the constant region of these isotypes and their effect on antibody avidity to pneumococcal polysaccharide, the polyreactive variable regions were expressed as IgG1 or IgG2 and subjected to kinetic analysis. The IgG1 antibodies uniformly had a stronger avidity to PPS14 and PPS23f compared to IgG2. To further document the importance of the constant region in antibody avidity and fine specificity, analysis of antibody f(ab)’2 fragment binding to PPS14 and PPS23f resulted in similar KD values. These studies suggest that antigen recognition by polyreactive antibodies is determined by a conserved variable light chain CDR3 length and longer, more flexible variable heavy CDR3s when compared to pneumococcal polysaccha-ride-specific sequences while differences in specific avidities are modulated by antibody isotype.
