Synthetic lethality is a novel model for cancer *** understand the function and mechanism of BEN domain-containing protein 4(BEND4)in pancreatic cancer,eight cell lines and a total of 492 cases of pancreatic neoplasia samples were included in this ***-specific polymerase chain reaction,CRISPR/Cas9,immunoprecipitation assay,comet assay,and xenograft mouse model were ***4 is a new member of the BEN domain *** expression of BEND4 is regulated by promoter region *** is methylated in 58.1%(176/303)of pancreatic ductal adenocarcinoma(PDAC),33.3%(14/42)of intraductal papillary mucinous neoplasm,31.0%(13/42)of pancreatic neuroendocrine tumor,14.3%(3/21)of mucinous cystic neoplasm,4.3%(2/47)of solid pseudopapillary neoplasm,and 2.7%(1/37)of serous cystic ***4 methylation is significantly associated with late-onset PDAC(>50 years,Pgnostic marker(Pgous end joining signaling by interacting with Ku80 and promotes DNA damage *** of BEND4 increased the sensitivity of PDAC cells to ATM ***,the present study revealed an uncharacterized tumor suppressor BEND4 and indicated that methylation of BEND4 may serve as a potential synthetic lethal marker for ATM inhibitor in PDAC treatment.

核磁共振(MR)热成像引导激光间隙热疗或立体定向激光消融术(Stereotactic laser ablation,SLA是开颅手术治疗脑海绵状血管瘤(Cerebral cavernous malformations,CCMs)所致局灶性癫痫的微创替代术。此研究检测了SLA治疗致痫性CCMs的安全性和有效性,回顾性分析连续19例伴有CCM的局部发作的患者。每例患者均接受CCMs和邻近皮质的SLA,继而进行标准的临床和影像学随访。除1例患者外,所有患者均患有慢性难治性癫痫(中位病程8年,年龄范围0.5~52岁)。病变位于颞叶(13例)、额叶(5例)和顶叶(1例)。CCMs在测温过程中会诱发磁化率伪影,但病灶周围皮层易于观察。在接受12个月以上随访的17例患者中,有14例(82%)达到了Engel I级,其中10例(59%)为Engel IA级。2例患者仅接受SLA后未达到无发作,在进一步行颅内电极引导下开放性切除术后达到了无发作。延迟的术后影像学检查证实了CCMs缩小(中位数减少83%)和周围皮质的消融。开放手术后对一个先前消融的CCM组织病理学检查证实闭塞。SLA未引起可检测的出血。两种症状性神经功能缺损(视觉和运动障碍)是可预见的,且均非永久残疾。在连续的回顾性研究中,MR热成像指导的SLA是致痫性CCMs开放手术治疗的有效替代方法。该方法无出血并发症,且临床上明显的神经功能缺损是可预见的。若有需要,SLA对后续的开放手术无任何障碍。

Response of saproxylic insect communities to logging history, tree species, stage of decay, and wood posture in the central Nearctic boreal forest

Saproxylic insect assemblages are essential functional components of forest ecosystems that can be affected by forest *** used a split-plot ANOVA design to analyze differences in selected saproxylic insects（all arthropod orders and dipteran and parasitic hymenopteran families） emerging from dead wood of sites with different logging histories（horse-logged,mechanically-logged and unlogged）,tree species（Populus and Picea）,stage of decay（early-and late-decay stages） and posture（standing and downed logs） in the boreal forest of central *** clear effects of logging history were seen for the studied taxa; however,interaction between logging history and other dead wood features was *** consistently emerged more from Populus than from Picea dead *** of the studied saproxylic families were more abundant in late-decay than in early-decay *** of the Cecidomyiidae,Ceratopogonidae,Empididae,Mycetophilidae and Sciaridae families,and hymenopterans of the Diapriidae and Ichneumonidae families were significantly more abundant in downed than in standing dead *** contrast,Mymaridae was most abundant in standing dead *** study provides evidence that some insects at high taxonomic levels respond differently to dead wood quality,and this could inform future management strategies in the boreal forest for the conservation of saproxylic fauna and their ecological functions.

