Background:The AbSeS-classification defines specific phenotypes of patients with intra-abdominal infection based on the(1)setting of infection onset(community-acquired,early onset,or late-onset hospital-acquired),(2)presence or absence of either localized or diffuse peritonitis,and(3)severity of disease expression(infection,sepsis,or septic shock).This classification system demonstrated reliable risk stratification in intensive care unit(ICU)patients with intra-abdominal *** study aimed to describe the epidemiology of ICU patients with pancreatic infection and assess the relationship between the components of the AbSeS-classification and ***:This was a secondary analysis of an international observational study(“AbSeS”)investigating ICU patients with intra-abdominal *** patients with pancreatic infection were included in this analysis(n=165).Mortality was defined as ICU mortality within 28 days of observation for patients discharged earlier from the *** with mortality were assessed using logistic regression analysis and reported as odds ratio(OR)and 95%confidence interval(CI).Results:The overall mortality was 35.2%(n=58).The independent risk factors for mortality included older age(OR=1.03,95%CI:1.0 to 1.1 P=0.023),localized peritonitis(OR=4.4,95%CI:1.4 to 13.9 P=0.011),and persistent signs of inflammation at day 7(OR=9.5,95%CI:3.8 to 23.9,P<0.001)or after the implementation of additional source control interventions within the first week(OR=4.0,95%CI:1.3 to 12.2,P=0.013).Gramnegative bacteria were most frequently isolated(n=58,49.2%)without clinically relevant differences in microbial etiology between survivors and ***:In pancreatic infection,a challenging source/damage control and ongoing pancreatic inflammation appear to be the strongest contributors to an unfavorable short-term *** this limited series,essentials of the AbSeS-classification,such as the setting of infection onset,diffuse

High-entropy alloys(HEAs)have the potential to be a paradigm-shift for rational catalyst discovery but this new type of alloy requires a completely new approach to predict the surface *** addition to the ligand effect perturbing the surface–adsorbate bond,the random configuration of elements in the surface will also induce local strain effects due to the varying radii of neighboring *** modelling of HEA surface reactivity requires an estimate of this effect:To what degree is the adsorption of intermediates on these lattice distorted atomic environments affected by local strain?In this study,more than 3,500 density functional theory(DFT)calculated adsorption energies of*OH and*O adsorbed on the HEAs IrPdPtRhRu and AgAuCuPdPt are statistically analyzed with respect to the lattice constants of the alloys and the surfaces of each individual binding *** is found that the inherent distortion of the lattice structure in HEAs releases the local strain effect on the adsorption energy as the atomic environment surrounding the binding atom(s)settles into a relaxed *** is even observed to be true for clusters of atoms of which the sizes deviate significantly from the atomic environment in which they are *** elucidates an important aspect of binding site interaction with the neighboring atoms and thus constitutes a step towards a more accurate theoretical model of estimating the reactivity of HEA surfaces.

Aluminum metal matrix composites （Al-MMCs） are difficult to machine. The reinforcement of aluminum using ceramic particles accelerates tool wear. Moreover, demanded machining accuracies or properties of the surface layer are difficult to achieve. In the present study, the effect of silicon carbide reinforcement particles on the surface layer of the workpiece was investigated using multiple cutting conditions for dry turning. Three differently reinforced Al-MMCs regarding the volume percent-age （17% and 30%） and the particle size （0.6 μm and 3 μm） and their non-reinforced matrix were considered as the workpiece materials. The reinforcement and the cutting condition affect the results of turning. A greater particle volume percent improves the surface roughness and decreases the tensile stress in the surface. The smaller particle size caused a lower tensile stress in the surface. A general effect of the particle size on the workpiece roughness can not be concluded. The most important cutting parameter for the surface layer of the workpiece is the feed. Greater feeds decrease the tensile stress in the surface, but deteriorate the surface quality.

Impaired axonal transport has been observed in patients with amyotrophic lateral sclerosis(ALS)and in animal models,suggesting that transport proteins likely play a critical role in the pathological mechanism of *** of Kinesin-family-member 5α(Kif5α),a neuron-specific isoform of heavy chain kinesin family,has been described in several neurological disorders,in humans and animal models,including *** this study,we determined Kif5αexpression by gene sequencing,quantitative reverse transcription-polymerase chain reaction,and western blot assay in the cervical spinal cord of wobbler mice and immunofluorescence staining in dissociated cultures of the ventral ***,we observed the distribution of Kif5αand mitochondria along motor neuronal branches by confocal *** results showed that Kif5αexpression was greatly dysregulated in wobbler mice,which resulted in altered distribution of Kif5αalong motor neuronal branches with an abnormal mitochondrial ***,our results indicate that dysregulation of Kif5 and therefore abnormal transport in motor neuronal branches in this ALS model could be causative for several pathological findings at the cellular level,like misallocation of cytoskeletal proteins or organelles like mitochondria.

