Treatment of biliary tract cancer with NVP-AEW541:Mechanisms of action and resistance

AIM:To investigate in vitro treatment with NVPAEW541,a small molecule inhibitor of insulin-like growth factor-1 receptor(IGF-1R),in biliary tract cancer (BTC),since this disease is associated with a poor prognosis due to wide resistance to chemotherapeutic agents and ***:Cell growth inhibition by NVP-AEW541 was studied in vitro in 7 human BTC cell lines by automated cell *** addition,the anti-tumoral mechanism of NVP-AEW541 was studied by Western blotting,cell cycle analysis and reverse transcription-polymerase chain reaction(RT-PCR).Anti-tumoral drug effect in combination with gemcitabine,5-fluorouracil(5-FU)and Polo-like kinase 1 inhibitor BI2536 was also studied. RESULTS:In vitro treatment with NVP-AEW541 suppressed growth in all human BTC cell lines,however response was lower in gallbladder *** withNVP-AEW541 was associated with dephosphorylation of IGF-1R and *** contrast,phosphorylation of p42/p44 and Stat3 and expression of Bcl-xL were inconsistently *** addition,treated cells showed cell cycle arrest at the G1/S-checkpoint and an increase in sub-G1 ***,IGF-1R and its ligands IGF-1 and IGF-2 were co-expressed in RT-PCR,suggesting an autocrine loop of tumor cell *** with gemcitabine,NVP-AEW541 exerted synergistic effects, particularly at low concentrations,while effects of combination with 5-FU or BI 2536 were only additive. CONCLUSION:Our findings suggest that NVP-AEW541 is active against BTC in vitro and potentiates the efficacy of gemcitabine.

Due to their very small size,nanoparticles can interact with all cells in the central nervous *** of the most promising nanoparticle subgroups are very small superparamagnetic iron oxide nanoparticles(VSOP)that are citrate coated for electrostatic *** determine their influence on murine blood-derived monocytes,which easily enter the injured central nervous system,we applied VSOP and carboxydextran-coated superparamagnetic iron oxide nanoparticles(Resovist).We assessed their impact on the viability,cytokine,and chemokine secretion,as well as iron uptake of murine blood-derived *** found that(1)the monocytes accumulated VSOP and Resovist,(2)this uptake seemed to be nanoparticle-and time-dependent,(3)the decrease of monocytes viability was treatment-related,(4)VSOP and Resovist incubation did not alter cytokine homeostasis,and(5)overall a 6-hour treatment with 0.75 mM VSOP-R1 was probably sufficient to effectively label monocytes for future *** homeostasis is not altered,it is safe to label blood-derived monocles with *** labeled monocytes can be used to study injured central nervous system sites further,for example with drug-carrying VSOP.

Background:Health policy formulations in India have witnessed a shift from a reactive approach to a more proactive approach over the last *** is therefore important to understand the effectiveness of recent national health policies(such as the National Rural Health Mission and the National Urban Health Mission)in addressing the varied needs of the heterogeneous population of ***:We use datasets from the National Sample Surveys carried out in 2004 and 2014 to understand the change in the health seeking behavior as a result of these *** choice of health care facilities and the associated expenditures are compared through descriptive analyses.A multinomial logistic regression is used to identify the significant parameters which contribute towards the share of health care providers in *** health status of two economically disparate Indian states(Bihar and Kerala)are also compared through specific metrics of ***:It is seen that due to increased availability of facilities in close proximity,both rural and urban residents prefer to avail of those facilities which will result in minimization of transportation *** effectiveness of national health policies is found to vary on a regional *** and health status have a strong correlation,thereby reinforcing that Bihar still lags far behind Kerala in terms of access to equitable health ***:Therefore,a hierarchical system,incorporating medical pluralism and tailor-made policies targeted at diverse health care demands,needs to be put in place to achieve Goal 3 of the Sustainable Development Goals as decreed by the United Nations,i.e.,“health for all”.

Electron donors(EDs)are widely used to improve the H 2 production performance of Schottky junction photocatalysts,but the functions of EDs are still unknown from the perspective of electron transfer ***,Pt nanocluster-decorated CdS nanorod is successfully prepared to construct a typical CdS/Pt Schottky *** nanoclusters with a diameter of∼2 nm are deposited on the surface of CdS nanorods by in situ photoreduction at sub-zero *** CdS/Pt photocatalyst using lactic acid shows a higher H_(2)production rate of 4762μmol g^(-1)h^(-1)compared to that using methanol,tri-ethanolamine,and *** understand the cause,the dynamics of photogenerated carriers in CdS/Pt photocatalysts during ED-assisted H_(2)production are revealed by femtosecond transient absorption *** the four organic EDs,lactic acid enables the fastest electron transfer rate of 1.8×10^(9)s^(-1)and the highest electron transfer efficiency of 76%at the CdS/Pt interface due to the most efficient hole *** work sheds light on the importance of efficient interfacial electron transfer for im-proving the photocatalytic performance of Schottky junction photocatalysts.

