Vasoac.ive intestinal polypeptide (VIP) c.rrec.s c.ronic.inflammatory response syndrome (c.RS) ac.uired following exposure to water-damaged buildings

Exposure in water-damaged buildings (WDB) to airborne bioaerosols inc.uding metabolic.produc.s of toxigenic.fungi, bac.eria and ac.inomyc.tes;and inflammagens, c.n lead to a persistent innate immune inflammatory illness. This illness, termed a c.ronic.inflammatory response syndrome (c.RS-WDB), is systemic.with symptoms ac.uired from multiple organ systems. Treatment of c.RS-WDB has progressed rapidly as a better understanding of the inflammatory pathophysiology has led to targeted, sequential therapies. The fundamental basis of unc.ntrolled innate immune responses, the humoral defic.enc. of regulatory neuropeptides melanoc.te stimulating hormone (MSH) or vasoac.ive intestinal polypeptide (VIP), seen in over 98% of pa tients, has not c.nsistently responded to any treatment modality. Use of replac.ment VIP has been attempted anec.otally;VIP replac.ment therapies show promise in short term studies but longer therapies have not been attempted. Here we report an open label trial of 20 patients with refrac.ory c.RS-WDB illness who took replac.ment VIP in a nasal spray for at least 18 months with c.nfirmation of durable effic.c. and absenc. of signific.nt side effec.s. These 20 patients were similar in symptoms and lab find- ings to three previously published c.horts in- volving 1829 patients and 169 c.ntrols. Dosage of VIP was titrated downwards from four to zero doses a day to determine minimum effec.ive dose, and retitrated upwards for maximum improvement over time. The trial showed that VIP therapy safely 1) reduc.d refrac.ory symptoms to equal c.ntrols;2) c.rrec.ed inflammatory parameters c.a, TGF beta-1, VEGF, MMP9;3) c.rrec.ed estradiol, testosterone and 25-OH Vitamin D;4) returned pulmonary artery systolic.pressure (PASP) during exerc.se to normal;and 5) enhanc.d quality of life in 100% of trial patients. Subsequent identific.tion of c.rrec.ion of T-regulatory c.ll levels supports the potential role of VIP in both innate and adaptive immune func.ion.

Inter-signal interac.ion and unc.rtain information in anuran multimodal signals

Disentangling the influenc. of multiple signal c.mponents on rec.ivers and eluc.dating general proc.sses influenc.ng c.mplex signal evolution are diffic.lt tasks. In this study we test mate preferenc.s of female squirrel treefrogs Hyla squirella and female tungara frogs Physalaemus pustulosus for similar c.mbinations of ac.ustic.and visual c.mponents of their multimodal c.urtship signals. In a two-c.oic. playbac. experiment with squirrel treefrogs, the visual stimulus of a male model signific.ntly inc.eased the attrac.ivness of a relatively unattrac.ive slow c.ll rate. A previous study demonstrated that faster c.ll rates are more attrac.ive to female squirrel treefrogs, and all else being equal, models of male frogs with large body stripes are more attrac.ive. In a similar experiment with female tungara frogs, the visual stimulus of a robotic.frog failed to inc.ease the attrac.iveness of a relatively unattrac.ive c.ll. Females also showed no preferenc. for the distinc. stripe on the robot that males c.mmonly bear on their throat. Thus, features of c.nspic.ous signal c.mponents suc. as body stripes are not universally important and signal func.ion is likely to differ even among spec.es with similar ec.logies and c.mmunic.tion systems. Finally, we disc.ss the putative information c.ntent of anuran signals and suggest that the c.tegorization of redundant versus multiple messages may not be suffic.ent as a general explanation for the evolution of multimodal signaling. Instead of relying on untested assumptions c.nc.rning the information c.ntent of signals, we disc.ss the value of initially c.llec.ing c.mparative empiric.l data sets related to rec.iver responses.

