Mechanism-driven strategies for prevention of rheumatoid arthritis

In seropositive rheumatoid arthritis(RA),the onset of clinically apparent inflammatory arthritis(IA)is typically preceded by a prolonged period of autoimmunity manifest by the presence of circulating autoantibodies that can include antibodies to citrullinated protein antigens(ACPA)and rheumatoid factor.This period before clinical IA can be designated preclinical RA in those individuals who have progressed to a clinical diagnosis of RA,and an“at‐risk”status in those who have not developed IA but exhibit predictive biomarkers of future clinical RA.With the goal of developing RA prevention strategies,studies have characterized immune phenotypes of preclinical RA/at-risk states.From these studies,a model has emerged wherein mucosal inflammation and dysbiosis may lead first to local autoantibody production,which should normally be transient,but instead is followed by a systemic spread of the autoimmunity as manifested by serum autoantibody elevations,ultimately driving the development of clinically identified joint inflammation.This model can be envisioned as the progression of disease development through serial“checkpoints”that in principle should constrain or resolve autoimmunity;however,instead,the checkpoints“fail”and clinical RA develops.herein we review the immune processes that are likely to be present at each step and the potential therapeutic strategies that could be envisioned to delay,diminish,halt,or even reverse the progression to clinical RA.Notably,these prevention strategies could utilize existing therapies approved for clinical RA,therapies approved for other diseases that target relevant pathways in the preclinical/at-risk state,or approaches that target novel pathways.

SARS-CoV-2 variants are associated with different clinical courses in children with MIS-C

Background Recent infection with SARS‑CoV-2 in children has been associated with multisystem inflammatory syndrome in children(MIS-C).SARS‑CoV-2 has undergone different mutations.Few publications exist about specific variants and their correlation with the severity of MIS-C.Methods This was a single-center,retrospective study including all patients admitted with MIS-C at Rady Children’s hospital-San Diego between May 2020 and March 2022.Local epidemiologic data,including viral genomic information,were obtained from public records.Demographics,clinical presentation,laboratory values,and outcomes were obtained from electronic medical records.Results The analysis included 104 pediatric patients.Four MIS-C waves were identified.Circulating variants in San Diego during the first wave included clades 20A to C.During the second wave,there were variants from clades 20A to C,20G,21C(Epsilon),20I(Alpha),and 20J(Gamma).The third wave had Delta strains(clades 21A,21I,and 21J),and the fourth had Omicron variants(clades 21K,21L,and 22C).MIS-C presented with similar symptoms and laboratory findings across all waves.More patients were admitted to the pediatric intensive care unit(PICU)(74%)and required inotropic support(63%)during the second wave.None of the patients required mechanical circulatory support,and only two required invasive ventilatory support.There was no mortality.Conclusions The various strains of SARS-CoV-2 triggered MIS-C with differing severities,with the second wave having a more severe clinical course.Whether the differences in disease severity across variants were due to changes in the virus or other factors remains unknown.

Applying heteroatom co-doped carbon nanotube for manifesting high performance in the electrochemical reduction of aqueous nitrogen oxide by gold nanoparticles

Electrochemical NO-to-Nh_(3) under ambient conditions could be a viable alternative having advantages in terms of energy consumption and exhaust gas recycling of NO,replacing a traditional ammonia synthesis method of the haber–Bosch process.In synthesizing boron(B-)and nitrogen(N-)co-doped carbon nanotube(CNT)based gold(Au)catalysts,B-dopants elevate the conductivity of carbon nanotube by sp2 hybridization on graphene and implant B–N domains within the graphene layer,and result in facilitating the embedding amount of Au accompanied by high dispersibility with low particle size.Theoretical density functional theory(DFT)calculations elucidate that the electron cloud transmitted from B-dopant to the active site of Au induces the Lewis acidic site,and the O-distal pathway occurs following a spontaneous reaction.Increment of the electron-deficient B-doping area accompanied by N-defects and B–O edges retains the major valence state of Au as Au^(δ+),and suppresses hydrogen evolution reaction(hER)by repulsing the hindrance of h*.This record exhibits the highest faradaic efficiency(FE)of 94.7%,and Nh_(3) yield rate of 1877.4μg·h^(-1)·mg_(cat)^(-1),which is the optimal yield over energy consumption in the field of the ambient reduction of aqueous NO.

The use of multisensory cues to locate mates can increase an organism's success by acting as a back-up plan whenone system fails, by providing additional information to the receiver, and by increasing their ability to detect mates using sensesthat have different ranges in a variable aquatic environment. In this contribution we review the sensory cues that male horseshoecrabs Limulus polyphemus are known to use when locating mates and then provide new data that shed light on this subject. Duringthe breeding season, females migrate into shore during high tides to spawn. Males attach to females as they approach thebeach or are attracted to pairs already spawning. Vision is well established as an important cue in attracting males. Althoughchemoreception is well known in other marine arthropods, and horseshoe crabs have the anatomy available, there are few studieson chemical cues in this species. Experiments are presented here that provide evidence for chemical cue use. We show that the attraction,and retention, of attached and satellite males to actively spawning females and mating pairs involves multimodal

Effects of Ginkgo biloba extract on cell proliferation and cytotoxicity in human hepatocellular carcinoma cells

