Functional specialization of Nicotiana attenuata phytochromes in leaf development and flowering time

Phytochromes mainly function in photoau- totrophic organisms to adjust growth in response to fluctuating light signals. The different isoforms of plant phytochromes often display both conserved and divergent roles, presumably to fine-tune plant responses to environmental signals and optimize fitness. Here we describe the distinct, yet partially redundant, roles of phytochromes NaPHYA, NaPHYB1 and NaPHYB2 in a wild tobacco species, Nicotiana attenuata using RNAi-silenced phytochrome lines. Consistent with results reported from other species, silencing the expression of NaPHYA or NaPHYB2 in N. attenuata had mild or no influence on plant develop- ment as long as NaPHYBI was functional; whereas silencing the expression of NaPHYB1 alone strongly altered flowering time and leaf morphology. Thecontribution of NaPHYB2 became significant only in the absence of NaPHYB1; plants silenced for both NaPHYB1 and NaPHYB2 largely skipped the rosette- stage of growth to rapidly produce long, slender stalks that bore flowers early： hallmarks of the shade- avoidance responses. The phenotyping of phyto- chrome-silenced lines, combined with sequence and transcript accumulation independent functional analysis, suggest the diversification of the phytochromes, and a dominant role of NaPHYB1 and NaPHYB2 in N. attenuata＇s vegetative and reproductive development.

The Optical and Two-Photon Properties of Amylose-Encapsulated Chromophore

Encapsulation of DASP (4-[4-(dimethylamino)styryl]-1-octylpyridinium bromide) with amylose, and conformational transitions of amylose upon complexation with the dyes were investigated using UV-vis, fluorescence and two-photon absorption properties in various Dimethyl Sulfoxide ( DMSO)/H2O mixtures. Encapsulation of the dye was achieved by dilution of DMSO stock solutions of the chromophores (1 × 10-3 M) and amylose (1 × 10-1 M) with DMSO, followed by a dropwise addition of H2O with continuous stirring at room temperature. At less than 40% aqueous DMSO solutions, the chromophores had a stable encapsulation with amylose and showed over 100 times the fluorescence intensity of non-encapsulated chromophores. Both of the dye solution with and without amylose also exhibit 2PA (two-photon absorption), as determined by nonlinear absorption measurements using 10 ns laser pulses at 800 nm. The respective values of two-photon cross-sections are 13.3 × 10-20 for encapsulated dye solution and 2.53 × 10-20 cm4/GW for non encapsulated dye solution. The data clearly indicate that the contribution of the encapsulated dye dominates the two-photon absorption cross-section of the non encapsulated dye at 800 nm.

Validation of Treatment Planning Dose Calculations: Experience Working with Medical Physics Practice Guideline 5.a.

Recently published Medical Physics Practice Guideline 5.a. (MPPG 5.a.) by American Association of Physicists in Medicine (AAPM) sets the minimum requirements for treatment planning system (TPS) dose algorithm commissioning and quality assurance (QA). The guideline recommends some validation tests and tolerances based primarily on published AAPM task group reports and the criteria used by IROC Houston. We performed the commissioning and validation of the dose algorithms for both megavoltage photon and electron beams on three linacs following MPPG 5.a. We designed the validation experiments in an attempt to highlight the evaluation method and tolerance criteria recommended by the guideline. It seems that comparison of dose profiles using in-water scan is an effective technique for basic photon and electron validation. IMRT/VMAT dose calculation is recommended to be tested with some TG-119 and clinical cases, but no consensus of the tolerance exists. Extensive validation tests have provided the better understanding of the accuracy and limitation of a specific dose calculation algorithm. We believe that some tests and evaluation criteria given in the guideline can be further refined.

＜正＞Background/Aims:Nitric Oxide （NO） and its interrelated redox species （collectively denoted as NOx） play a multi - faceted role in cancer progression. NOx production induces mutational events,and it has also been found to simultaneously inhibit apoptosis. In an effort to define the underlying mechanisms of apoptosis inhibition arising from nitric oxide exposure, we here develop a novel mouse model of lung adenocarcinoma. Methods: Four NOx donor compounds （DETA - NONOate, Spermine - NONOate, SNAP, and glyco - SNAP） were exposed to a novel murine lung adenocarcinoma cell line at increasing *** extent of NOx production was measured by the Griess reaction,spectrophotometrically,and via nitrotyrosine *** alkaline single- cell gel electrophoresis assay （the COMET assay） was used to measure DNA strand breaks （DSB’ s） and was correlated with cell viability. A fluorescence caspase activity assay was used to determine apoptosis and was correlated with apoptosis - related gene products using cDNA microarrays. Results:Kinetics,concentration,and mode of delivery all had an effect on NOx mediated DSB’s. At the highest concentration of NONOate donors, the DSB’ s correlated with cell death in a caspase - 9 dependent fashion. Caspase -9 activity appeared to be inhibited by thiol - based NOx donors, and since DTT reversed NO’s effect of apoptosis, inhibition is believed to have occurred via a S - nitrosylation pathway. This change in caspase activity was found to correlate with a number of transcriptional changes, suggesting that several key apoptosis steps are likely under transcriptional control via S - nitrosylation. Conclusions: The results of our murine model of lung cancer progression were found to parallel those previously reported in a human lung adenocarcinoma cell line. The model suggests that various apoptosis related gene products are likely regulated through S - nitrosylation in a caspase - 9 dependent manner. With this model in hand, we c