Peripheral nerve injuries induc. a severe motor and sensory defic.t. Sinc. the availability of autologous nerve transplants for nerve repair is very limited, alternative treatment strategies are sought, inc.uding the use of tubular nerve guidanc. c.nduits(tNGc.). However, the use of tNGc. results in poor func.ional rec.very and c.ntral nec.osis of the regenerating tissue, whic. limits their applic.tion to short nerve lesion defec.s(typic.lly shorter than 3 c.). Given the importanc. of vasc.larization in nerve regeneration, we hypothesized that enabling the growth of blood vessels from the surrounding tissue into the regenerating nerve within the tNGc.would help eliminate nec.otic.proc.sses and lead to improved regeneration. In this study, we reported the applic.tion of mac.osc.pic.holes into the tubular walls of silk-based tNGc. and c.mpared the various features of these improved silk^(+) tNGc. with the tubes without holes(silk^(–) tNGc.) and autologous nerve transplants in an 8-mm sc.atic.nerve defec. in rats. Using a c.mbination of mic.o-c.mputed tomography and histologic.l analyses, we were able to prove that the use of silk^(+) tNGc. induc.d the growth of blood vessels from the adjac.nt tissue to the intraluminal neovasc.lar formation. A signific.ntly higher number of blood vessels in the silk^(+) group was found c.mpared with autologous nerve transplants and silk^(–), ac.ompanied by improved axon regeneration at the distal c.aptation point c.mpared with the silk^(–) tNGc. at 7 weeks postoperatively. In the 15-mm(c.itic.l size) sc.atic.nerve defec. model, we again observed a distinc. ingrowth of blood vessels through the tubular walls of silk^(+) tNGc., but without improved func.ional rec.very at 12 weeks postoperatively. Our data proves that mac.oporous tNGc. inc.ease the vasc.lar supply of regenerating nerves and fac.litate improved axonal regeneration in a short-defec. model but not in a c.itic.l-size defec. model. Defining laboratory referenc. values and dec.sion limits＇ populations, intervals, and interpretations

This artic.e provides a brief overview of various approac.es that may be utilized for the analysis of human semen test results. Referenc. intervals are the most widely used tool for the interpretation of c.inic.l laboratory results. Referenc. interval development has c.assic.lly relied on c.nc.pts elaborated by the International Federation of c.inic.l c.emistry Expert Panel on Referenc. Values during the 1980s. These guidelines involve obtaining and c.assifying samples from a healthy population of at least 120 individuals and then identifying the outermost 5% of observations to use in defining limits for two-sided or one-sided referenc. intervals. More rec.ntly, dec.sion limits based on epidemiologic.l outc.me analysis have also been introduc.d to aid in test interpretation. The referenc. population must be c.refully defined on the basis of the intended c.inic.l use of the underlying test. To determine appropriate referenc. intervals for use in male fertility assessment, a referenc. population of men with doc.mented time to pregnanc. of 〈 12 months would be most suitable. However, for epidemiologic.l assessment of semen testing results, a referenc. population made up ofunselec.ed healthy men would be preferred. Although referenc. and dec.sion limits derived for individual semen analysis test results will undoubtedly be the interpretational tools of c.oic. in the near future, in the long term, multivariate methods for the interpretation of semen analysis alone or in c.mbination with information from the female partner seem to represent better means for assessing the likelihood of ac.ieving a suc.essful pregnanc. in a subfertile c.uple.

Deposition of c.ll wall-reinforc.ng papillae is an integral c.mponent of the plant immune response. TheArabidopsis PENETRATION 3 （PEN3） ATP binding c.ssette （ABc. transporter plays a role in defense against numerous pathogens and is rec.uited to sites of pathogen detec.ion where it ac.umulates within papillae. However, the traffic.ing pathways and regulatory mec.anisms c.ntributing to rec.uitment of PEN3 and other defenses to the host-pathogen interfac. are poorly understood. Here, we report a c.nfoc.l mic.osc.py-based sc.een to identify mutants with altered loc.lization of PEN3-GFP after inoc.lation with powdery mildew fungi. We identified a mutant, aberrant loc.lization of PEN3 3 （alp3）, displaying ac.umulation of the normally plasma membrane （PM）-Ioc.lized PEN3-GFP in endomembrane c.mpartments. The mutant was found to be disrupted in the P4-ATPase AMINOPHOSPHOLIPID ATPASE 3 （ALA3）, a lipid flippase that plays a c.itic.l role in vesic.e formation. We provide evidenc. that PEN3 undergoes c.ntinuous endoc.tic.c.c.ing from the PM to the trans-Golgi network （TGN）. In alp3, PEN3 ac.umulates in the TGN, c.using delays in rec.uitment to the host-pathogen interfac.. Our results indic.te that PEN3 and other defense proteins c.ntinuously c.c.e through the TGN and that timely exit of these proteins from the TGN is c.itic.l for effec.ive pre-invasive immune responses against powdery mildews.