AIM: To study the effect of Ginkgo biloba extract (EGb 761)containing 22-27% flavonoids (ginkgo-flavone glycosides) and 5-7% terpenoids (ginkgolides and bilobalides) on cell proliferation and cytotoxicity in human hepatocellular carcinoma (hCC) cells. METhODS: human hCC cell lines (hepG2 and hep3B) were incubated with various concentrations (0-1 000 mg/L) of EGb 761 solution. After 24 h incubation, cell proliferation andcytotoxicity were determined by 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2h-tetrazolium (MTS) assay and lactate dehydrogenase (LDh) release, respectively. After 48 h incubation, the expression of proliferating cell nuclear antigen (PCNA) and p53 protein was measured by Western blotting. RESULTS: The results showed that EGb 761 (50-1 000 mg/L)significantly suppressed cell proliferation and increased LDh release (PhepG2 and hep3B cells compared with the control group. The cell proliferation of hepG2 and hep3B cells treated with EGb 761 (1 000 mg/L) was 45% and 39% of the control group (Ph release of hepG2 cells without and with EGb 761 (1 000 mg/L) treatment was 6.7% and 37.7%, respectively, and that of hep3B cells without and with EGb 761 (1 000 rag/L) treatment was 7.2% and 40.3%, respectively. The expression of PCNA and p53 protein in hepG2 cells treated with EGb 761 (1 000 mg/L) was 85% and 174% of the control group, respectively. CONCLUSION: Ginkgo bilobaextract significantly can suppress proliferation and increase cytotoxicity in hepG2 and hep3B cells. Additionally, Ginkgo biloba extract can decrease PCNA and increase p53 expression in hepG2 cells.

Classifying Abdominal Fat Distribution Patterns byUsing Body Measurement Data

This study aims to explore new categorization that characterizes the distribution clusters of visceral and subcutaneous adipose tissues(VAT and SAT)measured by magnetic resonance imaging(MRI),to analyze the relationship between the VAT-SAT distribution patterns and the novel body shape descriptors(BSDs),and to develop a classifier to predict the fat distribution clusters using the BSDs.In the study,66 male and 54 female participants were scanned by MRI and a stereovision body imaging(SBI)to measure participants’abdominal VAT and SAT volumes and the BSDs.A fuzzy c-means algorithm was used to form the inherent grouping clusters of abdominal fat distributions.A support-vector-machine(SVM)classifier,with an embedded feature selection scheme,was employed to determine an optimal subset of the BSDs for predicting internal fat distributions.A fivefold cross-validation procedure was used to prevent over-fitting in the classification.The classification results of the BSDs were compared with those of the traditional anthropometric measurements and the Dual Energy X-Ray Absorptiometry(DXA)measurements.Four clusters were identified for abdominal fat distributions:(1)low VAT and SAT,(2)elevated VAT and SAT,(3)higher SAT,and(4)higher VAT.The cross-validation accuracies of the traditional anthropometric,DXA and BSD measurements were 85.0%,87.5% and 90%,respectively.Compared to the traditional anthropometric and DXA measurements,the BSDs appeared to be effective and efficient in predicting abdominal fat distributions.

A Metric Approach to hot Topics in Biomedicine via Keyword Co-occurrence

Purpose:To reveal the research hotpots and relationship among three research hot topics in b iomedicine,namely CRISPR,iPS(induced Pluripotent Stem)cell and Synthetic biology.Design/methodology/approach:We set up their keyword co-occurrence networks with using three indicators and information visualization for metric analysis.Findings:The results reveal the main research hotspots in the three topics are different,but the overlapping keywords in the three topics indicate that they are mutually integrated and interacted each other.Research limitations:All analyses use keywords,without any other forms.Practical implications:We try to find the information distribution and structure of these three hot topics for revealing their research status and interactions,and for promoting biomedical developments.Originality/value:We chose the core keywords in three research hot topics in biomedicine by using h-index.

hearing loss and cognitive decline among older adults with atrial fibrillation:the SAGE-AF study

Objective To examine the association between hearing loss and cognitive function cross-sectionally and prospectively among older adults with atrial fibrillation(AF).Methods Patients with AF≥65-year-old(n=1244)in the SAGE(Systematic Assessment of Geriatric Elements)-AF study were recruited from five internal medicine or cardiology clinics in Massachusetts and Georgia.hearing was assessed by a structured questionnaire at baseline.Cognitive function was assessed by Montreal Cognitive Assessment(MoCA)at baseline and one year.Cognitive impairment was defined as score≤23 on the MoCA.The associations between hearing loss and cognitive function were examined by multivariable adjusted logistic regression.Results Participants with hearing loss(n=451,36%)were older,more likely to be male,and have depressive symptoms than patients without hearing loss.At baseline,528(42%)participants were cognitively impaired.Individuals with hearing loss were significantly more likely to have cognitive impairment at baseline[adjusted odds ratio(OR)=1.37,95%confidence interval(CI):1.05–1.81].Among the 662 participants who did not have cognitive impairment at baseline and attended the one-year follow-up visit,106(16%)developed incident cognitive impairment.Individuals with,versus those without,hearing loss were significantly more likely to develop incident cognitive impairment at one year(adjusted OR=1.68,95%CI:1.07–2.64).Conclusions hearing loss is a prevalent but under-recognized factor associated with cognitive impairment in patients with AF.Assessment for hearing loss may be indicated among these patients to identify individuals at high-risk for adverse outcomes.
