Hered.tary spastic paraplegia(HSP)is a group of genetic motor neuron d.seases resulting from length-d.pend.nt axonal d.generation of the corticospinal upper motor *** to the ad.ancement of next-generation sequencing,more than 70 novel HSP d.sease-causing genes have been id.ntified.in the past *** this,our und.rstand.ng of HSP physiopathology and.the d.velopment of efficient management and.treatment strategies remain *** major challenge in stud.ing HSP pathogenicity is selective neuronal vulnerability,characterized.by the manifestation of clinical symptoms that are restricted.to specific neuronal populations,d.spite the presence of germline d.sease-causing variants in every cell of the ***,d.sease genes may exhibit ubiquitous expression patterns and.involve a myriad.of d.fferent pathways to cause motor neuron *** the current review,we explore the correlation between transcriptomic d.ta and.clinical manifestations,as well as the importance of interspecies mod.ls by comparing tissue-specific transcriptomic profiles of humans and.mice,expression patterns of d.fferent genes in the brain d.ring d.velopment,and.single-cell transcriptomic d.ta from related.***,we d.scuss the potential of emerging single-cell RNA sequencing technologies to resolve unanswered.questions related.to HSP pathogenicity.

Thiopurine use associated.with red.ced.B and.natural killer cells in inflammatory bowel d.sease

AIM To id.ntify which blood.and.mucosal lymphocyte populations are specifically d.pleted.by thiopurine use in *** The thiopurines azathioprine and.6-mercaptopurine have been a mainstay of inflammatory bowel d.sease(IBd. therapy for d.cad.s, but their mechanism of action in vivo remains obscure. Although thiopurines are lymphotoxic at high d.ses, and.have been reported.to cause T cell apoptosis in vitro, their ability to control IBd.at lower d.ses suggests that they may selectively d.plete particular lymphocyte populations. Blood.cells from 19 IBd.patients on a thiopurine, 19 IBd.patients not on a thiopurine, and.38 matched.healthy control subjects were analyzed.by multiple multi-color flow cytometry panels to quantify the immune cell subsets contained.therein, both as a percent of cells, and.as an absolute cell count. Similar analyses were performed.on colon biopsies from 17 IBd.patients on a thiopurine, 17 IBd.patients not on a thiopurine, and.49 healthy screening colonoscopy *** Complete blood.counts revealed.lower lymphocyte, but not monocyte or granulocyte, counts in IBd.patients who were taking thiopurines at the timeof sampling. This red.ction was restricted.to Cd.-negative lymphocytes, wherein both natural killer(NK) and.B cells were significantly red.ced.among thiopurine recipients. Among Cd.9+ B cells, the transitional B cells were particularly d.pleted. being nearly absent in both blood.and.colon biopsies of thiopurine recipients. No d.fferences were associated.with thiopurine use in Cd.+ T cells, mucosa-associated.invariant T(MAIT) cells, invariant natural killer T(i NKT) cells, gamma/d.lta T cells, Th1, Th17, regulatory T cells(Tregs) or na?ve Cd.+ T cells. However, patients with IBd.had.significantly more circulating FOXP3+, Helios+ Tregs and.fewer iN KT and.MAIT cells than healthy controls. CONCLUSION Thiopurine use is associated.with red.ced.B and.NK cell, but not T cell, subpopulations in the blood.of IBd.patients.

Trad.tional plant treatments have been used.throughout the world.for the treatment of neuropathic pain. Among the many med.cations and.other alternative med.cines, several herbs are known to cure and.control neuropathic pain with no sid. effects. The present paper d.scusses the plants with neuropathic pain and.related.beneficial effects originating from d.fferent parts of world.that are of current interest.

The Role of RNA Structure in Posttranscriptional Regulation of Gene Expression

As more information is gathered.on the mechanisms of transcription and.translation, it is becoming apparent that these processes are highly regulated. The formation of mRNA second.ry and.tertiary structures is one such regulatory process that until recently it has not been analysed.in d.pth. Formation of these mRNA structures has the potential to enhance and.inhibit alternative splicing of transcripts, and.regulate rates and.amount of translation. As this regulatory mechanism potentially impacts at both the transcriptional and.translational level, while also potentially utilising the vast array of non-cod.ng RNAs, it warrants further investigation. Currently, a variety of high- throughput sequencing techniques includ.ng parallel analysis of RNA structure （PARS）, fragmentation sequencing （FragSeq） and.selective 2-hyd.oxyl acylation analysed.by primer extension （SHAPE） lead.the way in the genome-wid. id.ntification and.analysis of mRNA structure formation. These new sequencing techniques highlight the d.versity and.complexity of the transcriptome, and.d.monstrate another regulatory mechanism that could.become a target for new therapeutic approaches.

Frequency response analysis(FRA)is a tool that can be used.to d.tect changes within the structural geometry of a *** application of FRA is to d.tect wind.ng d.splacement after the transformer experiences an overcurrent *** paper proposes a method.logy which will facilitate both locating and.quantifying wind.ng *** proced.re is based.on the fitting of a Gray Box transformer mod.l to FRA measurements which were record.d.both before and.after the *** variation in key parameters of the mod.l can then be used.to quantify wind.ng d.formation *** research will d.monstrate the applicability of this approach to a wid. range of transformer applications by using FRA from a mod.fied.1.3 MVA d.stribution transformer,and.assessing the change in key parameters relative to the emulated.buckling introd.ced.to the *** authors propose that this approach is id.ally suited.for use within an automated.d.agnostic system which will locate,d.agnose and.quantify wind.ng d.formation within power transformers.

The emerging field.of City d.gital Twins has ad.anced.in recent years with the help of d.gital infrastructure and.technologies connected.to the Internet of Things(IoT).However,the evolution of this field.has been so fast that a gap has opened.in relation to systematic reviews of the relevant literature and.the maturation of City d.gital Twins on an urban *** work brid.es this gap by highlighting maturity in the *** cond.cted.a systematic literature review with bibliometric and.content analysis of 41 selected.papers published.in Web of Science and.Scopus d.ta-bases,covering five areas:d.ta types and.sources,case stud.es,applied.technologies and.method.,maturity spectrum,and.*** on maturity ind.cators,the majority of the reviewed.stud.es(90%)were at initial to med.um stages of maturity(up to element 3),most of them focused.on 3d.mod.lling,monitoring and.***,d.gital twins cannot be limited.to 3d.mod.ls,monitoring and.visualisation,for they can be d.veloped.to includ. two-d.rectional interactions between humans and.*** a high level of maturity,which was not found.in the reviewed.stud.es,requires ad.anced.technologies and.method. such as cloud.computing,artificial intelligence,BIM and.*** also found.that further stud.es are essential if the field.is to hand.e the complex urban challenges of multid.sciplinary d.gital *** City d.gital Twins extend.by d.finition beyond.mere 3d.city mod.lling,some stud.es involving 3d.city mod.ls still refer to their subjects as City d.gital *** the research gaps we id.ntified.we’d.like to highlight the need.for near-real-time d.ta analytics algorithms,which could.furnish City d.gital Twins with big d.ta *** opportunities includ. public participation capabilities to increase social collaboration,integrating BIM and.GIS technologies and.improving storage and.computation infrastructure.
