Dear editor,Prostate cancer(Pca)remains a major healthcare burden in men globally[1].Most patients present with localized disease,and treatment is recommended based on risk classification systems like the National comprehensive cancer Network(NccN)[2].However,these methods are imprecise for estimating metastasis-free survival and prostate cancer-specific mortality and thus biomarkers that can predict tumor aggression are needed[3–5].Several studies have since characterized the molecular landscape of localized Pca in White[4,5]and Black/African-American men[6],but data is lacking in Asian *** chinese Prostate cancer Genome and Epigenome Atlas(cPGEA)reported on the genomic and epigenomic landscape of 208 Pca of men from china[7].comparative analyses between the cPGEA cohort and data from The cancer Genome Atlas(TcGA)revealed higher frequencies of Forkhead box A1(FOXA1)and chromodomain-helicase DNA-binding 1(cHD1)mutations,and lower frequencies of phosphatase and tensin homolog(PTEN)mutations and transmembrane protease serine 2-E26 transformationspecific related gene(TMPRSS2-ERG)fusion in chinese compared with White men[7].These preliminary findings highlight the presence of race-specific differences in molecular phenotypes of Pca.

The Difference in calcium Levels in Aspergillus nidulans Grown on Glucose or Pectin

Understanding the growth regulatory mechanisms in filamentous fungi is very important for the production of medicines for antifungal therapies. It is well established that ca2+ gradient is essential for hyphal growth and that one mechanism responsible for the ca2+ cellular concentration starts with the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) by receptor-regulated forms of phosphoinositide-specific phospholipase c (PI-PLc). In the present study the levels of calcium in Aspergillus nidulans wild type (A26) and plcA-deficient mutant (AP27) growing in a carbon source readily assimilated, as glucose or pectin a non-readily assimilated carbon source was investigated. Intracellular calcium levels in A26 were higher in the presence of glucose than in pectin, but lower in AP27 independently of the carbon source and in AP27 the vesicular calcium distribution occurred mainly at the apex of the hyphae. Delay in nuclear division was also observed if A26 and AP27 were grown in pectin presence when compared with growth in glucose. For the first time, it is demonstrated that the levels of intracellular ca2+ were higher when A. nidulans was growing in glucose than in a non readily assimilated carbon source as pectin. Further, it also showed that the plcA gene, although not essential, may be responsible for high-molecular weight carbon source recongnation, for the intracellular ca2+ levels maintenance and consequently by the nuclear division in A. nidulans.

＜正＞While many books and papers discuss the development of manufacturing from an American perspective, little discussion exists from a canadian perspective. This paper examines the factors that influenced developments in industry and manufacturing in canada from the seventeenth to the twentieth century. It also discusses the lessons that may be learnt from examining the approaches of canadian manufacturers. Although canada’s abundance of natural resources led to the development of primary industries in the seventeenth and eighteenth centuries, the manufacturing industry was not significant until the early nineteenth century. Of these, four representative manufacturing industries are discussed to illustrate the overall trend in the chronological evolution of canadian manufacturing in the nineteenth and twentieth centuries. The role and impact of factors such as transportation, electricity, and foreign investment, particularly by U.S. entrepreneurs, is reviewed to understand their impact on manufacturing as it has evolved to the present. Porter’s 1991 study and its follow up about canadian competitiveness are also analysed. Finally the lessons that other countries may learn from the canadian experience are also discussed.

Poly-I:c Decreases Dendritic cell Viability Independent of PKR Activation

Vaccination with tumor-antigen pulsed, monocyte-derived dendritic cells (Dcs) has emerged as a promising strategy in cancer immunotherapy. The standard Dc maturation cocktail consists of a combination of tumor necrosis factor-α (TNF-α)/interleukin (IL)-1β/IL-6 and prostaglandin E2 (PGE2) for generation of standard Dcs (sDcs). In order to improve IL-12p70 production and cytotoxic T-lymphocyte (cTL) induction, a novel cocktail composed of TNF-α/IL-1β/ interferon (IFN)-α/IFN-γ and polyinosinic:polycytidylic acid (Poly-I:c) has been introduced to generate so-called α-Type-1 polarized Dcs (αDc1s). We and others have previously performed a comprehensive comparison of sDcs and αDc1s. Here we demonstrate that the viability of αDc1s is lowered compared to sDcs and that Dc apoptosis is mediated by Poly-I:c. We speculated that activation of protein kinase R (PKR) could mediate the observed apoptosis, but despite significantly higher PKR expression in αDc1s compared to sDcs and induction of active threonine (Thr)446 autophosphorylation of PKR in αDc1s, Poly-I:c did not influence total PKR expression or autophosporylation, indicating PKR-independent Poly-I:c-induced Dc apoptosis.

