The objective of the present study is to propose and evaluate a novel multivariate approach for genetic mapping of complex categorical diseases. This approach results from an application of standard stepwise discriminant analysis to detect linkage based on the differential marker identity-by-descent (IBD) distributions among the different groups of sib pairs. Two major advantages of this method are that it allows for simultaneously testing all markers, together with other genetic and environmental factors in a single multivariate setting and it avoids explicitly modeling the complex relationship between the affection status of sib pairs and the underlying genetic determinants. The efficiency and properties of the method are demonstrated via simulations. The proposed multivariate approach has successfully located the true position(s) under various genetic scenarios. Systematic Errors Introduced into Sorghum Grain Yield Data: Does the Multiseed (<i>msd</i>) Trait Increase Sorghum Seed Yield?

Multiseed (msd) mutant sorghum [Sorghum bicolor (L.) Moench] lines with greatly increased seed numbers were developed. It was originally thought that the msd trait could increase grain yield several times in comparison with the wild type from which the mutant was derived. However, in a small plot trial, msd seed yield decreased when compared to the parent line. Herein we report results that msd seed yield remained either unchanged or slightly increased in comparison to the parent line. We suggest that attempts to measure msd sorghum seed yield were complicated due to systematic errors associated with the post-harvest processing methods, including threshing and pneumatic winnowing equipment that was used for harvest. That is, seed recovery and seed loss from individual panicles were affected by the post-harvest processing. When evaluating sorghum grain yield of types with different seed sizes, threshing and seed cleaning harvesting methods should be optimized for each sorghum line.

AS a metropolitan resident who is used to seeing towering buildings, an incessant stream of vehicles and people, I’ve always longed to have a taste of an ancient civilization, which is vigorous but exquisite, bold but elegant, natural but mysterious. I realized my dream when I went to Xi’an, a capital with a history of over 1,000 years. Xi’an was once the capital for eleven dynasties, including the Zhou, Qin, Han and Tang. This is the place where china’s ancient civilization developed. Ancestors at Banpo created exquisite painted pottery; the Western Zhou Dynasty (about 1,066-771 B.c.) saw the height of the splendors of the bronze culture; the Qin Emperor Shi Huang (Qin Dynasty, from about 221-206 B.c.)

Individuals with and without normal tension glaucoma exhibit comparable performance on tests of cognitive function

AIM:To evaluate aspects of cognition impacted by individuals with and without normal tension ***:Fifty normal tension glaucoma(NTG)and 50 control patients≥50 y of age were recruited from the UcSF Department of *** data and glaucoma parameters were extracted from electronic medical records for both *** of executive function[Executive Abilities:Measures and Instruments for Neurobehavioral Evaluation and Research(EXAMINER)]and learning and memory[california Verbal Learning Test–Second Edition(cVLT-II)]were administered to both NTG and ***,handedness,best-corrected visual acuity,maximum intraocular pressure,optic nerve cup-todisc ratio,visual field and optic nerve optical coherence tomography parameters,and a measure of general health(charlson comorbidity Index)were compared between NTG and controls as well as within NTG *** linear regression was used to compare group performances on the EXAMINER battery and cVLT-II while controlling for age,sex,and years of ***:NTG and controls were comparable with respect to age,sex,race,education,handedness,and the charlson comorbidity Index(P>0.05 for all).Performance on the EXAMINER composite score and the cVLT-II did not differ between NTG and controls(P>0.05 for both).cONcLUSION:This is the first prospective study in which the cognitive function of subject with NTG were evaluated using a comprehensive,computerized neurocognitive *** with NTG do not perform worse than unaffected controls on tests of executive function,learning,and memor *** do not suppor t the hypothesis that individuals with NTG are at higher risk for cognitive dysfunction and/or dementia.

Synthesis, characterization and biological activities of Schiff bases derived from 2-hydroxy-3-nitrobenzaldehyde

Four novel Schiff bases were synthesized by the condensation reaction of 2-hydroxyl-3-nitrobenzaldehyde with either benzothiazoles or benzimidazolamines. The compounds 1–4 were characterized by spectroscopic and analytical methods. The crystal structures of 6-methoxy-1,3-benzothiazol-2-amine and compound 2 are reported only. The 6-methoxy-1,3-benzothiazol-2-amine was found to crystallize in the orthorhombic crystal system in the space group Pbca, having eight molecules in the unit cell, while compound 2 was found to crystallize in the monoclinic crystal system in the space group P2_1/c. The biological activities of these compounds were investigated by evaluating their antibacterial, antifungal, antimalarial, anti-HIV and anti-trypanosomiasis activities. These compounds displayed very interesting antibacterial, antifungal, antimalarial, anti-trypanosomiasis, anti-HIV and low cytotoxicity activity against HeLa cells.

B lymphocytes regulate airway granulocytic inflammation and cytokine production in a murine model of fungal allergic asthma

Sensitization to fungi often leads to a severe form of asthma that is particularly difficult to manage clinically, resulting in increased morbidity and hospitalizations in these patients. Although B lymphocytes might exacerbate asthma symptoms through the production of IgE, these cells might also be important in the protective response against inhaled fungi. Through cytokine release and T-cell interactions, these lymphocytes might also influence the development and maintenance of airway wall fibrosis. JH-/- mice lack the JHgene for the heavy chain component of antibodies, which is critical for B-cell function and survival. These animals have facilitated the elucidation of the role of B iymphocytes in a number of immune responses; however, JH-/- mice have not been used to study fungal allergy. In this study, we examined the role of B lymphocytes using an Aspergillus fumigatus murine fungal aeroallergen model that mimics human airway disease that is triggered by environmental fungal exposure. We compared disease progression in sensitized wild-type BALB/c and JH-/- mice that were exposed to repeated fungal exposure and found no differences in airway hyperresponsiveness, overall pulmonary inflammation or collagen deposition around the large airways. However, the levels of the Th2-type cytokines IL-4 and IL-13 were significantly attenuated in the airways of JH-/- mice relative to the BALB/c controls. By contrast, levels of the inflammatory cytokines IL-17A and IL-6 were significantly elevated in the JH-/- animals, and there was significantly more robust airway eosinophilia and neutrophilia than in control animals. Taken together, these findings demonstrate that B lymphocytes help to regulate granulocytic responses to fungal exDosure in the pulmonary compartment.

Antioxidant and hepatoprotective effects of Ajuga nipponensis extract by ultrasonic-assisted extraction

Objective: To investigate suitable condition for extraction of the active components from Ajuga nipponensis(A. nipponensis). Methods: Orthogonal experimental design was used to determine the optimal extraction parameters for ecdysterones and flavonoids. Finally, the hepatoprotective abilities of A. nipponensis extracts were evaluated by ccl_4-induced animal models. Results:Maximum yields of flavonoids(7.87±0.10) mg/g and ecdysterones(0.73±0.02) mg/g could be obtained when the extraction time was 50 min, the extraction temperature was 60 ℃, and the ratio of sample to 70%(v/v) ethanol was 1:20(w/w). The antioxidant property of A. nipponensis was correlated to the concentration of its extracts. At 5 mg/m L, A. nipponensisextract scavenged 84.8% of DPPH radical and had absorbance values of 2.43±0.04 reducing power. Upon ccl_4-induced liver injury, glutamic oxaloacetic transaminase and glutamic pyruvic transaminase decreased significantly after the mice were treated with A. nipponensis. Histological researches also explained that A. nipponensis reduced the extent of liver lesions induced by ccl_4. conclusions: A. nipponensis exhibited potent antioxidant activity in chemical experimental models and hepatoprotective effect against ccl_4-induced liver damage.
