Hematologic and biochemical reference intervals for specific pathogen free 6-week-old Hampshire-Yorkshire crossbred pigs

Background： Hematologic and biochemical reference intervals depend on many factors, including age. A review of the literature highlights the lack of reference intervals for 6-wk-old specific pathogen free （SPF） Hampshire-Yorkshire crossbred pigs. For translational research, 6-wk-old pigs represent an important animal model for both human juvenile colitis and diabetes mellitus type 2 given the similarities between the porcine and human gastrointestinal maturation process. The aim of this study was to determine reference intervals for hematological and biochemical parameters in healthy 6-wk-old crossbred pigs. Blood samples were collected from 66 clinically healthy Hampshire-Yorkshire pigs. The pigs were 6 wks old, represented both sexes, and were housed in a SPF facility. Automated hematological and biochemical analysis were performed using an AdVIA 120 Hematology System and a Cobas 6000 C501 Clinical Chemistry Analyzer. Results： Reference intervals were calculated using both parametric and nonparametric methods. The mean, median, minimum, and maximum values were calculated. Conclusion： As pigs are used more frequently as medical models of human disease, having reference intervals for commonly measured hematological and biochemical parameters in 6-wk-old pigs will be useful. The reference intervals calculated in this study will aid in the diagnosis and monitoring of both naturally occurring and experimentally induced disease. In comparison to published reference intervals for older non SPF pigs, notable differences in leukocyte populations, and in levels of sodium, potassium, glucose, protein, and alkaline phosphatase were observed.

Chiral 1,1’-binaphthyl-linked diporphyrin‘tweezers’(R)-1/(S)-1 and the corresponding zinc(II)complexes(R)-2/(S)-2 were prepared as chiral host molecules,and their utility for chiral analyses(especially enantiomeric excess(ee)determinations)were ***(1-n-dodecyl)porphyrins were used for the first time as the interacting *** capabilities of the diporphyrin tweezers were investigated by titrations with(R,R)-and(S,S)-cyclohexane-1,2-diamine(CHdA).The host molecules could be used as multichannel probes of ee by using UVvis,circular dichroism(Cd),fluorescence emission and 1H nuclear magnetic resonance(1 H-NMR)*** configurations could also be differentiated using Cd or 1HNMR *** three optical techniques give good resolution of ee with reasonable sensitivity considering the low concentrations used(ca.10^-6 mol·L^-1).The ee determination of CHdA enantiomers using NMR spectroscopy is also possible because of the reasonably well separated resonances in the case of(R,R)-and(S,S)-***-metallated(R)-1/(S)-1 hosts could not be used to detect chiral information in a strongly acidic chiral *** work demonstrates the utility of 1,1’-binapthyllinked chiral hosts for chiral analysis of ditopically interacting enantiomers.

Are trends in SeaWiFS chlorophyll time-series unusual relative to historic variability

For selected locations in the Atlantic and Pacific Ocean, we compared surface ocean chlorophyll time series extracted from SeaWiFS imagery from 1997-2004 with the results of an ocean coupled circulation and biogeochemical model covering the period 1958-2004. during the 1997-2004 time period, linear trends in model and satellite time series were significantly correlated at most of the 44 sites we studied. Eleven sites were selected for further study, and we used the longer time series of the model to assess whether trends observed during the SeaWiFS period at these 11 sites were unusual in relation to those observed over the longer historical period covered by the model. The results show that the trends observed during the SeaWiFS period were not unusual and fell well within the range in magnitude of linear trends observed in other 8-year periods of model output. This result implies that the SeaWiFS satellite ocean color time series is not yet sufficiently long, on its own, to directly observe any long term changes in phytoplankton chlorophyll that may be occurring in the surface waters of the open ocean as a result of increased ocean stratification linked to global climate changed.

Cancer risk in primary sclerosing cholangitis: Epidemiology,prevention, and surveillance strategies

Primary sclerosing cholangitis(PSC) is a rare cholestatic liver disease characterized by progressive fibroinflammatory destruction of the intra-and/or extrahepatic biliary ducts. While its features and disease course can be variable,most patients with PSC have concurrent inflammatory bowel disease and will eventually develop liver cirrhosis and end-stage liver disease, with liver transplantation representing the only potentially curative option. Importantly,PSC is associated with a significantly increased risk of malignancy compared to the general population, mainly cholangiocarcinoma, gallbladder carcinoma,hepatocellular carcinoma, and colorectal cancer, with nearly 50% of deaths in patients with PSC being due to cancer. Therefore, robust surveillance strategies are needed, though uncertainty remains regarding how to best do so. In this review, we discuss the epidemiology, prevention, and surveillance of cancers in patients with PSC. Where evidence is limited, we present pragmatic approaches based on currently available data and expert opinion.

Impact of source tissue and ex vivo expansion on the characterization of goat mesenchymal stem cells

Background： There is considerable interest in using goats as models for genetically engineering dairy animals and also for using stem cells as therapeutics for bone and cartilage repair. Mesenchymal stem cells （MSCs） have been isolated and characterized from various species, but are poorly characterized in goats. Results： Goat MSCs isolated from bone marrow （BM-MSCs） and adipose tissue （ASCs） have the ability to undergo osteogenic, adipogenic and chondrogenic differentiation. Cytochemical staining and gene expression analysis show that ASCs have a greater capacity for adipogenic differentiation compared to BM-MSCs and fibroblasts. different methods of inducing adipogenesis also affect the extent and profile of adipogenic differentiation in MSCs. Goat fibroblasts were not capable of osteogenesis, hence distinguishing them from the MSCs. Goat MSCs and fibroblasts express Cd90, Cd105, Cd73 but not Cd45, and exhibit cytoplasmic localization of OCT4 protein. Goat MSCs can be stably transfected by Nucleofection, but, as evidenced by colony-forming efficiency （CFE）, yield significantly different levels of progenitor cells that are robust enough to proliferate into colonies of integrants following G418 selection. BM-MSCs expanded over increasing passages in vitro maintained karyotypic stability up to 20 passages in culture, exhibited an increase in adipogenic differentiation and CFE, but showed altered morphology and amenability to genetic modification by selection. Conclusions： Our findings provide characterization information on goat MSCs, and show that there can be significant differences between MSCs isolated from different tissues and from within the same tissue. Fibroblasts do not exhibit trilineage differentiation potential at the same capacity as MSCs, making it a more reliable method for distinguishing MSCs from fibroblasts, compared to cell surface marker expression.

