Despite the maturity of ensemble numerical weather prediction(NWP),the resulting forecasts are still,more often than not,*** such,forecast calibration tools have become *** those tools,quantile regression(QR)is highly competitive in terms of both flexibility and predictive ***,a long-standing problem of QR is quantile crossing,which greatly limits the interpretability of QR-calibrated *** this point,this study proposes a non-crossing quantile regression neural network(NCQRNN),for calibrating ensemble NWP forecasts into a set of reliable quantile forecasts without *** overarching design principle of NCQRNN is to add on top of the conventional QRNN structure another hidden layer,which imposes a non-decreasing mapping between the combined output from nodes of the last hidden layer to the nodes of the output layer,through a triangular weight matrix with positive *** empirical part of the work considers a solar irradiance case study,in which four years of ensemble irradiance forecasts at seven locations,issued by the European Centre for medium-Range Weather Forecasts,are calibrated via NCQRNN,as well as via an eclectic mix of benchmarking models,ranging from the naïve climatology to the state-of-the-art deep-learning and other non-crossing *** and stringent forecast verification suggests that the forecasts post-processed via NCQRNN attain the maximum sharpness subject to calibration,amongst all ***,the proposed conception to resolve quantile crossing is remarkably simple yet general,and thus has broad applicability as it can be integrated with many shallow-and deep-learning-based neural networks.

multiple myeloma(mm)is a common haematological malignancy that is associated with a high rate of venous thromboembolism(VTE)with almost 10% of patients suffering thrombosis during their disease *** studies have shown that,despite current thromboprophylaxis strategies,VTE rates in mm remain disappointingly *** pathophysiology behind this consistently high rate of VTE is likely multifactorial.A number of factors such as anti-thrombin deficiency or raised coagulation Factor VIII levels may confer resistance to heparin in these patients,however,the optimal method of clinically evaluating this is unclear at present,though some groups have attempted its characterisation with thrombin generation testing(TGT).In addition to testing for heparin resistance,TGT in patients with mm has shown markedly varied abnormalities in both endogenous thrombin potential and serum thrombomodulin *** from these thrombin-mediated processes,other mechanisms potentially contributing to thromboprophylaxis failure include activated protein C resistance,endothelial toxicity secondary to chemotherapy agents,tissue factor abnormalities and the effect of immunoglobulins/“m-proteins”on both the endothelium and on fibrin fibre *** thus appears clear that there are a multitude of factors contributing to the prothrombotic milieu seen in mm and further work is necessitated to elucidate which factors may directly affect and inhibit response to anticoagulation and which factors are contributing in a broader fashion to the hypercoagulability phenotype observed in these patients so that effective thromboprophylaxis strategies can be employed.

Tailoring the degradation rate of magnesium through biomedical nano-porous titanate coatings

A novel approach was developed to reduce the corrosion rate of magnesium(mg)metal,utilising titanate *** sputtering was used to deposit ca.500 nm titanium(Ti)coatings onto pure mg discs,followed by hydrothermal conversion and ion exchange reactions to produce sodium and calcium titanate *** confirmed the characteristic nanoporous structure of sodium and calcium titanate,with thicknesses ranging from ca.0.8 to 1.4μ*** analysis confirmed the presence of Ti^(4+)-o,Na-o,and Ca-o bonding,whilst Raman spectroscopy demonstrated characteristic vibrational modes(such as Tio 6 octahedral vibrations)of the sodium and calcium titanate perovskite ***,corrosion studies through potentiodynamic polarisation measurements demonstrated the NB/NH CaTC samples to be superior in reducing mg degradation,compared to other samples tested,through an increase in E_(corr)from-1.49 to-1.33 V,and the reduction in corrosion current density,i corr,from 0.31 to 0.06 mA/cm^(2)for mg and NB/NH CaTC samples,*** was a clear trend noted for the NB/NH samples,which showed an increase in E corr to more positive values in the following order:mgoatedoporous titanate coatings have potential to be applied onto degradable plates for bone fracture fixation,or other orthopaedic applications.

mycophenolate mofetil for maintenance of remission in steroid-dependent autoimmune pancreatitis