Li Zehou belongs among the ranks of the most important Chinese philosophers of the 20th and 21st centuries. In his complex theoretical system, he aimed to reconcile the Chinese cultural heritage with the demands of the contemporary world. Besides elaborating on traditional Chinese philosophy, Li launched many innovative views based on his understanding of specific developments in pre-modern and modern Western philosophy. His philosophy could be described as the search for a synthesis between Western and traditional Chinese thought and a specifically Chinese modernization. In order to provide a basic insight into Li＇s specific methods of combining Kant, early Marx and classical Chinese philosophy, the present article investigates his elaboration of the traditional Chinese paradigm of “the unity of nature and man” （tianren heyi 天人合一） through the lens of ideas about humanized nature （renhua de ziran ）人化的自然） and naturalized humans （ziranhua de ten 自然化的人）.

This study aims to analyze how changes in land use influenced the delivery of ecosystem services in Cezava,a South Moravian agricultural region in the Czech Republic,in the period of 1845–*** observation of this period covering more than 160 years made it possible to reflect on social forces driving processes of transformation in the *** capture the landscape multifunctionality and to indicate the environmental quality of the area under study,seven services provided in parallel by arable land,forests,and bodies of water were *** quantification of ecosystem services is based primarily on the transfer of values from the existing literature and on chronicle reviews and map *** looking back to the more distant past is a challenge and reliable information resources are lacking,a simple scoring method defining the functional features of the ecosystems was applied in order to evaluate the change of qualitative characteristics of the observed *** that,the findings of these integrated assessments were supported by an analysis performed using landscape metrics.A comparison of service provision over the decades revealed that regulating and cultural services were significantly reduced,while provisioning services increased due to the proliferation of arable land,land consolidation,and agricultural ***,a trend of improvement in the delivery of ecosystem services was introduced after *** several uncertainties,this study demonstrates that it is possible to analyze long-term land-use trends to generate more meaningful,spatially explicit information,which can form the basis for landscape planning and ecosystem management.

The Drivers of Child Mortality During the 2012–2016 Drought in La Guajira, Colombia

During the 2012–2016 drought in La Guajira,Colombia, child mortality rates rose to 23.4 out of *** of these children belonged to the Wayuu indigenous community, the largest and one of the most vulnerable in Colombia. At the municipal level, this study found a significant positive correlation between the average child mortality rate and households with a monthly income of less than USD 100, the number of people without access to health insurance, being part of the indigenous population,being illiterate, lacking sewage systems, living in rural areas, and large households with members younger than5 years old and older than 65 years old. No correlation was found with households without access to a water *** stepwise regression analysis showed that households with a monthly income of less than USD 100, no members older than 65 years old, but several children younger than5 years old, account for 90.4% of the child mortality *** study concludes that, if inhabitants had had better incomes or assets, as well as an adequate infrastructure,they could have faced the drought without the observed increase in child mortality.

This study investigates the influence of waste characteristics,especially zeta potential,on the properties of cement pastes and *** focus is to evaluate the impact of the zeta potential of cement particles and waste materials on the sedimentation speed,rheology,and hardening time of stabilized cement *** Cement II F 40,retarder additive,silica,and fly ash were used in the *** pastes were prepared,and during the stabilization period,their rheological properties and pH were *** zeta potential and sedimentation speed of the cement and waste particles were measured at the pH that the pastes presented during the entire stabilization *** the stabilization period,the pastes were subjected to the hardening time *** zeta potential analyses revealed diverse values for the different powder types,with the cement particles exhibiting a zeta potential of−3.0 mV,the silica particles exhibiting−10.5 mV,and the fly ash particles exhibiting−20.3 *** influence of the high zeta potential modulus was observed on the sedimentation speed,with the solution containing fly ash exhibiting a speed of 40.01μm/s,whereas the solution containing only cement exhibited a speed of 99.38μm/*** the pastes,the results indicate that the presence of fly ash particles with a significantly negative zeta potential led to a 16%reduction in hardening time compared to particles with a lower modulus of zeta *** tests showed that the inclusion of fly ash particles prevented the formation of *** the zeta potential influenced agglomerate formation and hardening time,it was found to have no effect on yield stress or viscosity.