Lunasin protease inhibitor concentrate(LPIC)is a novel combination of soy bioactive peptide lunasin,Kunitz and Bowman-Birk protease *** reported anti-inflammatory and anticancer properties of each one of them suggest LPIC as a promising candidate for the treatment of infl ammatory-related *** objective was to assess the in vivo anti-infl ammatory properties of ***,an in vitro test was performed in lipopolysaccharide(LPS)-activated RAW264.7 murine macrophages by measuring the production of nitric oxide(NO),interleukin-6(IL-6),and tumor necrosis factorα(TNF-α)as infl ammatory *** the in vivo model,ulcerative colitis(UC)was induced in mice via oral administration of dextran sodium sulfate(dSS).LPIC treatment was performed via daily intraperitoneal injection of 50 mg/kg body *** weight,visible blood in stool and stool consistency were scored daily as macroscopic indicators of disease *** blood was evaluated by the presence of hemoglobin in stool every third *** length,caecum weight,colonic myeloperoxidase activity(MPO),presence of pro-inflammatory cytokines in blood and colon,changes in the architecture,and expression of inducible nitric oxide synthase(i NOS)in colonic tissue were *** vitro,LPIC induced production of NO and maintained cytokine levels in comparison to activated untreated *** vivo,LPIC increased colonic bleeding and did not improve macroscopic markers of the disease,but reduced colonic IL-1βand IL-6,decreased systemic circulation of TNF-α,attenuated neutrophils infi ltration and i NOS expression in colonic tissue,and diminished the damage in colonic *** results suggest that combinations of peptides in LPIC may counteract the antiinfl ammatory properties in vitro;while in vivo,LPIC can signifi cantly reduce the histopathological damage,hence is a possible therapeutic strategy to attenuate UC.

Antioxidant and antihyperlipidemic activity of Costus speciosus against atherogenic diet-induced hyperlipidemia in rabbits

Objective: An extract of Costus speciosus(CSE), a herb widely used in folk medicine, was evaluated for its antioxidant, antihyperlipidemic and ameliorating effects on histopathological changes in atherogenic ***: Twenty-four male rabbits(Oryctolagus cuniculus) were divided into 4 groups. Three groups were fed a diet containing 3% saturated fat and 1.3% cholesterol for 40 d. One of these was sacrificed on the 40 th day and was called the pathogenic(P) group; the other two groups received treatment for another 30 d as follows: one received 0.8 g/(kgád) of CSE and the other was given 0.01 g/(kgád) of simvastatin. The normal group was sacrificed on the 70 th day and used as a ***: CSE showed radical-scavenging ability. Administration of CSE for a 30-day period resulted in a significant decrease in total cholesterol, triacylglycerol, low-density lipoprotein and aspartate aminotransferase compared to the P group, while levels of hemoglobin, packed corpuscular volume and red blood cells were elevated. With respect to studies performed on the heart, a decrease in malondialdehyde and an increase in reduced glutathione were noted. Total protein increased in the liver, heart and aorta after treatment with CSE and also a marked improvement in histopathological parameters was ***: The present findings indicate that the C. speciosus rhizome possesses antiatherogenic and antioxidant properties which may provide protective effects against oxidative stress in atherosclerotic rabbits.

Singlet Oxygen Generation and Photoinduced Charge Separation of Tetra Polyethyleneglycol Functionalized Zinc Phthalocyanine-Fullerene dyad

Utilization of polyethyleneglycol （PEG） functionalized zinc phthalocyanine, （PEG）4ZnPc in （i） singlet oxygen generation, and （ii） in building energy harvesting donor-acceptor systems using fullerene, C60 as an acceptor via the well-known metal-ligand axial coordination approach is reported. The （PEG）4ZnPe was found to be capable of producing singlet oxygen with a quantum yield, ooa of 0.77 in toluene, a value higher than that obtained for pristine （t-bu）4ZnPc （φ△=0.54） carrying no PEG groups, revealing its usefulness in photodynamic therapy applications. Spectroscopic studies revealed efficient binding of phenylimidazole functionalized fullerene, C60 Im with l ： 1 stoichiometry to （PEG）4ZnPc. Binding constant K for the formation of （PEG）4ZnPc：ImC60 dyad was found to be 6 × 103 M 1 revealing moderate stability. Geometric and electronic studies of the dyad was arrived by B3LYP/3-21G（＊） method. The HOMO level was found to be on zinc phthalocyanine entity while the LUMO level was found to be on the C60 entity suggesting formation of （PEG）4ZnPc＊＋：ImC60＊ charge separated state during the process of electron transfer reaction. Redox studies on the （PEG）4ZnPc：ImC60 dyad enabled accurate determination of the oxidation and reduction potentials of the donor-acceptor system, and to evaluate free-energy changes associated for the charge separation process. Kinetics of photoinduced charge separation and recombination in the （PEG）4ZnPc：ImC60 dyad was investigated using femtosecond transient absorption studies. Relatively long-lived charge separated states were confirmed for the dyad suggesting their potential usefulness in energy harvesting applications.