Utilizing <i>Spirogyra grevilleana</i>as a Phytoremediatory Agent for Reduction of Limnetic Nutrients and <i>Escherichia coli</i>Concentrations

The freshwater alga Spirogyra grevilleana was used in an experimental biofiltration system to reduce levels of Escherichia coli, nitrates, and phosphates. Water collected from a 2.32 ha lake in Atlanta, georgia, USA was pumped at a constant rate (6.17 × 10-1 m3·hr-1) through the algal filtration devices with low and high concentrations of S. grevilleana. Effluent water was tested over time for E. coli, nitrate, phosphate, dissolved oxygen, and pH levels. Both concentrations of S. grevilleana reduced E. coli by 100% and significantly reduced nitrate concentrations (30% ± 13%) and phosphate concentrations (23% ± 5%) while maintaining dissolved oxygen and pH at normal levels. Utilizing S. grevilleana in an algal filtration device could potentially provide a sustainable, flexible, and low-cost method of E. coli reduction in freshwater lakes worldwide. Initial results indicate that the use of S. grevilleana in conjunction with an algal filtration device is potentially capable of creating potable water.

Modelling and Analysis of TCP Performance in Wireless Multihop Networks

Researchers have used extensive simulation and experimental studies to understand TCP performance in wireless multihop networks. In contrast, the objective of this paper is to theoretically analyze TCP performance in this environment. By examining the case of running one TCP session over a string topology, a system model for analyzing TCP performance in multihop wireless networks is proposed, which considers packet buffering, contention of nodes for access to the wireless channel, and spatial reuse of the wireless channel. Markov chain modelling is applied to analyze this system model. Analytical results show that when the number of hops that the TCP session crosses is ?xed, the TCP throughput is independent of the TCP congestion window size. When the number of hops increases from one, the TCP throughput decreases ?rst, and then stabilizes when the number of hops becomes large. The analysis is validated by comparing the numerical and simulation results.

Advances in plant phenotyping technologies are dramatically reducing the marginal costs of collecting multiple phenotypic measurements across several time ***,most current approaches and best statistical practices implemented to link genetic and phenotypic variation in plants have been developed in an era of single-time-point ***,we used time-series phenotypic data collected with an unmanned aircraft system for a large panel of soybean(glycine max(L.)Merr.)varieties to identify previously uncharacterized ***,we focused on the dissection of canopy coverage(CC)variation from this rich data *** also inferred the speed of canopy closure,an additional dimension of CC,from the time-series data,as it may represent an important trait for weed ***-wide association studies(gWASs)identified 35 loci exhibiting dynamic associations with CC across developmental *** time-series data enabled the identification of 10 known flowering time and plant height quantitative trait loci(QTLs)detected in previous studies of adult plants and the identification of novel QTLs influencing *** novel QTLs were disproportionately likely to act earlier in development,which may explain why they were missed in previous single-time-point ***,this time-series data set contributed to the high accuracy of the gWASs,which we evaluated by permutation tests,as evidenced by the repeated identification of loci across multiple time *** novel loci showed evidence of adaptive selection during domestication,with different genotypes/haplotypes favored in different geographic *** summary,the time-series data,with soybean CC as an example,improved the accuracy and statistical power to dissect the genetic basis of traits and offered a promising opportunity for crop breeding with quantitative growth curves.

1 IntroductionCulture systems capable of expanding and/or maintaining hematopoietic stem cells will not only facilitate our understanding of stem cell biology, but also broaden clinical applications. Among various in vitro hematopoietic culture systems, co-cultures of marrow or CD34+ cells with an adherent stromal layer that can produce cytokines and extracellular matrix components most effectively supports long-term hematopoiesis ( LTC ), mimicking the bone marrow micro-environment.

Optical spectroscopy can be used to quickly characterise the structural properties of individual ***,it cannot be applied to biological assemblies because light is generally blind to the spatial distribution of the component *** insensitivity arises from the mismatch in length scales between the assemblies(a few tens of nm)and the wavelength of light required to excite chromophores(≥150 nm).Consequently,with conventional spectroscopy,ordered assemblies,such as the icosahedral capsids of viruses,appear to be indistinguishable isotropic spherical *** limits potential routes to rapid high-throughput portable detection appropriate for point-of-care ***,we demonstrate that chiral electromagnetic(EM)near fields,which have both enhanced chiral asymmetry(referred to as superchirality)and subwavelength spatial localisation(∼10 nm),can detect the icosahedral structure of virus ***,they can detect both the presence and relative orientation of a bound virus *** illustrate the potential uses of the exquisite structural sensitivity of subwavelength superchiral fields,we have used them to successfully detect virus particles in the complex milieu of blood serum.
