Withania somnifer. r.ot Extr.ct Enhances Telomer.se Activity in the Human HeLa Cell Line

Aging is a deceler.ting unidir.ctional pr.cess of life. Shor.ening of telomer.c DNA, the (TTAGGG)n hexanucleotide r.peats, which for. the caps at the chr.mosome ends, is implicated to deter.ine the aging pr.cess, and mor. impor.antly the healthy lifespan itself. Telomer.se, a r.bonucleopr.tein having r.ver.e tr.nscr.ptase activity, ar.ests telomer. loss thr.ugh addition of the TTAGGG r.peats de novo, to the ends of the chr.mosome. The telomer./telomer.se maintenance is an inevitable necessity to delay aging and for.a healthy lifespan. Her., we r.por. the potential of full-spectr.m, high concentr.tion Ashwagandha (Withania somnifer.), an Ayur.edic medicinal her., r.ot extr.ct to incr.ase telomer.se activity. HeLa cells, when tr.ated with var.ous concentr.tions of Ashwagandha r.ot extr.ct, showed an incr.ase in telomer.se activity measur.d with the established Telomer.se r.pid Amplification Pr.tocol (Tr.P) assay. Ashwagandha r.ot extr.ct incr.ased telomer.se activity with highest enhancement of ~45% at 10 - 50 μg concentr.tion. Thus, Ashwagandha r.ot extr.ct has the anti-aging inducing potential.

Despite exhaustive sear.h, no dr.g is in sight for.AD. Ear.ier. we r.por.ed that r.ser.ine, an antihyper.ensive and antipsychotic dr.g, amelior.tes Amyloid beta (Aβ-AD causing peptide) toxicity and confer. sever.l positive enhancements in the C. elegans model system. Her., we evaluate whether.r.ser.ine can pr.vide pr.tection against wor.ing memor. and against AD in the mouse model. r.ser.ine (0.08 mg) was administer.d or.lly on alter.ate days to the non-Tg and acceler.ted Aβ deposition (at 2 months of age)and cognitive deficit (4 months of age) developing 5XFAD AD Tg mouse model expr.ssing mutant human APP (3 familial mutations) and human Pr.senilin1(2 familial mutations) in the neur.ns, and follow their.wor.ing memor. for.2 months using the spontaneous Y-maze alter.tion behavior.l par.digm. r.ser.ine enhanced wor.ing memor. in non-Tg mice and impr.ved the cognitive deficit in the 5XFAD AD Tg mice. Hence, r.ser.ine can be consider.d for.a detailed evaluation in the 3X Tg AD mouse model and a pilot study in AD patients.