Objective: Prostate cancers(Pca) in Asian individuals are molecularly distinct from those found in their caucasian *** is no risk stratification tool for Asian men with rapid biochemical recurrence(BcR) following radical prostatectomy(Rad P). This study aims to assess the detection rate of ^(68)Ga-prostate-specific membrane antigen-positron emission tomography/computed tomography(PSMA-PET/cT) for diagnosis of clinical recurrence and as a treatment decision making tool in Asian patients with BcR post-Rad ***: ^(68)Ga PSMA-PET and cT body with/without bone scan [conventional workup(cWU)] were performed in 55 Asian patients with BcR within 36 months post-Rad P. Two blinded reviewers assessed the images. Detection rates of ^(68)Ga PSMAPET/cT were evaluated, and impact on management was reviewed by comparison with ***: Median time to BcR post-Rad P was 8.1 months. Detection rate for ^(68)Ga PSMA-PET/cT was 80%(44/55). A positive scan was significantly associated with increasing prostate-specific antigen(PSA) level [odds ratio(OR) = 1.13(95% cI 1.05–1.30), P =0.017], but not with higher Gleason grade or shorter PSA doubling time. compared to cWU, ^(68)Ga PSMA-PET/cT detected an additional 106 lesions in 33/44 patients with a positive scan, resulting in a change in management in 25/44(56.8%) patients: 10 to hormonal therapy(HT) and whole pelvis radiotherapy(RT) in addition to bed RT, and 15 to palliative HT ***: In the present report, we demonstrated the diagnostic and treatment decision utility of ^(68)Ga PSMA-PET/cT in Asian men with rapid BcR. Detection of small volume nodal and systemic recurrences at low PSA levels(cation with HT-RT or palliative HT in polymetastatic disease.

Better Glycaemic Outcome, Low Levels of Acute Severe complications, and High Patient Satisfaction in Routine Practice in Type 1 Diabetes Treated with an Insulin Pump

Treatment of patients with Type 1 diabetes with continuous subcutaneous insulin infusion (cSII) was re-introduced in Denmark in this millennium and initiated in the diabetes clinic, Fredericia Hospital, in 2005. The aim of the present study was to present yearly data of quality from 2005 to 2013 from a clinical database of patients treated with cSII in routine practice. Methods: A database was established in 2009-2010. Data from 2005 to 2009 are retrospective, data from 2009 to 2013 prospective. From 2009, patient satisfaction was measured by validated questionnaires. Results: By 31 December 2013, the database contained data from 143 active patients. HbA1c (median and range) decreased from 64 (40 - 126) mmol/mol (8.0 (5.8 - 13.7)%) (n = 104) to 60 (36 - 98) mmol/mol (7.6 (5.4 - 11.1)%) (n = 134) (p cSII. The improved glycaemic control was maintained each year until ≥5 years after initiation of cSII (p change in weight. The number of attacks of severe hypoglycaemia was reduced from 60 attacks in 21 patients (n = 104) the year before cSII to only 5 attacks in 5 patients in 2013 (n = 134) (p corresponding to an incidence of 3.7 episodes per 100 patient years. Each year after initiation of cSII until ≥5 years, the number of episodes of severe hypoglycaemia was reduced (p ce 2005, nine episodes of ketoacidosis have been registered in eight patients corresponding to an incidence of 1.4 episodes per 100 patient years. Patient satisfaction was high one year after initiation of cSII (p conclusion: change of treatment to cSII is accompanied by an improvement in glycaemic control, very pronounced reduction in severe hypoglycaemia, low levels of diabetic ketoacidosis, and pronounced patient satisfaction.

Medication Side Effect Profiles in PD Patients in a Safety-Net Hospital

Background: compulsive behavior, dyskinesias, motor fluctuations, and hallucinations are common Parkinson’s disease (PD) medication side effects. These are yet to be examined in relation to race and level of education. The goal of this analysis was to identify socioeconomic or clinical variables that are associated with compulsive behavior, dyskinesias, motor fluctuations, and hallucinations in patients in a safety-net hospital. Methods: A movement disorder patient database containing 452 patients with idiopathic PD was analyzed for differences in PD medication side effects using univariate and multivariate logistic regression analysis. Race, sex, and level of education were evaluated as possible confounders. Results: A greater proportion of the patients in this study were caucasian males. The only variable associated with compulsive behavior was age, with higher age having a protective effect (p = 0.0336). Disease duration (defined as time since the onset of symptoms), diagnosis duration (time since formal diagnosis), and level of education were significantly associated with dyskinesia inunivariate analysis (p =ctively). However, diagnosis duration was the only variable significantly associated with dyskinesia in multivariate analysis (p = 0.0038), in addition to a borderline significant association when comparing individuals with graduate degree to those who had completed high school education or less (p = 0.0599), with a protective effect of higher education. Disease duration, diagnosis duration, and use of monoamineoxidase inhibitors were also significantly associated with motor fluctuations in the univariate analysis, while only diagnosis duration was significantly associated with motor fluctuations in multivariate analysis (p = 0.0035) with longer diagnosis duration associated with higher risk of motor fluctuations.