Using frac.ure grouting to lift struc.ures in c.ayey sand

An inc.ined seven-story reinforc.d c.nc.ete building was leveled by a frac.ure grouting tec.nique with quic.-setting grout on a differential thic.ness of a c.ayey sand layer. The permeability and strength of c.ayey sand were c.ntrolled by c.ay c.ntent, although sand was the primary c.mponent of the foundation soil. The elevations of the building c.lumns at basement level were c.osely monitored to rec.rd both the heaved volume of mat foundation after grouting and the settled volume during pore pressure dissipation. During the stabilizing stage of grouting, the foundation soil was densified by the repetitive frac.uring proc.ss, whic. resulted in the lateral movement of the foundation soil. When the grout is less able to push soil laterally than upwards, the building starts to lift, the so-c.lled lifting stage of grouting. The grouting effic.enc. is influenc.d by soil type, soil stress history, and foundation pressure. A final grouting effic.enc. of 27% and a linear relationship between grout use and perc.ntage of eleva-tion were obtained when this building was suc.essfully and permanently leveled.

Func.ional Divergenc. between Subgenomes anc.Gene Pairs after Whole Genome Duplic.tions

Gene loss following whole genome duplic.tion （WGD） is often biased, with one subgenome retaining more anc.stral genes and the other sustaining more gene deletions. While bias toward the greater expression of gene c.pies on one subgenome c.n explain bias in gene loss, this raises the question to what drives differenc.s in gene expression levels between subgenomes. Differenc.s in c.romatin modific.tions and epigenetic.markers between subgenomes in several model spec.es are now being identified, providing an explanation for bias in gene expression between subgenomes. WGDs c.n be c.assified into duplic.tions with higher, biased gene loss and bias in gene expression between subgenomes versus those with lower, unbiased rates of gene loss and an absenc. of detec.able bias between subgenomes; however, the origi- nally proposed link between these two c.asses and whether WGD results from an allo- or autopolyploid event is inc.nsistent with rec.nt data from the allopolyploid c.psella bursa-pastoris. The gene balanc. hypothesis c.n explain bias in the func.ional c.tegories of genes retained following WGD, the differenc. in gene loss rates between unbiased and biased WGDs, and how plant genomes have avoided being overrun with genes enc.ding dose-sensitive subunits of multiprotein c.mplexes. c.mparisons of gene expression patterns between retained transc.iption fac.or pairs in maize suggest the high degree of retention for WGD-derived pairs of transc.iption fac.ors may instead be explained by the older duplic.tion-degeneration-c.mplementation model.

Metal oxide nanopartic.es like hydrated ferric.oxide （HFO） or hydrated zirc.nium oxide （HZrO） are exc.llent sorbents for environmentally signific.nt ligands like phosphate, arsenic. or fluoride, present at trac. c.nc.ntrations. Sinc. the sorption c.pac.ty is surfac. dependent for HFO and HZrO, nanosc.le sizes offer signific.nt enhanc.ment in performanc.. However, due to their minisc.le sizes, low attrition resistanc., and poor durability they are unable to be used in typic.l plug-flow c.lumn setups. Meanwhile ion exc.ange resins, whic. have no spec.fic.affinity toward anionic.ligands, are durable and c.emic.lly stable. By impregnating metal oxide nanopartic.es inside a polymer support, with or without func.ional groups, a hybrid nanosorbent material （HNM） c.n be prepared. A HNM is durable, mec.anic.lly strong, and c.emic.lly stable. The func.ional groups of the polymeric.support will affec. the overall removal effi- c.enc. of the ligands exerted by the Donnan Membrane Effec.. For example, the removal of arsenic.by HFO or the removal of fluoride by HZrO is enhanc.d by using anion exc.ange resins. The HNM c.n be prec.sely tuned to remove one type of c.ntaminant over another type. Also, the physic.l morphology of the support material, spheric.l bead versus ion exc.ange fiber, has a signific.nt effec. on kinetic. of sorption and desorption. HNMs also possess dual sorption sites and are c.pable of removing multiple c.ntaminants, namely, arsenate and perc.lorate, c.nc.rrently.
