Crystallization and preliminary crystallographic studies of insulin T_3R_3 states created in different conditions

In the presence of a series of small organic molecules including 1, 4 dioxane, DMF, isopropanol, rDNA human insulin, mutants A21 Asn→Ser and (A21 Asn→Ser, B27 Thr→Arg) human insulin have been crystallized in both Zn 2+ containing and Zn 2+ free system. Preliminary crystallographic analyses show that they all belong to T 3R 3 structural type. The results indicate that, except phenolic derivatives and lyotropic anions reported before, some other appropriate organic molecules can also induce the T 3R 3 conformation transition of insulin.

The modification of nanostructured materials is of great interest due to controllable and unusual inherent properties in such materials. Single phase Fe doped Zn O nanostructures have been fabricated through simple, versatile and quick low temperature solution route with reproducible results. The amount of Fe dopant is found to play a significant role for the growth of crystal dimension. The effect of changes in the morphology can be obviously observed in the structural and micro-structural investigations, which may be due to a driving force induced by dipole-dipole interaction. The band gap of Zn O nanostructures is highly shifted towards the visible range with increase of Fe contents, while ferromagnetic properties have been significantly *** prepared nanostructures have been found to be nontoxic to SH-SY5 Y Cells. The present study clearly indicates that the Fe doping provides an effective way of tailoring the crystal dimension, optical band-gap and ferromagnetic properties of Zn O nanostructure-materials with nontoxic nature, which make them potential for visible light activated photocatalyst to overcome environmental pollution, fabricate spintronics devices and biosafe drug delivery agent.

Crystallization and preliminary crystallographic analyses of human insulin mutant B9(Ser→Asp)in different aggregation forms

B9(Ser→Asp) insulin, one of the fast-acting insulin mutants produced by protein engineering, has been crystallized. Three crystal forms have been obtained, which belong to orthorhombic, tetragonal and rhombohedral system, respectively. Each crystal form contains different aggregation units of the mutant, i.e. dimer or hexamer. The diffraction data of the former two have been collected beyond 0.2 nm resolution. The structural analyses and comparisons will provide some information about insulin’s self-association and the structural basis of the fast absorption produced from the mutation.

The Netherlands is "well known" for its nitrogen problems; it has one of the highest reactive nitrogen (Nr) emission densities in the world. It is a small country at the delta of several large European rivers. Ever since the industrial revolution, there has been a growing excess of nutrients and related emissions into the atmosphere (ammonia, nitrogen oxides and nitrous oxide)and into groundwater and surface water (nitrate), leading to a large range of cascading environmental impacts. Vehicular traffic, sewage and animal husbandry are the main sources of oxidized and reduced forms of Nr. This paper provides an overview of the origin and fate of nitrogen in the Netherlands, the various reported impacts of nitrogen, the Dutch and European policies to reduce nitrogen emissions and related impacts. In addition, ways are presented to go forward to potentially solve the problems in a European perspective. Solutions include the improvement of nitrogen efficiencies in different systems, technological options and education.

Three sulphur-rich commercial crude oils have been studied,which contain sulphur as high as up to4—12%.These samples were collected from Tertiary hypersaline lake sediments of the Jianghan Basin,Hubei Province at different depths,but above the oil generation threshold（2200m）.FPD—GC and GC-MSdata show that aromatic fractions of the crude oils are composed of different homologues of sulphur-containing compounds,including long-chain normal alkyl-thiophenes and-thiolanes,long-chainisoprenoid-thiophenes and-thiolanes,and *** is worth noting that the distribution patternsof long-chain alkyl-thiophenes and-thiolanes from two shallow-seated crude oils are quite similar to thoseof normal alkanes showing marked even-odd *** seems that the even-odd predominance ofsulphur-containing compounds decreases with increasing burial depth of the crude *** majorcomponent of aliphatic fraction is phytane,and similarly the major peaks of aromatic fractions also representC20isoprenoid *** preliminary conclusions have been drawn from the above discussion:（1）Abundant sulphur-containing compounds may be used as an indicator of low mature or immature crude oils produced fromhypersaline lake sediments;（2）Sulphur-containing compounds are considered to be early diageneticproducts of reactions between elemental sulphur or sulfides and alkanes or their precursors（phytols,fattyacids,alcohols,etc.）,or of bacterial activities,but not direc■ inputs of organisms.

目的探讨CT灌注成像技术对定量评价肿瘤组织血流动力学改变,区分肿瘤组织和正常组织、肿瘤中心组织和肿瘤边缘组织的有效性,以及定量监测肿瘤微血管系对抗血管生成药物治疗反应的有效性。方法 15只一侧大腿植入VX2肿瘤的新西兰大白兔分为2组,1组行抗血管治疗(n=11),1组为对照组(n=4)。于肿瘤种植1周后、治疗1周后和治疗2周后进行DEC-CT扫描,分别计算肿瘤区、肿瘤边缘区、肿瘤中心区和正常组织区的灌注参数:血流量(blood flow,bf)、血容量(blood volume,BV)、平均通过时间(meantransit time,MTT)和表面通透性(permeability surface,PS),并对各灌注参数值进行统计分析。结果肿瘤区和正常组织区的bf、PS、MTT值分别为(149.32±30.99)ml/(***)、(28.24±5.15)ml/(***)、(2.79±0.66)s和(32.18±4.10)ml/(***)、(11.88±0.71)ml/(***)、(6.57±0.90)s,两者比较差异均有统计学意义(Pbf、PS、MTT值分别为(175.70±34.43)ml/(***)、(32.43±6.52)ml/(***)、(3.87±0.84)s和(19.07±3.58)ml/(***)、(18.75±2.10)ml/(***)、(8.11±1.44)s,两者比较差异均有统计学意义(Pbf、BV、PS、MTT值在治疗后第2周后从187.57ml/(***)、5.21ml/100g、30.74ml/(***)、3.87s改变为118.48ml/(***)、4.21ml/100g、24.02ml/(***)、4.27s,对照组从129.74ml/(***)、3.22ml/100g、25.85ml/(***)、3.68s改变为362.33ml/(***)、10.86ml/100g、39.31ml/(***)、3.54s,改善明显。结论 CT灌注成像技术能从血流动力学方面提供肿瘤定量信息,并能定量监测肿瘤微血管系对抗血管生成药物治疗的反应。