Systemic corticosteroids represent the standard treatment for autoimmune pancreatitis with IgG4-associated *** steroid-dependent disease,azathioprine has been used for maintenance of *** mofetil has been used for transplant immunosuppression and more recently for autoimmune hepatitis;however,there are no case reports to date on the use of mycophenolate mofetil in adult patients with autoimmune pancreatitis.A patient with IgG4-mediated autoimmune pancreatitis and IgG4-associated cholangitis refractory to steroids and intolerant of azathioprine was treated with mycophenolate mofetil,which inhibits de novo guanosine synthesis and blockade of both B and T lymphocyte *** of mycophenolate mofetil and uptitration to 1000 mg by mouth twice daily over a treatment period of 4 mo was associated with improvement in the patient's energy level and blood glucose control and was not associated with any adverse *** patient was managed without a biliary ***,there was a return of symptoms,jaundice,increase in transaminases,and hyperbilirubinemia when the prednisone dose reached 11 mg per *** the first report of mycophenolate mofetil use in an adult patient with IgG4-associated autoimmune pancreatitis and IgG4-associated cholangitis,the introduction of mycophenolate mofetil was safe and well-tolerated without adverse events,but it did not enable discontinuation of the *** mofetil and other immunomodulatory therapies should continue to be studied for maintenance of remission in the large subset of patients with refractory or recurrent autoimmune pancreatitis.

Validation of the Rockall scoring system for outcomes from non-variceal upper gastrointestinal bleeding in a Canadian setting

AIm: To validate the Rockall scoring system for predicting outcomes of rebleeding, and the need for a surgical procedure and death. mETHoDS: We used data extracted from the Registry of Upper Gastrointestinal Bleeding and Endoscopy including information of 1869 patients with non-variceal upper gastrointestinal bleeding treated in Canadian hospitals. Risk scores were calculated and used to classify patients based on outcomes. For each outcome, we used χ2 goodness-of-fit tests to assess the degree of calibration, and built receiver operating characteristic curves and calculated the area under the curve (AUC) to evaluate the discriminative ability of the scoring system. RESULTS: For rebleeding, the χ2 goodness-of-fit test indicated an acceptable fit for the model [χ2 (8) = 12.83, P = 0.12]. For surgical procedures [χ2 (8) = 5.3, P = 0.73] and death [χ2 (8) = 3.78, P = 0.88], the tests showed solid correspondence between observed proportions and predicted probabilities. The AUC was 0.59 (95% CI: 0.55-0.62) for the outcome of rebleeding and 0.60 (95% CI: 0.54-0.67) for surgical procedures, representing apoor discriminative ability of the scoring system. For the outcome of death, the AUC was 0.73 (95% CI: 0.69-0.78), indicating an acceptable discriminative ability. CoNCLUSIoN: The Rockall scoring system provides an acceptable tool to predict death, but performs poorly for endpoints of rebleeding and surgical procedures.

Physical separation from the mate diminishes male＇s attentiveness towards other females： a study in monogamous prairie voles microtus ochrogaster

We tested whether continuous cohabitation in monogamous voles affects the mated male＇s atten-tiveness to his breeding partner versus another female. Each male was housed in a 3-chamber ap-paratus with a Focal female （FF） and a Control female （CF） for 13 days then placed in a T-maze toassess his attentiveness to and memory of those females. The Distal male remained physically sep-arated from both females, but received their distal cues. The Separate male cohabited with the FFfor 3 days then remained physically separated from both females. The Disrupt male＇s continuouscohabitation with the FF was disrupted by having him physically separated from her after 10 daysand placed with the CF for the last 3 days. The Continuous male cohabited continuously with the FFfor 13 days. With females in the T-maze, the Separate and Disrupt males spent more time near theFF＇s box and the Disrupt males spent more time manipulating the FF＇s box than the CF＇s box. TheSeparate males groomed themselves more when near the FF＇s box than the CF＇s box. The Distaland Continuous males＇ attentiveness to the two females did not differ. Results suggest that phys-ical distance from the partner may reduce male＇s attentiveness toward other potential *** voles might be similar to socially monogamous primates in using tactile cues as a signal formaintaining their social bonds.

Crouzon syndrome mouse model exhibits cartilage hyperproliferation and defective segmentation in the developing trachea

Crouzon syndrome is the result of a gain-of-function point mutation in FGFR2. mimicking the human mutation, a mouse model of Crouzon syndrome(Fgfr2342Y) recapitulates patient deformities, including failed tracheal cartilage segmentation, resulting in a cartilaginous sleeve in the homozygous mutants. We found that the Fgfr2C342Y/C^342Y mutants exhibited an increase in chondrocytes prior to segmentation. This increase is due at least in part to over proliferation. Genetic ablation of chondrocytes in the mutant led to restoration of segmentation in the lateral but not central portion of the trachea. These results suggest that in the Fgfr2C342Y/C342Y mutants, increased cartilage cell proliferation precedes and contributes to the disruption of cartilage segmentation in the developing trachea.