In this work two different fluorochromes （Alexa 594 and Alexa 680） are conjugated to the same monoclonal antibody （Cetuximab） for obtaining a characteristic M-shaped dual-peak spectrum. Dual-labeling of Cetuximab by mixing both fluorochromes before the conjugation step gives spectral results similar to those of mixing of fluorochrome-labeled Cetuximab after the conjugation step （P 〉 0.05）. In conclusion, both methods may be used equivalently for producing a dual-labeled single-antibody probe. Future studies may test whether the M-shaped spectrum may increase the diagnostic confidence in tumor-targeted multispectral optical imaging.

High-quality DNA extraction is a crucial step in metagenomic *** by different isolation kits impairs the comparison across datasets.A trending topic is,however,the analysis of multiple metagenomes from the same patients to draw a holistic picture of microbiota associated with *** thus collected bile,stool,saliva,plaque,sputum,and conjunctival swab samples and performed DNA extraction with three commercial *** each combination of the specimen type and DNA extraction kit,20-gigabase(Gb)metagenomic data were generated using short-read *** profiles of the specimen types showed close proximity to each other,we observed notable differences in the alpha diversity and composition of the microbiota depending on the DNA extraction *** kit outperformed all selected kits on every *** reached consistently good results using the Qiagen QiAamp DNA Microbiome *** on the specimen,our data indicate that over 10 Gb of sequencing data are required to achieve sufficient resolution,but DNA-based identification is superior to identification by mass ***,longread nanopore sequencing confirmed the results(correlation coefficient>0.98).Our results thus suggest using a strategy with only one kit for studies aiming for a direct comparison of multiple microbiotas from the same patients.

Background: To study novel treatment modalities for pancreatic ductal adenocarcinoma(PDAC), we need to transfer the knowledge from in vitro to in vivo. It is important to mirror the clinical characteristics of the typically local invasive growth of pancreatic cancer and the distant spread resulting in liver metastasis. Notably, for xenotransplant studies using human specimen, two models, i.e. subcutaneous(s.c.) and orthotopic(o.t.) transplantation are widely used. Methods: The subcutaneously and orthotopically inoculated Colo357 Bcl-x L cell-derived tumors were directly compared with and without TNF-related apoptosis inducing ligand(TRAIL) treatment. The size of primary tumors, number of liver metastasis and the histologic markers Ki67, M30, TNF-α and CD31 were assessed. Results: Upon TRAIL treatment, the primary tumors did not change their size, neither in the s.c. nor in the o.t. approaches. But when s.c. was compared to o.t., the size of the s.c. tumors was more than twofold bigger than that of the o.t. tumors( P<0.01). However, mice with orthotopically inoculated PDAC cells developed liver metastasis upon TRAIL treatment much more frequently( n=13/17) than mice with subcutaneously inoculated PDAC cells( n=1/11)( P<0.01). As a likely driving force for this increased metastasis, a higher TNF-α staining intensity in the o.t. tumors was observed by immunohistochemistry. Conclusions: These data from a direct side-by-side comparison underline the importance of the proper inoculation site of the PDAC cells. Local invasion and liver metastases are a hallmark of PDAC in the clinic;the o.t. model is clearly superior in reflecting this setting. Moreover, a serious side-effect of a possible new therapeutic compound became obvious only in the o.t. model.

Sharing sensitive data is a specific challenge for research infrastructures in the field of life sciences. For that reason a toolbox has been developed, providing resources for researchers who wish to share and use sensitive data, to support the workflows for handling these kinds of digital objects. Common and community approved annotations are required to be compliant with FAIR principles(Findability, Accessibility,Interoperability, Reusability). The toolbox makes use of a tagging(categorisation) system, allowing consistent labelling and categorisation of digital objects, in terms relevant to data sharing tasks and activities. A pilot study was performed within the Horizon 2020 project EOSC-Life, in which 2 experts from 6 life sciences research infrastructures were recruited to independently assign tags to the same set of 10 to 25 resources related to sensitive data management and data sharing(in total 110). Summary statistics of agreement and observer variation per research infrastructure are provided. The pilot study has shown that experts were able to attribute tags but in most cases with a considerable observer variation between experts. In the context of CWFR(Canonical Workflow Frameworks for Research), this indicates the necessity for careful definition,evaluation and validation of parameters and processes related to workflow descriptions. The results from this pilot study were used to tackle this issue by revising the categorisation system and providing an updated version.
